도 6. 시토케라틴 (녹색)으로 및 염색체 21 (적색) 및 X 염색체 (청색)에 대한 FISH 프로브로 염색된 태아 세포. 화살표는 태아 세포에서 염색체 21의 세 개의 카피를 가리킨다. 모체 세포의 배경은 각각 두 개의 카피의 염색체 21을 함유한다.
Claims (18)
- 모체 혈액 샘플에서 태아 세포를 분리하는 방법으로서, 상기 방법은
a. 모체 혈액 샘플 또는 이들의 분획을 내피 세포 마커를 암호화하는 유전자 또는 내피 세포 마커에 관련된 리간드에 상보성인, 적어도 10개의 인접한 뉴클레오티드를 포함하는 잡종화 프로브와 접촉시키는 단계로서, 상기 내피 세포 마커는 CD105, CD146, CD141, 비멘틴, VCAM, ICAM, VEGFR-1, VEGFR-2, VEGFR-3, ITGA5, ITGB5, CDH11 또는 CDH3으로 구성된 그룹으로부터 선별되고,
b. 상기 내피 세포 마커에 특이적인 세포를 선별하고 이로 인해 내피 세포의 표현형을 갖는 세포에 대해 샘플 또는 이들의 분획을 농화하는 단계
c. 내피 세포 표현형을 나타내는, b)에서 선별된 세포를 상피 세포 마커를 암호화하는 유전자에 상보성인, 적어도 10개의 인접한 뉴클레오티드 또는 상피 세포 마커에 관련된 리간드를 포함하는 잡종화 프로브와 접촉시키는 단계로서, 상기 상피 세포 마커는 CK1, CK2, CK3, CK4, CK5, CK6, CK7, CK8, CK9, CK10, CK13, CK14, CK15, CK16, CK17, CK18 또는 CK19로 구성된 그룹으로부터 선별되고,
d. 단계 c)의 상피 세포 마커에도 결합하는 내피 세포의 표현형을 갖는 세포를 검출하는 단계
를 포함하며, 단계 a-d는 어떤 순서로도 수행될 수 있는 방법. - 제 1항에 있어서, 내피 세포 마커는 확인할 세포의 표면에 위치하는 것을 특징으로 하는 방법.
- 제 1항에 있어서, 상피 세포 마커는 확인할 세포의 세포 내에 위치하는 것을 특징으로 하는 방법.
- 내피 및 상피의 특징을 갖는 세포에 대하여 모체 혈액 샘플 또는 이들의 분획을 농화하는 방법으로서, 상기 방법은
a. 모체 혈액 샘플 또는 이들의 분획을
i. 내피 세포 마커를 암호화하는 유전자에 상보성인, 적어도 10개의 인접한 뉴클레오티드를 포함하는 잡종화 프로브 또는
ii. 내피 세포 마커에 관련된 리간드
와 접촉시키는 단계로서,
상기 내피 세포 마커는 CD105, CD146, CD141, 비멘틴, VCAM, ICAM, VEGFR-1, VEGFR-2, VEGFR-3, ITGA5, ITGB5, CDH11 또는 CDH3으로 구성된 그룹으로부터 선별되고,
b. 이전 단계의 잡종화 프로브 또는 리간드에 결합하는 세포를 선별하고 이로 인해 이전 단계의 잡종화 프로브 또는 리간드에 결합하는 세포에 대해 샘플 또는 이들의 분획을 농화하는 단계
c. 샘플 또는 이들의 분획을
i. 상피 세포 마커를 암호화하는 유전자에 상보성인, 적어도 10개의 인접한 뉴클레오티드를 포함하는 잡종화 프로브 또는
ii. 상피 세포 마커에 관련된 리간드
와 접촉시키는 단계로서,
상기 상피 세포 마커는 CK1, CK2, CK3, CK4, CK5, CK6, CK7, CK8, CK9, CK10, CK13, CK14, CK15, CK16, CK17, CK18 또는 CK19로 구성된 그룹으로부터 선별되고,
d. 이전 단계 c.의 잡종화 프로브 또는 리간드에 또한 결합하는 세포를 선별하고 이로 인해 내피 및 상피의 특징을 갖는 세포에 대해 샘플 또는 이들의 분획을 농화하는 단계
를 포함하는 방법. - 제 4항에 있어서, 샘플 또는 이들의 분획의 태아 세포를 확인하는 단계를 추가로 포함하는 것을 특징으로 하는 방법.
- 제 5항에 있어서, 확인은 태아 세포 상에 또는 안에 상피 세포 마커 관련된 리간드 또는 상피 세포 마커 관련된 잡종화 프로브의 존재를 검출하는 단계를 포함하는 것을 특징으로 하는 방법.
- 제 1항에 있어서, 모체 혈액 샘플은 전혈인 것을 특징으로 하는 방법.
- 제 1항에 있어서, 샘플 또는 이들의 분획의 세포는 선별 단계 후 고정 단계를 받게 되는 것을 특징으로 하는 방법.
- 제 8항에 있어서, 샘플 또는 이들의 분획의 세포는 고정 단계 후 침투화제와 접촉시킴으로써 침투화되는 것을 특징으로 하는 방법.
- 제 1항에 있어서, 내피 세포 마커는 CD105, CD146 또는 CD141로 구성된 그룹으로부터 선별되는 것을 특징으로 하는 방법.
- 제 1항에 있어서, 내피 세포 마커는 CD105인 것을 특징으로 하는 방법.
- 제 1항에 있어서, 상피 세포 마커는 CK8, CK18, CK19 또는 CK7로 구성된 그룹으로부터 선별되는 것을 특징으로 하는 방법.
- 제 4항에 있어서, 모체 혈액 샘플은 전혈인 것을 특징으로 하는 방법.
- 제 4항에 있어서, 샘플 또는 이들의 분획의 세포는 선별 단계 후 고정 단계를 받게 되는 것을 특징으로 하는 방법.
- 제 14항에 있어서, 샘플 또는 이들의 분획의 세포는 고정 단계 후 침투화제와 접촉시킴으로써 침투화되는 것을 특징으로 하는 방법.
- 제 4항에 있어서, 내피 세포 마커는 CD105, CD146 또는 CD141로 구성된 그룹으로부터 선별되는 것을 특징으로 하는 방법.
- 제 4항에 있어서, 내피 세포 마커는 CD105인 것을 특징으로 하는 방법.
