Abstract
Description
도 2. 키메라 Mov19와 huMov19 중쇄와 경쇄 가변 도메인 및 muFR1-21과 huFR1-21 중쇄와 경쇄 가변 도메인의 배열. 쥐 대응물과 함께 Mov19와 Fr1-21 가변 영역에 대한 재표면처리(resurfacing)된 서열의 배열. A)와 C) 경쇄 가변 도메인; B)와 D) 중쇄 가변 도메인. 대시 "-"는 쥐 서열과 동일함을 나타낸다. 이들 CDR(Kabat 정의)은 밑줄로 표시된다.

(A)는 인간 엽산염 수용체 1 에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편이고;
(L)은 링커이고; 그리고
(C)는 세포독성제이고,
여기서 상기 링커 (L)은 (A)를 (C)에 연결하고;
상기 항체 또는 이의 항원 결합 단편이 하기를 포함하는 면역접합체:
(a) SSFGMH(SEQ ID NO:72) 의 HC CDR1 아미노산 서열; YISSGSSTIS(SEQ ID NO:73) 의 HC CDR2 아미노산 서열; 및 EAYGSSMEY(SEQ ID NO:74) 의 HC CDR3 아미노산 서열을 포함하는 VH; 및
(b) RASQNINNNLH(SEQ ID NO:69) 의 LC CDR1 아미노산 서열; YVSQSVS(SEQ ID NO:70) 의 LC CDR2 아미노산 서열; 및 QQSNSWPHYT(SEQ ID NO:71) 의 LC CDR3 아미노산 서열을 포함하는 VL. - 제 12 항에 있어서, 상기 항체 또는 이의 항원 결합 단편이 YISSGSSTISYADSVKG (SEQ ID NO:85) 의 HC CDR2 아미노산 서열을 포함하는 면역접합체.
- 제 13 항에 있어서, 상기 항체 또는 이의 항원 결합 단편이 SEQ ID NO:101 의 VH 아미노산 서열 및 SEQ ID NO:100 의 VL 아미노산 서열을 포함하는 면역접합체.
- 제 14 항에 있어서, 상기 항체가 SEQ ID NO:117 의 HC 아미노산 서열 및 SEQ ID NO:116 의 LC 아미노산 서열을 포함하는 면역접합체.
- 제 12 항에 있어서, 상기 링커가 친수성 링커 또는 디카르복실산 기초된 링커인 면역접합체.
- 제 12 항에 있어서, 상기 링커가 비-절단성 링커인 면역접합체.
- 제 17 항에 있어서, 상기 링커가 N-숙신이미딜-[(N-말레이미도프로피온아미도)-테트라에틸렌글리콜] 에스테르(NHS-PEG4-말레이미드)인 면역접합체.
- 제 17 항에 있어서, 상기 링커가 N-숙신이미딜 4-(말레이미도메틸) 시클로헥산카르복실레이트(SMCC)인 면역접합체.
- 제 12 항에 있어서, 상기 링커가 절단성 링커인 면역접합체.
- 제 20 항에 있어서, 상기 링커가 N-숙신이미딜 4-(2-피리딜디티오)-2-설포부타노에이트(설포-SPDB)인 면역접합체.
- 제 12 항에 있어서, 상기 링커가 N-숙신이미딜 4-(2-피리딜디티오)펜타노에이트(SPP); N-숙신이미딜 4-(2-피리딜디티오)-2-설포펜타노에이트(설포-SPP); N-숙신이미딜 4-(2-피리딜디티오)부타노에이트(SPDB); N-숙신이미딜 4-(2-피리딜디티오)-2-설포부타노에이트(설포-SPDB); N-숙신이미딜 4-(말레이미도메틸) 시클로헥산카르복실레이트(SMCC); N-설포숙신이미딜 4-(말레이미도메틸) 시클로헥산카르복실레이트 (설포SMCC); N-숙신이미딜-4-(요오드아세틸)-아미노벤조에이트 (SIAB); 및 N-숙신이미딜-[(N-말레이미도프로피온아미도)-테트라에틸렌글리콜] 에스테르(NHS-PEG4-말레이미드)로 이루어진 군으로부터 선택되는 면역접합체.
- 제 12 항에 있어서, 상기 세포독성제가 메이탄시노이드, 독소루비신, 벤조디아제핀, 탁소이드, CC-1065, 듀오카르마이신, 칼리키아미신, 돌라스타틴, 토메이마이신, 렙토마이신 및 아우리스타틴으로 이루어진 군으로부터 선택되거나, 또는 상기 세포독성제의 전구 약물인 면역접합체.
- 제 23 항에 있어서, 상기 세포독성제가 메이탄시노이드인 면역접합체.
- 제 24 항에 있어서, 상기 세포독성제가 N(2')-데아세틸-N(2')-(3-메르캅토-1-옥소프로필)-메이탄신(DM1) 또는 N(2')-데아세틸-N(2')-(4-메르캅토-4-메틸-1-옥소펜틸)-메이탄신(DM4)인 면역접합체.
- 제 12 항 내지 제 25 항 중 어느 한 항에 있어서, 2-6 개 (C) 를 추가로 포함하는 면역접합체.
- 제 1 항 내지 제 6 항 중 어느 한 항의 항체 및 약제학적으로 허용가능한 담체를 포함하는, 종양 성장 억제 또는 암 치료용 약제학적 조성물.
- 제 12 항 내지 제 25 항 중 어느 한 항의 면역접합체 및 약제학적으로 허용가능한 담체를 포함하는, 종양 성장 억제 또는 암 치료용 약제학적 조성물.
- 제 28 항에 있어서, 면역접합체가 (A) 당 2-6 개 (C)를 포함하는 약제학적 조성물.
- 제 1 항 내지 제 6 항 중 어느 한 항의 항체 또는 이의 항원 결합 단편을 포함하는, 암 진단용 시약.
- 제 30 항에 있어서, 방사성표지(radiolabel), 형광단(fluorophore), 발색단(chromophore), 영상화제(imaging agent) 및 금속 이온(metal ion)으로 이루어진 군으로부터 선택되는 표지(label)를 추가로 포함하는 시약.
- 제 1 항 내지 제 6 항 및 제 12 항 내지 제 25 항 중 어느 한 항의 항체 또는 면역접합체를 포함하는, 종양 성장 억제 또는 암 치료용 키트.
- 제 1 항 내지 제 6 항 및 제 12 항 내지 제 25 항 중 어느 한 항의 항체 또는 면역접합체를 포함하는, 개체에서 종양 성장을 억제하기 위한 의약.
- 제 33 항에 있어서, 종양이 난소 종양, 뇌 종양, 유방 종양, 자궁 종양, 자궁내막 종양, 췌장 종양, 신장 종양, 복막 종양 및 폐 종양으로 이루어진 군으로부터 선택되는 의약.
- 제 1 항 내지 제 6 항 및 제 12 항 내지 제 25 항 중 어느 한 항의 항체 또는 면역접합체를 포함하는, 개체에서 암을 치료하기 위한 의약.
- 제 35 항에 있어서, 암이 난소암, 뇌암, 유방암, 자궁암, 자궁내막암, 췌장암, 신장암, 복막암 및 폐암으로 이루어진 군으로부터 선택되는 의약.
- 제 1 항 내지 제 6 항 중 어느 한 항의 항체 또는 이의 항원 결합 단편을 생산하는 분리된 세포.
- 제 1 항 내지 제 6 항 중 어느 한 항의 항체 또는 이의 항원 결합 단편을 암호화하는 핵산 서열을 포함하는 분리된 폴리뉴클레오티드.
