Claims (9)
- 장내세균과(Enterobacteriaceae)에 속하고 L-아미노산 생산능을 갖는 세균을 배지에서 배양하여, 배양물 중에 L-아미노산을 생산 축적시켜, 당해 배양물로부터 L-아미노산을 채취함을 포함하는 L-아미노산의 제조 방법으로서, 상기 세균은 본래 피롤로퀴놀린 퀴논을 조효소로서 사용하는 글루코스 탈수소효소(GCD)의 활성을 갖지만, 당해 GCD의 활성이 저하되도록 개변된 세균으로, 상기 GCD의 활성의 저하가
(1) gcd 유전자의 일부 또는 전부의 결손,
(2) gcd 유전자에 다른 서열의 삽입,
(3) gcd 유전자에 변이의 도입, 및
(4) gcd 유전자의 프로모터 서열의 개변으로 이루어진 그룹으로부터 선택되는 하나 이상의 방법에 의해 달성되고,
상기 세균이, 판토에아(Pantoea)속, 엔테로박터(Enterobacter)속, 에르위니아(Erwinia)속, 클렙시엘라(Klebsiella)속, 프로비덴시아(Providencia)속, 살모넬라(Salmonella)속, 세라티아(Serratia)속, 모르가넬라(Morganella)속, 및 예르시니아(Yersinia)속으로부터 선택되는 속에 속하는 세균인, 방법. - 삭제
- 제1항에 있어서, 상기 gcd 유전자가, 서열번호 2의 아미노산 서열을 암호화하는 DNA 또는 이의 변이체인, 방법.
- 제1항에 있어서, 상기 L-아미노산이 L-글루탐산, L-리신, L-트레오닌, L-아르기닌, L-히스티딘, L-이소류신, L-발린, L-류신, L-페닐알라닌, L-티로신, L-트립토판, 및 L-시스테인으로 이루어지는 그룹으로부터 선택되는 1종 또는 2종 이상의 L-아미노산인, 방법.
- 제4항에 있어서, 상기 L-아미노산이 L-글루탐산 또는 L-시스테인인, 방법.
- 제5항에 있어서, 상기 L-아미노산이 L-글루탐산이며, 상기 세균에서 시트르산 신타제, 메틸 시트르산 신타제, 포스포에놀피루브산 카복실라제, 및 글루타메이트 데하이드로게나제로 이루어지는 그룹으로부터 선택되는 1종 또는 2종 이상의 효소의 활성이 증강되어 있는, 방법.
- 제5항에 있어서, 상기 L-아미노산이 L-시스테인이며, 상기 세균에서 3-포스포글리세레이트 데하이드로게나제, 세린 아세틸트랜스퍼라제, 및 황산염/티오황산염 수송계로 이루어진 그룹으로부터 선택되는 1종 또는 2종 이상의 활성이 증강되거나, 또는 yeaS 유전자의 발현이 증강되거나, 또는 상기 활성 및 발현이 둘 다 증강되는, 방법.
- 삭제
- 제1항에 있어서, 상기 세균이 판토에아 아나나티스(Pantoea ananatis)인, 방법.
