G1=군 1; G2=군 2; G3=군 3; G4=군 4; G5=군 5; G6=군 6.

도 25a-b는 독소 투여되거나 독소 부재하에 이어서 SCS 또는 유리체내로 투여된 저 또는 고 용량의 TA로 처리된 동물의 눈의 와이드-필드 안저 이미지이다. 와이드-필드 안저 이미지는 -1일에 리포폴리사카라이드 (LPS) 주입 직전에, 처리 후 0일 및 3일에 비히클, 0.2 mg (저 용량) 또는 2.0 mg (고 용량)의 트리암시놀론 아세토니드 주입 직전에 이미지화한 것이다. 평형 염 용액 및 비히클 주입된 군 1 눈은 정상적인 외형을 유지하였다. 군 1 눈을 제외한 모든 눈에서 LPS 주입 후 24시간째에 후안부에 실질적인 혼탁함이 발생하였다. SCS로의 저 용량 및 고 용량 mg TA 및 고 용량 TA IVT로의 처리는 처리전 모양과 가까운 안저 이미지를 유도한 반면, 저용량 TA IVT로의 처리는 비히클 처리된 눈에 비해 단지 약간 개선된 이미지를 유도하였다. 2.0 mg TA IVT 주입된 눈은 중심 유리체에서 가시적인 TA의 큰 고형 데폿 (depot)을 가졌다.
도 26은 평형 염 용액 (BSS) 또는 100 ng의 리포폴리사카라이드 (LPS)의 유리체내 주입 후 3일에 또는 비히클, 0.2 mg TA, 또는 2.0 mg TA의 SCS 또는 IVT 투여 후 72 시간에 눈의 안구 조직병리를 보여준다. 검사한 눈 어느 것도 조직병리에서 실질적인 조직, 구조 또는 독소 변화의 증거를 갖지 않았다. 평형 염 용액 (BSS) 또는 100 ng의 리포폴리사카라이드 (LPS)의 유리체내 (IVT) 주입 후 3일 및 비히클, 0.2 mg 트리암시놀론 아세토니드 (저 용량 TA), 또는 2.0 mg의 트리암시놀론 아세토니드 (고 용량 TA)의 맥락막위 (SCS) 또는 IVT 주입 후 72 시간에서의 눈의 안구 조직병리. 헤마톡실린 및 에오신 염료.

C. LPS IVT 및 SCS로 비히클 주입된 전안부 (군 2). 기준 막대: 1 mm.

E. LPS IVT 및 SCS로 저 용량 TA 주입된 전안부 (군 3). 기준 막대: 1 mm.

Claims (240)
- 후안구 장애(posterior ocular disorder)의 치료가 필요한 인간 대상체에서 후안구 장애를 치료하는데 사용하기 위한 약물 제형으로서,
약물 제형은 후안구 장애의 치료가 필요한 인간 대상체의 눈의 맥락막위 공간(SCS)으로 수술 형태를 제외한 비-외과적(non-surgical) 투여를 위해 제형화되고,
약물 제형이 투여되면 삽입 부위로부터 유동하여 눈의 후안부에 국소화되며,
약물이 트리암시놀론이고,
후안구 장애가 포도막염 (uveitis) 또는 포도막염-관련 황반 부종 (uveitis-associated macular edema)이고,
비-외과적 투여가 개구를 지닌 팁 말단을 가지며 500 μm 내지 2000 μm의 길이를 갖는 중공 바늘을 삽입 부위에서 공막 내로 삽입하고, 삽입된 바늘을 통해 SCS로 약물 제형을 주입하는 것을 포함하고, 상기 비-외과적 투여가 션트 (shunt) 또는 캐눌라 (cannula)를 사용하지 않고 수행되는, 약물 제형. - 제 1항에 있어서, 상기 바늘이 공막 내로 80°내지 100°의 각도로 삽입되는, 약물 제형.
- 제 1항에 있어서, 마이크로입자의 현탁액을 포함하는 약물 제형.
- 제 1항에 있어서, 나노입자의 현탁액을 포함하는 약물 제형.
- 제 3항에 있어서, 마이크로입자의 D50이 2 μm 또는 그 미만인 약물 제형.
- 제 3항에 있어서, 마이크로입자의 D99가 10 μm 또는 그 미만인 약물 제형.
- 제 3항에 있어서, 마이크로입자의 D99가 5 μm 또는 그 미만인 약물 제형.
- 제 4항에 있어서, 나노입자의 D99가 10 μm 또는 그 미만인 약물 제형.
- 제 4항에 있어서, 나노입자의 D99가 5 μm 또는 그 미만인 약물 제형.
- 제 1항에 있어서, 트리암시놀론이 트리암시놀론 아세토니드인 약물 제형.
- 제 1항 내지 제 10항 중의 어느 한 항에 있어서, SCS로 투여되는 경우 치료 반응을 유발시키기에 충분한 약물의 투여량이 유리체내, 앞방내, 국소, 비경구 또는 경구 투여되는 경우의 치료 반응을 유발시키기에 충분한 약물의 투여량보다 적은 약물 제형.
- 제 1항 내지 제 10항 중의 어느 한 항에 있어서, 눈의 후안부에서의 약물 보유량이 유리체내, 앞방내, 국소, 비경구 또는 경구 투여되는 경우의 눈의 후안부에서의 약물 보유량보다 많은 약물 제형.
- 제 1항 내지 제 10항 중의 어느 한 항에 있어서, 약물의 전신 노출량이, 약물이 유리체내, 앞방내, 국소, 비경구 또는 경구 투여된 경우의 약물의 전신 노출량보다 적은 약물 제형.
