Abstract
本发明涉及一种微生物活菌组合制剂及其制备方法。该制剂的活菌成份是枯草芽孢杆菌(Bacillus subtilis)和粘红酵母(Rhodotorula glutinis);其制备采用微生物液体深层发酵混菌培养技术生产,先接种枯草芽孢杆菌,后接种粘红酵母,联合培养时间15-100小时。该制剂能显著提高畜禽机体抗病力和饲料吸收利用能力,且生产成本较低。
Description
技术领域:
本发明涉及一种微生物活菌组合制剂及其制备方法,属于微生物发酵工程技术领域。
背景技术:
自上世纪中叶发现饲料中添加抗生素具有明显抑菌、促生长效果以来,抗生素等药物性饲料添加剂对预防动物疾病、提高动物生产性能和经济效益方面发挥了巨大作用,推动了畜禽养殖业尤其是集约化养殖业的发展。但是后来人们发现,在饲料中长期使用和滥用抗生素,产生了另人担忧的问题;病原菌抗性显著增强,动物自身免疫力下降,药物残留引起人类健康危害,排泄物引起环境污染等,严重影响了人类的公共卫生和安全。各国纷纷采取措施对饲用抗生素的种类、剂量和使用方法等方面严加限制,欧盟已决定2006年开始全面禁止在饲料中使用抗生素。
微生态制剂是指将已知的有益微生物通过现代生物工程技术制备成应用于动物的生物制剂,它能有效补充畜禽体内的有益微生物,改善动物体内菌群平衡,提高机体抗病力和饲料吸收利用能力。由于微生态制剂无残留、不产生抗性,且具有预防疾病和促进动物生长的作用,可有效克服长期使用抗生素造成的一些不良影响,被认为是饲用抗生素潜在的、天然的、对环境和健康无害的替代物。微生态制剂按微生物种类可分为芽孢杆菌制剂、乳酸杆菌制剂、酵母制剂及复合制剂等,它们在畜禽及水产养殖方面的应用发展很快,1989年全球饲用微生态制剂总销售额为7500万美元,1993年为1.22亿美元,2000年为1.86亿美元,2006年估计可达3.5亿美元。
发明内容:
本发明目的是要提供一种新型微生物活菌组合制剂。
为实现上述目的采取的技术方案:采用复合微生态制剂,其活菌成份是枯草芽孢杆菌(Bacillus subtilis)和粘红酵母(Rhodotorula glutinis),采用液体深层发酵混菌培养技术生产。
该制剂能显著提高机体抗病力和饲料吸收利用能力,且生产成本低廉。
本发明的具体内容如下:
1.活性菌株鉴定:
(1)枯草芽孢杆菌ZHBal 78:
菌体杆状,很少成链,通常为0.7-0.8×2.0-3.0微米,革兰氏阳性,细胞原生质染色均匀,运动、周生鞭毛。芽孢椭圆形或柱状,0.9×0.6微米,中生或偏中生,菌落呈细皱状、干燥或颗粒状。生长温度15-55℃,接触酶和V-P反应阳性,溶菌酶阳性,V-P培养液中的pH 5.8,在7%氯化钠中生长,pH 5.7生长。利用葡萄糖、蔗糖、阿拉伯糖、木糖和甘露醇产酸,水解淀粉,液化明胶,能利用柠檬酸盐,还原硝酸盐成亚硝酸盐,分解酪素,不分解酪氨酸,石蕊牛奶产碱胨化。需氧菌,厌氧生长和卵黄反应阴性,在葡萄糖琼脂和酪氨酸琼脂上不形成可溶性黑色素,不水解马尿酸盐,不利用丙酸盐。
(2)粘红酵母ZHRh028:
细胞卵形到球形,2.3-5.0×4.0-7.8微米,单个或几个细胞连结成串,多端出芽。在麦芽汁琼脂斜面上菌苔呈现珊瑚红到橙红色,菌落表面光滑、有光泽、粘稠有时发硬,其横切面扁平到有较宽的凸起,边缘不规则到整齐,不形成菌丝,无假菌丝。不发酵任何糖,同化硝酸钾,分解杨梅苷,不产生类淀粉。可同化葡萄糖、麦芽糖、半乳糖、蔗糖、海藻糖、纤维二糖、L-山梨糖、乙醇、D-果糖、松三糖、甘油等,不能同化乳糖、蜜二糖、D-甘露糖、鼠李糖、D-木糖、菊糖、棉子糖、肌醇、可溶性淀粉、D-阿拉伯糖、L-阿拉伯糖、D-山梨醇和赤癣醇。无有性生殖现象,生长温度25-36℃,pH 2.5-10.0范围均能生长。
2.制备工艺:
(1)生产菌种:
本发明所用的微生物菌种为枯草芽孢杆菌ZHBa178(Bacillus subtilis ZHBa178),保藏于CGMCC,保藏号CGMCC NO.1744,保藏日期为2006年06月27日;粘红酵母ZHRh028(Rhodotorula glutinis ZHRh028),保藏于CGMCC,保藏号CGMCC NO.1745,保藏日期为2006年06月27日。
(2)培养基:
①种子斜面培养基:
枯草芽孢杆菌:牛肉膏0.5% 蛋白胨1.0% 氯化钠0.5%
葡萄糖0.2% 琼脂2.0% pH 7.0-7.2
粘红酵母: 葡萄糖2% 蛋白胨1.0% 酵母膏0.2%
琼脂2.0% pH 6.0
②发酵培养基:
培养基中的营养源采用常用的碳源和氮源。其中碳源可为淀粉、糊精、蔗糖、葡萄糖、饴糖等;氮源可为蛋白胨、黄豆饼粉、酵母膏、玉米浆、尿素、硫酸铵等。在此基础上再添加一些其它营养成份,如食盐、磷酸盐、钾盐、钙盐、镁盐等。
(3)培养方法:
本发明所述的微生物活菌组合制剂,通过液体深层发酵技术分步接种混和培养。首先接种枯草芽孢杆菌,培养温度为34-39℃;培养10-30小时后接种粘红酵母,培养温度降至28-33℃,联合培养15-100小时后放罐。
(4)产品后处理:
培养液调制成一定的含菌量,添加适量防腐剂后分装,即为水剂产品;或加入适量吸附剂吸附,干燥粉碎后分装,即为粉剂产品。

本发明的显著优点:
1.多效:
产品富含枯草芽孢杆菌和粘红酵母二种有益活性菌,以及由二菌培养时代谢产生的β-胡萝卜素、维生素、及多种酶等生理活性物质,因而具备多重功效。具有改善畜禽体内菌群平衡,提高动物免疫力,增加饲料适口性,促进动物对饲料的消化吸收,以及净化水质提高水产养殖环境质量等功能。
2.安全:
本产品系由有益菌组成的活菌制剂,动物饲用后无任何副作用,无残留问题,不引起耐药性,是相对非常安全的活性饲料添加剂。
3.低成本:
生产采用枯草芽孢杆菌、粘红酵母分步接种后混和培养工艺,与分别培养后混和工艺相比,可大大降低原材料成本和能源动力消耗,因而成本明显降低。

具体实施方式:
本发明有以下实施例:
实施例一:活菌总数大于50亿/ml水剂产品的制备
在70L发酵罐中,按葡萄糖3%、硫酸铵0.4%、酵母膏0.2%、磷酸氢二钾0.02%、硫酸镁0.02%配成50L培养基。按常规方法灭菌后冷却至37℃,接种1个茄子瓶的枯草芽孢杆菌生产菌种,培养温度为37℃,转速250转/分;培养18小时后接种1个茄子瓶粘红酵母生产菌种,培养温度降至28℃,联合培养20小时后放罐。加水稀释成总活菌数50亿/ml以上的悬液,再加0.1%苯甲酸钠作防腐剂,分装即得产品。该产品活菌总数大于50亿/ml,其中枯草芽孢杆菌大于35亿/ml,粘红酵母大于15亿/ml。
实施例二:总活菌数大于100亿/克粉剂产品的制备
在70L发酵罐中,按蔗糖4.0%、玉米浆0.5%、尿素0.3%、磷酸氢二钾0.02%、硫酸镁0.02%、氯化钠0.2%配成45L培养基。按常规方法灭菌后冷却至35℃,接种1个茄子瓶的枯草芽孢杆菌生产菌种,培养温度为36℃,转速220转/分;培养24小时后接种2个茄子瓶粘红酵母生产菌种,培养温度降至30℃,联合培养76小时后放罐。加适量玉米芯粉吸附并调制到总活菌数100亿/克以上,沸腾干燥后分装即为产品。该产品总活菌数大于100亿/克,其中枯草芽孢杆菌大于60亿/克,粘红酵母大于40亿/克。
Claims (2)
1、一种微生物活菌组合制剂,其特征是制剂的活菌成份为枯草芽孢杆菌Bacillussubtilis和粘红酵母Rhodotorula glutinis。
2、一种专用于权利要求1所述微生物活菌组合制剂的生产方法,采用微生物液体深层发酵混菌培养技术生产,其制备工艺的特征如下:
(1)生产菌种:
采用枯草芽孢杆菌Bacillus subtilis ZHBa178和粘红酵母Rhodotorula glutinisZHRh028,菌种保藏于CGMCC,保藏号分别为CGMCC NO.1744和CGMCC NO.1745;
(2)发酵培养基:
培养基中的营养源采用碳源和氮源,其中碳源为淀粉、糊精、蔗糖、葡萄糖或饴糖;氮源为蛋白胨、黄豆饼粉、酵母膏、玉米浆、尿素或硫酸铵,并添加食盐、磷酸盐、钾盐、钙盐或镁盐;
(3)培养方法:
通过液体深层发酵技术分步接种混和培养,首先接种枯草芽孢杆菌,培养温度为34-39℃,培养10-30小时后接种粘红酵母,培养温度降至28-33℃,联合培养15-100小时后放罐;
(4)产品后处理:
培养液调制后,再添加防腐剂,分装即为水剂产品;或加入吸附剂吸附,干燥粉碎后分装即为粉剂产品。
