Rγ2がヘテロアリールであるとき、Rγ2は、好ましくはキノリニル、ピリジルまたは2-ベンゾフラニルである。
最も好ましくは、抗糖尿病性D-フェニルアラニン誘導体はナテグリニドまたはその薬学的に許容される塩である。
引用の特許文献に開示されている対応する立体異性体および結晶改変などの対応の多形も同様に含まれる。
ICR-CDIマウス(雄、5週令、体重:約20 g)を18時間絶食させた後、試験対象として使用する。本発明の組合せ物および活性成分単独を0.5% CMC-0.14M 塩化ナトリウム緩衝液(pH 7.4)に懸濁する。こうして得た溶液を一定の量で試験動物に経口投与する。予め定めた時間後に、対照群に対する血糖値の低下パーセントを決定する。
本試験は、本発明の組合せ製剤または医薬組成物の相乗作用を特に証明する。DPP-IV仲介状態、特に2型糖尿病に対する有益な作用を、この試験の結果を通して直接に、または当業者に既知の試験方法を変ることにより決定することができる。
Claims (16)
- 遊離または薬学的に許容される塩の形態のジペプチジルペプチダーゼ−IV(DPP-IV)阻害剤および少なくとも1つのさらなる抗糖尿病性化合物または該化合物の薬学的に許容される塩、および場合により少なくとも1つの薬学的に許容される担体を含む、同時に、別々に、または連続的に使用するための組合せ物。
- さらなる抗糖尿病性化合物が、タンパク質チロシンホスファターゼ(PTPases)阻害剤、非小分子模倣化合物およびグルタミン-フルクトース-6-リン酸アミドトランスフェラーゼ(GFAT)阻害剤のようなインスリン情報伝達モジュレーター、グルコース-6-ホスファターゼ(G6Pase)阻害剤、フルクトース-1,6-ビスホスファターゼ(F-1,6-BPase)阻害剤、グリコーゲンホスホリラーゼ(GP)阻害剤、グルカゴン受容体アンタゴニストおよびホスホエノールピルビン酸・カルボキシキナーゼ(PEPCK)阻害剤のような異常調節された肝グルコース産生に影響する化合物、ピルビン酸デヒドロゲナーゼ・キナーゼ(PDHK)阻害剤、インスリン増感剤、インスリン分泌促進剤、α-グルコシダーゼ阻害剤、胃排出阻害剤、インスリンおよびα2-アドレナリンアンタゴニストよりなる群から選択されるまたは該化合物の薬学的に許容される塩である、請求項1に記載の組合せ物。
- 組合せ製剤または医薬組成物である、請求項1または2に記載の組合せ物。
- DPP-IV仲介状態の予防、進行遅延または処置のために同時に、別々にまたは連続的に使用するための組合せ製剤である、請求項3に記載の組合せ物。
- DPP-IV阻害剤が、遊離形態または酸付加塩形態の式(I):
a) R1R1aN(CH2)m-(ここで、
R1は、場合により低級アルキル、低級アルコキシ、ハロゲン、トリフルオロメチル、シアノまたはニトロでモノ-または独立的にジ-置換されたピリジニルまたはピリミジニル部分;または場合により低級アルキル、低級アルコキシまたはハロゲンでモノ-または独立的にジ-置換されたフェニルであり;
R1aは、水素または(C1-8)アルキルであり;
mは、2または3である);
b) 場合により1-位を(C1-3)ヒドロキシアルキルでモノ置換された(C3-12)シクロアルキル;
c) R2(CH2)n-(ここで:
R2は、場合により低級アルキル、低級アルコキシ、ハロゲンまたは場合によりフェニル環がヒドロキシメチルでモノ置換されたフェニルチオで、モノ-または独立的にジ-または独立的にトリ-置換されたフェニル;(C1-8)アルキル;場合により(C1-8)アルキルでモノ-または多-置換された[3.1.1]二環性炭素環部分;場合により低級アルキル、低級アルコキシまたはハロゲンでモノ-または独立的にジ-置換されたピリジニルまたはナフチル部分;シクロヘキセン;またはアダマンチルであり;かつ
nは、1〜3であるか;または
R2は、場合により低級アルキル、低級アルコキシまたはハロゲンでモノ-または独立的にジ-置換されたフェノキシであり;かつ
nは、2または3である);
d) (R3)2CH(CH2)2-、(ここで、各R3は、独立的に、場合により低級アルキル、低級アルコキシまたはハロゲンでモノ-または独立的にジ-置換されたフェニルである);
e) R4(CH2)p-(ここで、R4は2-オキソピロリジニルまたは(C2-4)アルコキシであり;そして
pは、2〜4である);
f) 場合により1-位が(C1-3)ヒドロキシアルキルでモノ置換されたイソプロピル;
g) R5(ここで、R5はインダニル;場合によりベンジルで置換されたピロリジニルまたはピペリジニル部分;場合により(C1-8)アルキルでモノ-または多置換された[2.2.1]-または[3.1.1]-二環性炭素環部分;アダマンチル;または場合によりヒドロキシまたはヒドロキシメチルで、または場合により低級アルキル、低級アルコキシまたはハロゲンでモノ-または独立的にジ-置換されたフェニルで、モノ-または独立的に多-置換された(C1-8)アルキルである);
h) 置換されたアダマンチル

のN-(N'-置換グリシル)-2-ベンジルシアノピロリジンである、請求項1〜4のいずれかに記載の組合せ物。 - DPP-IV阻害剤が、(S)-1-[(3-ヒドロキシ-1-アダマンチル)アミノ]アセチル-2-シアノ-ピロリジンおよび(S)-1-{2-[5-シアノピリジン-2-イル)アミノ]エチル-アミノアセチル}-2-シアノ-ピロリジンよりなる群から選ばれる遊離形態または酸付加塩形態の式Iの化合物である、請求項5の組合せ物。
- インスリン増感剤が、抗糖尿病性チアゾリジンジオンおよび抗糖尿病性バナジウム含有化合物よりなる群から選ばれる、請求項2〜4のいずれかに記載の組合せ物。
- インスリン分泌促進剤が、スルホニル尿素誘導体、抗糖尿病性フェニル酢酸誘導体および抗糖尿病性D-フェニルアラニン誘導体よりなる群から選ばれる、請求項2に記載の組合せ物。
- 抗糖尿病性D-フェニルアラニン誘導体が、式X:
Rγ1は、水素、C1〜C5アルキル、C6〜C12アリール、C6〜C12アリールアルキル、
Rγ2は、C6〜C12アリール、ヘテロ6員環、ヘテロ5員環、シクロアルキルまたはシクロアルケニル(いずれの基も1以上の置換基を有し得る)よりなる群から選択され;そして
Rγ3は、水素およびC1〜C5アルキルより選択する(ただし、Rγ1およびRγ3がともに水素原子であるとき、Rγ2は置換または非置換のフェニルまたはナフチル以外である)。]
の化合物、その薬学的に許容される塩、または人体内または動物体内でそのものに変換され得る前駆物質である、請求項8に記載の組合せ物。 - DPP-IV阻害剤が、(S)-1-[(3-ヒドロキシ-1-アダマンチル)アミノ]アセチル-2-シアノ-ピロリジンおよび(S)-1-{2-[5-シアノピリジン-2-イル)アミノ]エチル-アミノアセチル}-2-シアノ-ピロリジンから選択され、さらなる抗糖尿病性化合物が、ナテグリニド、レパグリニド、メトホルミン、ロシグリタゾン、ピオグリタゾン、トログリタゾン、グリソキセピド、グリブリド、グリベンクラミド、アセトヘキサミド、クロロ-プロパミド、グリボルヌリド、トルブタミド、トラザミド、グリピジド、カルブタミド、グリキドン、グリヘキサミド、フェンブタミド、トルシクラミド、グリメピリドおよびグリクラジドから選択されるか、または該化合物の薬学的に許容される塩である、請求項1に記載の組合せ物。
- DPP-IV仲介状態を処置する方法であって、処置を必要とする温血動物に、併用で治療上有効な量の遊離または薬学的に許容される塩形態のDPP-IV阻害剤、および少なくとも1つのさらなる抗糖尿病性化合物(好ましくは、タンパク質チロシンホスファターゼ(PTPases)阻害剤、非小分子模倣化合物およびグルタミン-フルクトース-6-リン酸アミドトランスフェラーゼ(GFAT)阻害剤のようなインスリン情報伝達モジュレーター、グルコース-6-ホスファターゼ(G6Pase)阻害剤、フルクトース-1,6-ビスホスファターゼ(F-1,6-BPase)阻害剤、グリコーゲンホスホリラーゼ(GP)阻害剤、グルカゴン受容体アンタゴニストおよびホスホエノールピルビン酸・カルボキシキナーゼ(PEPCK)阻害剤のような異常調節された肝グルコース産生に影響する化合物、ピルビン酸デヒドロゲナーゼ・キナーゼ(PDHK)阻害剤、インスリン増感剤、インスリン分泌促進剤、α-グルコシダーゼ阻害剤、胃排出阻害剤、インスリンおよびα2-アドレナリンアンタゴニストよりなる群から選ばれる)または該化合物の薬学的に許容される塩を投与することを含む方法。
- 哺乳動物の体型を改善する方法であって、該哺乳動物に、遊離または薬学的に許容される塩形態のDPP-IV阻害剤、および少なくとも1つのさらなる抗糖尿病性化合物(好ましくは、タンパク質チロシンホスファターゼ(PTPases)阻害剤、非小分子模倣化合物およびグルタミン-フルクトース-6-リン酸アミドトランスフェラーゼ(GFAT)阻害剤のようなインスリン情報伝達モジュレーター、グルコース-6-ホスファターゼ(G6Pase)阻害剤、フルクトース-1,6-ビスホスファターゼ(F-1,6-BPase)阻害剤、グリコーゲンホスホリラーゼ(GP)阻害剤、グルカゴン受容体アンタゴニストおよびホスホエノールピルビン酸・カルボキシキナーゼ(PEPCK)阻害剤のような異常調節された肝グルコース産生に影響する化合物、ピルビン酸デヒドロゲナーゼ・キナーゼ(PDHK)阻害剤、インスリン増感剤、インスリン分泌促進剤、α-グルコシダーゼ阻害剤、胃排出阻害剤、インスリンおよびα2-アドレナリンアンタゴニストよりなる群から選ばれる)または該化合物の薬学的に許容される塩を、グルコース代謝に影響を与え、美容的に有益な減量に作用するのに有効な量で投与することを含む方法。
- DPP-IV仲介状態に対する併用で治療上有効な量の、請求項1〜10のいずれかに記載の組合せ物、および少なくとも1つの薬学的に許容される担体を含む医薬組成物。
- DPP-IV仲介状態の予防、進行遅延または処置のための薬剤の製造における、請求項1〜10のいずれかに記載の組合せ物の使用。
- 美容的に有益な減量を達成するための、哺乳動物の美容的処置における、請求項1〜10のいずれかに記載の組合せ物の使用。
- 活性物質として請求項1〜10のいずれかに記載の組合せ物を、DPP-IV仲介状態の予防、進行遅延または処置における、および哺乳動物の体型の改善の方法における同時の、別々のまたは連続的な使用のための指示書とともに含む市販用包装物。
