パエニバチルス種は、以前はバチルス属に含まれていた通性嫌気性、内生胞子形成、グラム陽性生物である。バチルスもグラム陽性の属である。本開示において、パエニバチルス及びバチルスの分離株は、植物の成長を促進し、真菌及び卵菌類(F.オキシスポルム(oxysporum)、R.ソラニ、又はP.アルティマムを含むがこれらに限定されない)によって引き起こされる苗の立枯れを抑制するために使用される。一実施形態では、パエニバチルス及びバチルスの分離株は、より成熟した植物において病原体に感染した根、栄養組織、及び/又は生殖組織を制御する。

実施例1 リゾクトニア及びピシウムのin vitro阻害
4つの分離株MS1479、MS2379、MS2414、及びMS2820を、真菌病原体に対して、4つの生産培地であるTSB培地(30g/Lのトリプチックソイブロス(TSB、Sigma-Aldrich 78907))、BS3培地、BS3-M2、及びGB6-M3で試験した。
4つのパエニバチルス分離株の生物活性に対するGB6培地における窒素源の影響を調査するために、GB6-Mベースの培地を、窒素源を変更して抗生物質の活性を高めることによりさらに改変した。それぞれ250mlの生産培地を含む16個の1Lバッフル付きフラスコに、LB培地からの2%(5ml)の種菌を接種した。これらを26℃、200rpmで72時間培養した。4つの生産培地には、GB6-M3、GB6-M7、GB6-M8、GB6-M9が含まれる。
30個の種子を50mlの遠心チューブ内で、0.1ml/種子のWB量で処理した。処理した種子(0.1ml/種子)を室温で2ヵ月保存した。各処理から3個の種子をcfuアッセイに使用した。
図3は、全ブロス(WB)で処理したダイズ種子の結果を示し、TSB、BS3、GB6-M、又はLB培地で培養したMS1479、MS2379、MS2379、及びMS2414は比較的安定しており、2ヵ月の保管後も、種子処理直後に採取した初期cfuと同様のcfu/mlを含む。処理された種子は、畑に播種する前に、合理的な期間、例えば、少なくとも2ヵ月間保存できると考えられる。
パエニバチルス処理及び未処理のダイズ種子を滅菌土壌で発芽させ、8日後に根を洗浄し、滅菌水中で5分間高出力で超音波処理した。次に、スパイラルプレーターを使用してこの水を塗抹した。図4に示すように、未処理のダイズ種子からの根は、細菌コロニーの数が少なく形態も様々であり、処理された種子は、均一で優勢なコロニー型のパエニバチルスのコロニーを多数生成した。この結果から、処理された種子ではパエニバチルスが持続し、土壌環境において苗の根で増殖したことが示唆された。
全ブロスを、TSB、BS3、BS3-M2、又はGB6-M3培地で増殖させたMS1479、MS2379、MS2414、及びMS2820から無菌的に収集し、9つの真菌病原体のin vitro防除について試験した。各分離株について2μlの全ブロスを、プレート上に細菌又は真菌病原体を有するプレート上に、又はプレートの中央に置かれた病原体コロニー化寒天キューブ上にスポットした。全てのプレートを適切な条件でインキュベートした:30℃インキュベーター(キサントモナス・ペルフォランス(Xanthomonas perforans)(“Xp”)及びシュードモナス・シリンガエトマト病原型(“Ps”));25℃インキュベーター(マクロフォミナ・ファセオリナ(“Mp”)、リゾクトニア・ソラニ(“Rs”)、ボトリチス・シネレア(“Be”));25℃グロースチャンバ(シュードモナス・シリンガエ(“Ps”)、ピシウム・アルティマム(“Pu”)、ピシウム・イレギュラーレ(“Pi”)、及びフザリウム・ビルグリフォルメ(“Fv”)。次に、プレートを、抗生作用により引き起こされた透明帯の直径について測定した。in vitro阻害データを表7に示す。
ポットアッセイでは、WB又はその希釈液を、種子コーティングとして、又は病原体を接種した土壌を含むポット内の模擬畝内施用として適用した。バイオコントロール効果を評価するために、出芽率、植物成長段階、及び病気の評価の測定値を用いた。全ブロス(WB)又はWB希釈液を、模擬畝内処理又はダイズ種子に適用される種子処理のいずれかとして適用した。模擬畝内施用については、病原体を接種したピートライトミックス培養土に形成した0.5cmの深さの窪みに種子を置き、その上に1mlのバイオコントロール処理剤又は陰性対照及び陽性対照をピペッティングして、上記培養土を被せた。種子処理は、接種した培養土に植える前のダイズ種子を、バイオコントロール処理剤及び対照処理剤でコーティングすることにより適用した。陰性対照は、接種したポットの種子に適用した逆浸透精製(RO)水とした。陽性対照は、標識された割合で適用されたSatori(商標)(アゾキシストロビン)及びSubtilex(商標)(枯草菌MBI600株)とした。さらに、病原体接種を行わない模擬接種治療を各試験に含めた。接種は、セメントミキサーを使用して、RO水200ml中のリゾクトニア・ソラニコロニー化ポテトデキストロース培養物と、4Lのピートライトミックス(例えば、Sunshine LC1ポッティングミックス)の2つを完全にブレンドすることによって作成された1Lのスラリーを完全に混合することによって行った。実験単位は1ポットあたり5個の種子とし、処理は完全に無作為化した配置で、4個の複製で実施した。試験は、LEDランプで照らされた照明カート(昼16時間/夜8時間)において、26~28℃で7~10日間実施した。ポットには、必要に応じてRO水を灌注した。出芽率、植物成長段階(Mungerら、2008年)、及び病気の重症度(0、病気なし~5、枯死)の測定値を各試験の終了時に各植物から集め、実験単位の平均値をJMPバージョン11(SAS、Cary、NC)を用いて分析した。
処理された種子を、各処理からの100個の種子とともに保持した。これらの種子は、R.ソラニ防除用の植物試験グループによって試験した。精度を高めるために、アッセイの各処理について10個の複製を行った(各処理に40の植物)。病気評価の統計分析を図6に示す。GB6-M培地で増殖させたMS2414で処理した種子は、対照と比較して耐病性が大幅に向上した。
GB6-M培地で増殖させたMS2379又はMS2414の発酵WBをアマランスと混合して、圃場試行研究用の種子処理剤として適用される混合物を生成した(表10)。全ての処理で、目標濃度は20μl水/種子であり、これは60ml/460g種子に相当する。ダイズ種子460gごとに60mlの液体を合計6回添加した。毎回、460gの種子に10mlの液体を加え、15秒間混合した。コーティングされた種子は、添加と添加の間に換気下で5~10分間乾燥させた。最終的なコーティング種子を、安全キャビネット内で一晩乾燥させた。
7つのバチルス分離株MS0633、MS2335、MS2652、MS2658、MS2681、MS2697、及びMS2712を、4つのパエニバチルス分離株(MS1479、MS2379、MS2414、MS2820)と共にバイオコントロール活性について試験した。この実験では、それぞれ50mlのLB培地を含む11個の250ml振盪フラスコに、0.1mlの凍結バイアルの解凍物又はプレートからのコロニーを接種した。種菌を30℃、200rpmで約18時間一晩培養した。それぞれ250mlのGB6-M8培地を含む11個の1リットルのバッフル付きフラスコに2%(5ml)の種菌を接種した。バチルス分離株の培養条件には、28℃及び72時間の200rpmの振盪速度が含まれていた。4つのパエニバチルス分離株の培養条件には、26℃及び72時間の200rpmの振盪速度が含まれていた。
GB6-M8培地で増殖させた11の分離株(4つのパエニバチルス及び7つのバチルス)の発酵全ブロスを、ポットでのR.ソラニ感染に対して試験した。植物の発芽、植物の発育、及び病気の重症度の点で、処理の効果は大きく異なる。種子を深さ1cmの窪みに置いた後、WBを各種子の上に1mlずつピペットで滴下することにより、模擬畝内施用として適用した。
ダイズのピシウム病に対する細菌分離株の効果を、Brodersら、Plant Dis 91:727-735(2007)の方法を変更したものを用いて試験した。100mlの漂白剤(5.25%NaOCl)と3.5mlのHCl(10N)を合わせて発生させた塩素ガスを使用してダイズの種子を表面滅菌した。次いで、ダイズ種子を、種子当たり約20μlのバイオコントロール処理剤又は対照処理剤でコーティングし、滅菌培養フード内で完全に乾燥させた。陰性対照は、種子に適用された滅菌蒸留水であった。陽性対照は、標識された割合で適用されたメタラキシル及びSubtilex(商標)(枯草菌MBI600株)であった。さらに、病原体接種のない模擬接種処理が各試験に含まれていた。水寒天(0.8%寒天)プレートの中央に、V8寒天で増殖させた7日齢のP.イレギュラーレの培養物の縁から切り取った10mm角の塊を接種した。次に、病原体が置かれた同じ日に、単一の処理の10個の種子が、各接種された水寒天プレートの周囲に直ちに無菌的に置かれた。実験単位には、プレートあたり10個の処理種子が含まれ、1つの処理につき5個の複製を行った。プレートをランダムに配置し、蛍光灯下(昼16時間、夜8時間)、16℃で7日間、さらに同じ照明下で25℃でさらに7日間インキュベートした。プレートあたり10個の種子のうち発芽した種子の数と、プレート内の各種子の植物発達段階(Mungerら、2008年)を記録し、JMPバージョン11(SAS、Cary、NC)を使用して分析した。
20L発酵槽に播種するための接種材料は、2つの1Lバッフル付きフラスコ内の2つの250mlのLB培地に、それぞれ1mlの凍結バイアルの解凍物から0.5mlを接種することにより調製し、28℃及び200rpmの振盪速度で一晩インキュベートした(約16~18時間)。インキュベーション後、MS2379の種菌は7.43のpHと3.24のOD600を示し、MS2414の種菌は7.30のpHと2.83のOD600を示した。
この実験は、病害防除のための葉面処理のUV安定性を試験するためであった。全ブロス濾液、全ブロス、ならびにMS2379及びMS2414の画分をUV光下に置いた。処理の長さは、処理するサンプルの量及び種類に応じて異なる。当業者は、UV処理の期間を決定することができる。
種子発芽アッセイは、ピシウム・イレギュラーレに対するGB6-M8培地で増殖させた分離株の全ブロスのバイオコントロール活性を調査するために使用された。図17に示すように、いくつかの全ブロス微生物処理(MS2379、MS2820、及びMS1479)で、植物の発育と種子の発芽の結果は、両方のメトリックで陽性化学物質対照メタラキシルに類似しており、Subtilexよりも優れてさえいた。
圃場調査のプロトコルを表17に示す。
GB6-M31培地で発酵させたMS2379(LPI-6592)のバイオコントロール効果を、以下の4種類の芝生の病気について、芝生の病気の確立された区画において試験した:炭疽病(コレトトリカム・セレアレ(Colletrotrichum cereale))、ブラウンパッチ病(リゾクトニア・ソラニ)、ダラースポット病(スクレロチニア・ホモエオカルパ)、及びピシウム胴枯病(ピシウム種)。炭疽病の区画には人為的に接種し、他の区画は天然の接種源で感染させた。
植物の葉の病気に対する2つの新しい培地GB6-M32及びGB6-M34でのMS2379発酵物の有効性を、GB6-M31培地のMS2379(LPI-6592)と比較した。細菌分離株を、発酵中のpH制御(pH5.8~6.0)あり又はなしで発酵させた。pH制御発酵では、滅菌ボトル内の滅菌1N NaOH又は10%H2SO4を用いて、発酵中にpHを5.6~6.0の範囲に自動的に制御した。サンプルを、分離されたキャノーラの葉に10%(v/v)の有効成分の濃度で適用した。試験には4つの対照処理を含めた:未処理対照(病原体のみを用いた未処理)。化学殺真菌剤Dyna-Shield(商標)フルジオキソニル(Lovel and Products)、有効成分は0.06%(v/v)のフルジオキソニル;市販のバイオコントロール剤Serenade ASO(商標)(Bayer Crop Science)、有効成分は3%(v/v)の枯草菌QST713;模擬接種(病原体なし)。MS2379及びSerenade ASO処理のコロニー形成ユニット(CFU)を、1mlあたり3×108内生胞子で正規化した。処理は完全乱塊法で配置し、各処理について5個の複製で実施した。温室で栽培された10日齢のキャノーラ植物から均一サイズの本葉を使用直前に切り取り、逆浸透精製水で30分間完全に洗浄した。
真菌の病気に対する有効性を試験するために、細菌分離株の濃縮も行った。GB6-M34で増殖させたMS2379(発酵中pH5.6~6.0)の発酵全ブロス4Lを、PM-500(分画分子量は500,000Da)中空繊維濾過カートリッジを搭載したKOCHデモ濾過ユニットの容器に注入した。限外濾過は、循環ポンプをオンにし、透過側の圧力を100kPa(1bar)に調整して実施した。透過液の量が2Lに達したときに濾過を停止し、非透過物を回収した。透過液を、非透過物が約10倍濃縮されるまで、PM-5(分画分子量は5,000Da)中空繊維フィルターを用いてさらに濾過した。限外濾過からの全ての非透過物及び透過液を、CFU/ml、粘度(cP)及びプロテアーゼ活性について試験した。図22に示すように、CFUは非透過物において約2倍に濃縮され(図22A)、PM5-非透過物は最も高いプロテアーゼ活性を示した(図22B)。
保存後の細菌発酵物の安定性を試験するために、事前にプロピレングリコールに溶解させた0.03%BIT(1,2-ベンズイソチアゾリン-3-オン)を発酵物(GB6-M31でのMS2379)に加えた。GB6-M31培地中のMS2379の保存されたWB約8Lを量り取り、50%クエン酸又は1N NaOHで目標pH+/-0.1となるようにpH6.5、6.0、5.5、5.0、4.5に調整した。様々な製剤成分を添加した後、必要に応じてpHを再調整した。200mlWB製剤を250mlボトルに加えたものを、同じサイズの各ボトルで2つ作成した。1セットのサンプルを25℃、もう1セットを40℃に置いた。表19に、各サンプルの製剤及びpHをまとめて示す。2セットのサンプルには0.5%プロピレングリコールを使用し、そのうちの1つは凍結融解試験に使用した。バイオプロテクター(Lallemand製)は、生物学的種子処理又は他の作用に使用できるアジュバントである。

Claims (7)
- パエニバチルス(Paenibacillus)又はバチルス(Bacillus)の細菌分離株、あるいはそれらの変異体を含む農業用組成物であって、前記細菌分離株は、BS3、BS3-M2、BS3-M9、BS3-M10、GB6-M、GB6-M3、GB6-M7、GB6-M8、GB6-M9、GB6-M22、GB6-M23、GB6-M10、GB6-M31、GB6-M33、GB6-M34、又はそれらの組合せを含む培地で発酵及び/又は培養される、農業用組成物。
- 培養培地はBS3-M2、BS3-M9、BS3-M10、GB6-M、GB6-M3、GB6-M7、GB6-M8、GB6-M9、GB6-M22、GB6-M23、GB6-M10、GB6-M31、GB6-M33、GB6-M34、又はそれらの組合せを含む、請求項1に記載の農業用組成物。
- 培養培地はGB6-M10、GB6-M31、GB6-M33、GB6-M34、又はそれらの組合せを含む、請求項1又は2に記載の農業用組成物。
- 細菌分離株は、
(1)シード培地に細菌分離株を接種すること、及び
(2)生産培地で培養物を拡大すること
を含むプロセスにより発酵される、請求項1に記載の農業用組成物。 - シード培地は、BS3、BS3-M2、BS3-M9、BS3-M10、GB6-M、GB6-M3、GB6-M7、GB6-M8、GB6-M9、GB6-M22、GB6-M23、GB6-M10、GB6-M31、GB6-M33、GB6-M34、又はそれらの組合せを含む、請求項4に記載の農業用組成物。
- 生産培地は、GB6-M10、GB6-M31、GB6-M33、GB6-M34、又はそれらの組合せを含む、請求項4に記載の農業用組成物。
- パエニバチルス(Paenibacillus)又はバチルス(Bacillus)の細菌分離株、あるいはそれらの変異体を含む農業用組成物であって、前記細菌分離株は、(i)5~20g/Lの大豆ペプトン、2~10g/Lの尿素、0~5g/LのCaCl2、2~10g/LのKH2PO4、2~10g/LのK2HPO4、及び10~30g/Lのスクロース;(ii)10~30g/Lの低脂肪大豆粉、1~5g/LのCaCl2、2~10g/LのKH2PO4、2~10g/LのK2HPO4、及び10~30g/Lのスクロース;(iii)5~30g/Lのマルトデキストリン、5~25g/Lのデキストロース、1~10g/Lの酵母エキス、0.1~5g/Lの硫酸アンモニウム、及び0.2~3g/LのCaCO3;(iv)5~40g/Lのマルトデキストリン、5~20g/Lのデキストロー ス、2~15g/Lの酵母エキス、2~15g/Lの低脂肪大豆粉、0.2~1.5g/Lの硫酸アンモニウム、及び0.5~3g/LのCaCO3;又は(v)5~20g/Lの低脂肪大豆粉、0.5~5g/LのCaCl2、2~10g/LのKH2PO4、2~10g/LのK2HPO4、10~30g/Lのスクロース、及び0.1~5g/Lの硫酸アンモニウムを含む培地で発酵及び/又は培養される、農業用組成物。
