Description
本発明は、全てが2008年2月5日に出願された米国仮出願第61/026,300号、第61/026,309号、第61/026,324号、第61/026,336号、第61/026,357号、及び第61/026,374号の優先権を主張し、その開示内容全体が参考として本明細書で援用される。
本発明は、微生物、例えば尿のような生体サンプル中の微生物、例えばバクテリアの同定及び定量化を行うためのシステムに関する。より具体的には、本発明は、使い捨てカートリッジ及び先細り表面を有する光学カップ又はキュベットと、光学式読取装置及び熱コントローラを含む光学システムと、光学式分析装置及び改良された分光計とを備えるシステムに関する。このシステムは、その使い捨てカートリッジをサンプルプロセッサで利用することができ、光学カップ又はキュベットを光学式分析装置で利用することができる。
サンプルの温度を冷却して制御するためのシステムは光学式分析装置内に位置する。
光学式分析装置内のサンプルの温度を冷却して制御するための前記システムは、周囲温度から所望の温度にサンプルを冷却し、且つ光学式分析装置でのサンプルの処理が完了するまでサンプルの温度を所望の温度に実質的に維持するように構成される。
ムから約8インチ離れている。本発明において、図19のこの照明ビーム234の長さは、図18の照明装置216によって減少され、照明装置216はフィルタホイール223を組み込んでいる。フィルタホイール223は、複数の狭帯域フィルタ、即ち、紫外線領域のフィルタを担持してもよい。本例において、図18の光源218からの放射は、260nm〜300nmの範囲の波長に制限され得る。或いは、フィルタホイール223は、全光スペクトル及び関連する波長を提供するフィルタを担持してもよい。また、上記に論じたように、フィルタホイール223の代わりに線形可変(linear varying)フィルタを使用してもよい。図18の照明装置216の第1の光学システム220及び第2の光学システム221におけるターニングミラー(図示しない)は、主に紫外帯域を反射するカスタムフィルタである。
別の態様に係る光学分析における複数の光学カップ内のサンプルの温度を冷却及び制御するシステムにおいて、
各々が光学式分析装置による光学分析対象のサンプルを含有する使い捨て光学カップを支持する複数の使い捨てカートリッジを支持するためのカルーセルであって、各々が前記使い捨てカートリッジの一つと関連する複数の開口を有するカルーセルと、
各々が前記カルーセル内の開口の一つと関連する複数の開口を有するターンテーブルと、
熱電クーラーと、
前記サンプルの温度を冷却して制御するために前記ターンテーブルの複数の開口を通して冷却空気を循環するための前記ターンテーブルに関連するファンと、を備える。
好適には、前記システムにおいて、前記ターンテーブルはアルミニウム製であり、前記光学カップ及び前記使い捨てカートリッジはプラスチック製であり、前記アルミニウム製のターンテーブル及び前記プラスチック製の使い捨てカートリッジ及び前記光学カップを経由する対流冷却が発生して前記光学カップ内のサンプルを急速に冷却する。
好適には、前記システムにおいて、前記サンプルの温度を冷却して制御するためのシステムは光学式分析装置内に位置する。
好適には、前記システムにおいて、前記光学式分析装置内の前記サンプルの温度を冷却して制御するための前記システムは、周囲温度から所望の温度に前記サンプルを冷却し、且つ前記光学式分析装置での前記サンプルの処理が完了するまで前記サンプルの温度を所望の温度に実質的に維持するように構成される。
別の態様に係る光学式分析装置における複数の光学カップ内のサンプルの温度を冷却して制御するためのシステムは、
各々が光学式分析装置による光学分析対象の前記サンプルを含有する使い捨て光学カップを支持する複数の使い捨てカートリッジを支持するためのカルーセルであって、各々が使い捨てカートリッジの一つと関連する複数の開口を有するカルーセルと、
各々が前記カルーセル内の開口の一つと関連する複数の開口を有するターンテーブルと、
前記サンプルを冷却して前記サンプルの温度を制御するために、熱電クーラーから前記ターンテーブルへ冷却空気を運搬するための、且つ前記ターンテーブルから前記熱電クーラーへ冷気を運搬するための、前記ターンテーブルに関連する複数の通路を有する、前記ターンテーブル下方に位置するアルミニウムブロックと、を備える。
好適には、前記別の態様のシステムにおいて、前記ターンテーブルはアルミニウム製であり、前記光学カップ及び前記使い捨てカートリッジはプラスチック製であり、前記アルミニウム製のターンテーブル及び前記プラスチック製の使い捨てカートリッジ及び前記光学カップを経由する対流冷却が発生して前記光学カップ内のサンプルを急速に冷却する。
好適には、前記別の態様のシステムにおいて、前記サンプルの温度を冷却して制御するためのシステムは光学式分析装置内に位置する。
好適には、前記別の態様のシステムにおいて、前記光学式分析装置内の前記サンプルの温度を冷却して制御するための前記システムは、所望の温度に前記サンプルを冷却し、且つ前記光学式分析装置での前記サンプルの処理が完了するまで前記サンプルの温度を所望の温度に実質的に維持するように構成される。
別の態様に係る光学カップに含有される生体液体サンプル中の汚染物質の存在を分析するための光学式読取装置内で使用するための分光計は、
前記生体液体サンプルを含有する前記光学カップから照明ビームを受光するための集光レンズシステムと、
前記光学カップから出た後に第1の光路を移動する前記照明ビームが通過する前記集光レンズシステムに隣接配置された分光計スリットと、
前記第1の光路の前記照明ビームを受光するための前記分光計スリットに隣接配置された第1の円柱レンズと、
前記第1の円柱レンズを通過する前記照明ビームをコリメート光にするため、且つ第2の光路内に前記照明ビームを反射するため少なくとも第1のミラーを含むミラーシステムと、
前記第1のミラーから反射された前記照明ビームを受光し、前記照明ビームをスペクトル成分に分光して複数の分光ビームを形成し、第3の光路に沿って前記分光ビームを反射するための前記第2の光路に位置する平面回折格子と、
前記ミラーシステムに対して位置決めされる前記第2の円柱レンズと、
前記ミラーシステムは前記複数の分光ビームを受光するため及び第4の光路の前記第2の円柱レンズに向かって前記複数の分光ビームを集束するための少なくとも第2のミラーを更に含み、
前記サンプル中の汚染物質の存在を分析するための前記第2の円柱レンズを通過する前記複数の分光ビームを受光するための前記第2の円柱レンズに隣接配置されたCCDデバイスを備える。
別の態様に係る光学分析に使用するための生体サンプルを保持するための光学カップは、
前記生体サンプルを含有する容器を備え、
前記容器は、前記生体サンプルを受容するための開口、及び前記開口に対して内側下方に延出する下部先細り領域を有し、
前記下部先細り領域は、前記開口を通った照明ビームを受け取るために、および、前記生体サンプルを通った前記照明ビームを反射させるために、形成された表面を有する。
Claims (7)
- 光学カップに含有された生体サンプルの光学分析のための光学式読取装置に発生したコリメートビームの信号対ノイズ比を増加する方法であって、
a)照明ビームを生成するための光源を提供するステップと、
b)前記光源の照明ビームの移動路を屈曲するようにフィルタ及びターニングミラーを含む第1の光学システムに前記照明ビームを向けるステップと、
c)ステップb)で生成した前記照明ビームの移動路を45°の角度で屈曲するようにフィルタ及びターニングミラーを含む第2の光学システムにステップb)で生成した前記照明ビームを向けるステップと、
d)前記生体サンプル内のバクテリアを分析するために、サンプルの底部のみを観察するよう制御された集光デバイス、次に検出デバイスに向けられる蛍光発光を生成するために、前記光学カップ内の生体サンプルに向けられるコリメートビームを生成するため、ステップc)で得られた前記照明ビームをスリットに向けるステップであって、ここで、ステップc)で得られた前記照明ビームを、前記サンプルと平行して前記サンプル内を一往復横断した後、入射時の移動路に沿って反射されるように制御するステップと、を含む方法。 - 前記光学カップは、前記生体サンプルを受容するための開口、及び前記開口に対して内側下方に延出する下部先細り領域を有し、前記下部先細り領域は、前記開口を通った照明ビームを受け取るために、および、前記生体サンプルを通った前記照明ビームを反射させるために形成された表面を有する請求項1に記載の方法。
- 前記光学カップは反射表面を有する矩形容器を含み、平行に間隔を空けた2つの側壁と、平行に間隔を空けた2つの端壁と、水平床とを有し、前記2つの端壁のうち一方の端壁である第1の端壁は前記水平床に連続する先細り領域を備えている請求項2に記載の方法。
- 前記照明ビームが、前記光学カップの床を照射しないように制御され、前記集光デバイスが、前記光学カップの床のみを観察するように制御された請求項3に記載の方法。
- 前記光源が、キセノン、LED、及びジュウテリウムの1つを含む請求項1に記載の方法。
- 前記第1の光学システムと前記第2の光学システムとの間に、フィルタホイール又は線形可変フィルタの少なくとも一方を設けるステップを含む請求項1に記載の方法。
- 前記信号対ノイズ比が73である請求項1に記載の方法。
