Claims (32)
- 효소의 활성에 대한 나트륨이온의 영향을 측정함으로써 유체중에 있는 나트륨이온을 측정하는 방법.
- 제1항에 있어서, 유체가 생물학적 또는 비-생물학적 유체임을 특징으로 하는 방법.
- 제2항에 있어서, 생물학적 유체가 혈액, 혈청, 혈장, 뇨, 땀, 뇌척수액, 림프 또는 장분비물, 추출액 또는 누출액이고 비-생물학적 유체가 물 또는 수성추출물 또는 혼합물임을 특징으로 하는 방법.
- 제1항에 있어서, 효소가 트랜스퍼라아제, 히드롤라아제, 옥시도리덕타아제 또는 리아제임을 특징으로 하는 방법.
- 제4항에 있어서, 트랜스퍼라아제가 전이한 함유그룹이고 히드롤라아제가 글루코시다아제 또는 펩티드결합에 작용하는 것임을 특징으로 하는 방법.
- 제5항에 있어서, 히드롤라아제가 α- 또는 β-D-갈락토시다아제, 카복시펩티나아제 A, 콜라게나아제, 아밀라아제, 칼파인 Ⅰ, 칼파인 Ⅱ, 디펩티딜 아미노펩티다아제 Ⅰ(카텝신 C) 또는 포스포글리콜레이트 포스포타아제임을 특징으로 하는 방법.
- 제5항에 있어서, 틀내스퍼라아제가 피루베이트 키나아제, 헥소키나아제, 아데닐레이트 키나아제, 피리독살 키나아제 또는 아세테이트 키나아제이고 히드롤라아제는 α- 또는 β-D-갈락토시다아제 또는 카복시펩티다아제 A임을 특징으로 하는 방법.
- 제4항에 있어서, 옥시도리덕타아제가 공여체분자의 CH-OH 그룹에 또는 알데히드에 또는 공여체의 옥소그룹에 작용함을 특징으로 하는 방법.
- 제4항에 있어서, 옥시도리덕타아제가 글리세롤 디하이드로게나아제, 아세트알데히드 디하이드로게나아제 또는 티로시나아제(카테롤 옥시다이제)임을 특징으로 하는 방법.
- 제7항에 있어서, 효소 피루베이트 키나아제 또는 β-D-갈락토시다아제를 사용하여 나트륨이온을 측정함을 특징으로 하는 방법.
- 제1항에 있어서, 방해 이온이 결합제로 차폐됨을 특징으로 하는 방법.
- 제1항에 있어서, 시료를 희석하는 것이 불가능할 경우에 측정될 나트륨이온을 결합제에 의해 측정에 적합한 수준의 농도를 감소시키거나 측정할 나트륨이온에 대한 효소의 감도를 감소시킴을 특징으로 하는 방법.
- 제11항 또는 12항중의 어느 한 항에 있어서, 방해이온 또는 검체가 크립탄드, 코로난드, 포난드, 크라운 에테르, 스페란드, 헤미스페란드, 칼릭사렌 또는 이들의 조합물, 천연적으로 존재하는 이온공여체, 시클릭펩티드, 콤플렉손 또는 킬레이트제 및 이들의 유도체에 의해 결합됨을 특징으로 하는 방법.
- 제13항에 있어서, 나트륨이온을 측정하는 경우에 방해하는 칼륨이온이 KryptofixR222에 의해 결합됨을 특징으로 하는 방법.
- 제12항에 있어서, β-갈락토시다아제를 사용하여 나트륨이온을 측정하는 경우와 유리나트륨이온의 농도를 결합제로 감소시키거나 나트륨이온에 대한 효소의 감도를 리튬이온으로 감소시킴을 특징으로 하는 방법.
- 제12항 또는 제15항중에서 어느 한 항에 있어서, 결합제가 KryptofixR221임을 특징으로 하는 방법.
- 제12항에 있어서, 크립탄드, 코로난드, 포단드, 크라운 에테르, 스페란드, 헤미스페란드, 칼릭사렌 또는 이들의 조합물, 천연적으로 존재하는 이온공여체, 시클로펩티드, 콤플렉손 또는 킬레이트제 및 이들의 유도체로 구성되는 결합제가 존재하고, 이것이 지시이온과 함께 착체를 형성하고 이로부터 측정될 이온에 의해 지시이온이 대체되고 효소의 활성에 대한 대체된 지시이온의 영향을 분석하고, 이에 의해 검체농도가 간접적으로 측정됨을 특징으로 하는 방법.
- 제17항에 있어서, 효소가 피루베이트 키나아제이고, 지시이온이 칼륨이고, 결합제가 KryptofixR221이고 측정될 이온이 나트륨임을 특징으로 하는 방법.
- 제17항에 있어서, 측정될 이온이, 착물로 부터 pH 민감성 결합제로 지시이온을 대체하는 pH값의 변화를 유발함을 특징으로 하는 방법.
- 제1항에 있어서, 민감한 지시효소의 경쟁적 억제제인 이온이 분석중에 포함됨을 특징으로 하는 방법.
- 제20항에 있어서, 측정된 이온이 나트륨이고 경쟁적 억제제가 리튬이온임을 특징으로 하는 방법.

- 제31항에 있어서, 트리스-HCl, pH 8.7(37℃), 티티오트레이톨, 황산마그네슘, 염화리튬, EGTA(리튬염), 인간의 혈청알부민, β-갈락토시다아제, NPG, KryptofixR221, ADP(유리산), PEP(중화된 트리스염), NADH, GDH를 함유하는 분석으로, 반응속도를 측정하여 나트륨이온의 농도를 분석하고, 배양혼합물의 pH를 낮춘후 LDH,PK,MnCl2,LiCl 성분을 가하는 것을 특징으로 하는 방법.
