상기 감소 또는 불활성화는 상기 GDH를 코딩하는 유전자 내로 하나 이상의 염기쌍의 삽입에 의한 삽입 돌연변이, 상기 유전자 내의 하나 이상의 염기쌍의 결실을 갖는 결실 돌연변이, 및 상기 유전자 내의 넌센스 코돈 또는 상이 코돈을 도입시키는 염기쌍의 전이(transition) 또는 전환(transversion) 돌연변이로 이루어지는 군으로부터 선택되는 하나 이상의 돌연변이 방법에 의하여 유발되는 것인 L-오르니틴 생산능이 향상된 코리네박테리움 글루타미쿰.
- 제1항에 있어서,
상기 감소 또는 불활성화는 상기 GDH를 코딩하는 유전자의 일부분과 항생제 마커를 포함하는 재조합 벡터에 의한 형질전환에 의해 유발되는 것인 L-오르니틴 생산능이 향상된 코리네박테리움 글루타미쿰.
- 제3항에 있어서,
상기 항생제 마커는 카나마이신 내성부여 유전자인 것인 L-오르니틴 생산능이 향상된 코리네박테리움 글루타미쿰.
- 제3항에 있어서,
상기 재조합 벡터는 pK18mobsacB 벡터로부터 유래된 것인 L-오르니틴 생산능이 향상된 코리네박테리움 글루타미쿰.
- 제3항에 있어서,
상기 재조합 벡터는 추가로 sacB 유전자를 포함하는 것인 L-오르니틴 생산능이 향상된 코리네박테리움 글루타미쿰.
- 제6항에 있어서,
상기 재조합 벡터는 도 2a에 도시된 개열지도를 갖는 pSJC3513, 도 2b에 도시된 개열지도를 갖는 pSJC3515 또는 도 2c에 도시된 개열지도를 갖는 pSJC3517인 것인 L-오르니틴 생산능이 향상된 코리네박테리움 글루타미쿰.
- 제1항에 있어서,
상기 GDH를 코딩하는 유전자는 서열번호 1의 염기서열로 구성되는 NCgl0281, 서열번호 2의 염기서열로 구성되는 NCgl2582 또는 서열번호 3의 염기서열로 구성되는 NCgl2053인 것인 L-오르니틴 생산능이 향상된 코리네박테리움 글루타미쿰.
- 제1항에 있어서,
상기 코리네박테리움 글루타미쿰은 코리네박테리움 글루타미쿰 SJC8250(KACC9 1631P), 코리네박테리움 글루타미쿰 SJC8251(KACC9 1632P) 또는 코리네박테리움 글루타미쿰 SJC8252(KACC9 1633P)인 것인 L-오르니틴 생산능이 향상된 코리네박테리움 글루타미쿰.
- (ⅰ) 글루코스 디하이드로게나제(GDH)를 코딩하는 유전자의 일부 서열이 변이된 폴리뉴클레오티드 단편을 수득하는 단계;
(ⅱ) 상기 수득한 폴리뉴클레오티드 단편을, L-오르니틴을 생산할 수 있는 숙주세포에 도입될 수 있고 염색체상에서 상동재조합을 수행할 수 있는 벡터에 도입하여 재조합 벡터를 수득하는 단계;
(ⅲ) 상기 수득한 재조합 벡터를 숙주세포에 도입하여 상동재조합체를 수득하는 단계; 및,
(ⅳ) 상기 상동재조합체 중에서 GDH의 활성이 감소 또는 불활성화된 균주를 선발하는 단계를 포함하는, 제1항 내지 제9항 중 어느 한 항에 기재된 L-오르니틴 생산능이 향상된 코리네박테리움 글루타미쿰의 제조방법.
- (ⅰ) 탄소원의 일부 또는 전부로서 글루코스가 포함된 배양 배지에 제1항 내지 제9항 중 어느 한 항의 L-오르니틴 생산능이 향상된 코리네박테리움 글루타미쿰을 접종하고 배양하여 배양물을 수득하는 단계; 및,
(ⅱ) 상기 배양물로부터 L-오르니틴을 회수하는 단계를 포함하는 L-오르니틴을 생산하는 방법.
