Claims (1)
- 【特許請求の範囲】 1.(i)それぞれ、配列番号1、配列番号2、配列番号3及び配列番号7に 示されるアミノ酸配列から選択されたアミノ酸配列を有し;又は(ii)1又は複 数の前記アミノ酸配列と少なくとも80%の相同性を示し;そして/又は1つの前 記アミノ酸配列を有するα−アミラーゼに対して生じさせた抗体との免疫学的交 差反応性を示し;そして/又は1つの前記アミノ酸配列を有するα−アミラーゼ をコードするDNA 配列と同じプローブとハイブリダイズするDNA 配列によりコー ドされる親α−アミラーゼの変異体であって;前記変異体においては、 (a)前記親α−アミラーゼの少なくとも1つのアミノ酸残基が欠失されてお り;そして/又は (b)前記親α−アミラーゼの少なくとも1つのアミノ酸残基が異なったアミ ノ酸残基により置換されており;そして/又は (c)少なくとも1つのアミノ酸残基が前記親α−アミラーゼに対して挿入さ れており; 前記変異体がα−アミラーゼ活性を有し、そして前記親α−アミラーゼに対し ての次の性質の少なくとも1つを示し:高められた熱安定性;酸化に対しての高 められた安定性;及び低められたCa2+依存性;但し、前記変異体のアミノ酸配列 は、それぞれ配列番号1、配列番号2、配列番号3及び配列番号7に示されるア ミノ酸配列のいづれとも同一でないことを特徴とする変異体。 2.前記親α−アミラーゼの少なくとも1つの酸化可能アミノ酸残基が欠失さ れており、又は前記酸化可能アミノ酸残基よりも酸化に対して低い感受性である 異なったアミノ酸残基により置換されている請求の範囲第1項記載の変異体。 3.前記酸化可能アミノ酸残基が、メチオニン、トリプトファン、システイン 及びチロシンから成る群から選択される請求の範囲第2項記載の変異体。 4.前記酸化可能アミノ酸残基が、配列番号1に示されるアミノ酸残基のM9, M10,M105,M202,M208,M261,M309,M382,M430 又はM440であり、又はそれら に対して同等のメチオニンである請求の範囲第2又は3項記載の変異体。 5.配列番号1に示されるアミノ酸配列における次の置換:M9L; M10L ; M105 L ; M202L,T,F,I,V;M208L ; M261L ; M309L ; M382L ; M430L ; M440L の1つであるか、又はそれらに対して同等のメチオニン置換を含んで成る請求 の範囲第4項記載の変異体。 6.前記メチオニン残基がトレオニンにより置換されている請求の範囲第3〜 5のいづれか1項記載の変異体。 7.配列番号1に示されるアミノ酸配列のF180,R181,G182,T183,G184又は K185であるか、又はそれらに対して同等の少なくとも1つのアミノ酸が欠失され ている請求の範囲第1〜6のいづれか1項記載の変異体。 8.前記欠失されたアミノ酸が、前記アミノ酸残基のいづれか2種の残基又は それらの2種の残基に対して同等である請求の範囲第7項記載の変異体。 9.前記欠失が、R181*+G182*;又はT183*+G184*であるか、又はそれに対し て同等である請求の範囲第8項記載の変異体。 10.配列番号1に示されるアミノ酸配列における次の置換:K269R ; P260E ; R124P ; M105F,I,L,V;M208F,W,Y;L217I ;V206I,L,Fの1つであ るか、又はそれに対して同等のアミノ酸置換を含んで成る請求の範囲第1〜9の いづれか1項記載の変異体。 11.配列番号1に示されるアミノ酸配列における次の置換:Y243F ; K108R ; K179R ; K239R ; K242R ; K269R ; D163N ; D188N ; D192N ; D199N ; D205N ; D207N ; D209N ; E190Q ; E194Q ; N106D の1つであるか、又はそれに対して同 等のアミノ酸置換を含んで成る請求の範囲第1〜10のいづれか1項記載の変異体 。 12.請求の範囲第1〜11のいづれか1項記載のα−アミラーゼをコードするDN A配列を含んで成るDNA構造体。 13.請求の範囲第12項記載のDNA構造体を担持する組換え発現ベクター。 14.請求の範囲第12項記載のDNA 構造体又は請求の範囲第13項記載のベクター により形質転換される細胞。 15.微生物である請求の範囲第14項記載の細胞。 16.細菌又は菌類である請求の範囲第15項記載の細胞。 17.グラム陽性細菌、たとえばバシラス・サブチリス、バシラス・リケニホル ミス、バシラス・レンタス、バシラス・ブレビス、バシラス・ステアロサーモフ ィラス、バシラス・アルカロフィラス、バシラス・アミロリクエファシエンス、 バシラス・コアグランス、バシラス・サーキュランス、バシラス・ラウタス、バ シラス・ツリンギエンシス、又はストレプトマイセス・リビダンスもしくはスト レプトマイセス・ムリナス、又はグラム陰性細菌、たとえばE.コリである請求 の範囲第16項記載の細胞。 18.請求の範囲第1〜11のいづれか1項記載のα−アミラーゼ変異体を生成す るための方法であって、請求の範囲第14〜17のいづれが1項記載の細胞が、前記 α−アミラーゼ変異体の生成の助けとなる条件下で培養され、そして前記α−ア ミラーゼ変異体が続いて、培養物から回収されることを含んで成る方法。 19.洗浄及び/又は皿洗いへの請求の範囲第1〜11のいづれか1 項記載のα−アミラーゼ変異体の使用。 20.請求の範囲第1〜11のいづれか1項記載のα−アミラーゼ変異体を、場合 によっては、非ダスチング粒質物、安定化された液体又は保護された酵素の形で 含んで成る洗剤添加剤。 21.前記添加剤1g当たり酵素タンパク質0.02〜200mg を含んで成る請求の範 囲第20項記載の洗剤添加剤。 22.さらに、他の酵素、たとえばプロテアーゼ、リパーゼ、ペルオキシダーゼ 、もう1つの澱粉分解性酵素、及び/又はセルラーゼを含んで成る請求の範囲第 20又は21項記載の洗剤添加剤。 23.請求の範囲第1〜11のいづれか1項記載のα−アミラーゼ変異体、及び界 面活性剤を含んで成る洗剤組成物。 24.さらに、他の酵素、たとえばプロテアーゼ、リパーゼ、ペルオキシダーゼ 、もう1つの澱粉分解性酵素及び/又はセルラーゼを含んで成る請求の範囲第23 項記載の洗剤組成物。 25.請求の範囲第1〜11のいづれか1項記載のα−アミラーゼ変異体、及び界 面活性剤を含んで成る手動又は自動皿洗い用洗剤組成物。 26.さらに、他の酵素、たとえばプロテアーゼ、リパーゼ、ペルオキシダーゼ 、もう1つの澱粉分解性酵素及び/又はセルラーゼを含んで成る請求の範囲第25 項記載の皿洗い用洗剤組成物。 27.請求の範囲第1〜11のいづれか1項記載のα−アミラーゼ変異体、及び界 面活性剤を含んで成る手動又は自動洗濯用組成物。 28.さらに他の酵素、たとえばプロテアーゼ、リパーゼ、ペルオキシダーゼ、 もう1つの澱粉分解性酵素及び/又はセルラーゼを含んで成る請求の範囲第27項 記載の洗濯用組成物。 29.織物糊抜きへの請求の範囲第1〜11のいづれか1項記載のα−アミラーゼ 変異体の使用。
