Abstract

본 발명은 세포를 약 38℃ 내지 약 39℃의 온도에서 배양함으로써 아데노바이러스-형질전환된 포유동물 세포중에서 작용성 재조합 단백질을 고수준으로 생성시키는 방법에 관한 것이다. 상기 방법은 어떤 세포계에서는 총 단백질 발현 수준이 증가되도록 하고, 다른 세포계에서는 작용성 단백질의 수준이 증가되도록 한다.

Claims (10)

  1. 아데노바이러스-혈질전환된 재조합 포유동물 숙주 세포를 약 38℃ 내지 39℃의 온도에서 배양함을 포함하는, 상기 아데노바이러스-형질전환된 재조합 포유동물 숙주 세포에서 단백질 C의 생성을 증가시키기 위한 방법.
  2. 제1항에 있어서, 상기 재조합 포유동물 숙주 세포가 293 세포인 방법.
  3. 제1항에 있어서, 상기 배양 온도가 약 38℃인 방법.
  4. 제1항에 있어서, 상기 배양 온도가 약 39℃인 방법.
  5. 제2항에 있어서, 상기 배양 온도가 약 38℃인 방법.
  6. 제2항에 있어서, 상기 배양 온도가 약 38.5℃인 방법.
  7. 제2항에 있어서, 상기 배양 온도가 약 39℃인 방법.
  8. 재조합 AV12숙주 세포중의 재조합적으로 생성된 단백질 C분자를 약 38℃ 내지 약 39℃의 온도에서 배양함을 포함하는, 상기 재조합적으로 생성된 단백질 C분자의-카복실화 수준을 증가시키는 방법.
  9. 제8항에 있어서, 상기 재조합 AV12 숙주 세포를 약 38℃에서 배양하는 방법.
  10. 제8항에 있어서, 상기 재조합 AV12 숙주 세포를 약 39℃에서 배양하는 방법.
