Description
도 2는, E.coli ccrNXA 오페론을 발현시키는 이. 콜라이(E. coli) L-글루탐산 생산균의 L-글루탐산 생산 배양 결과를 도시한 도면이다.
도 3은, 이. 콜라이(E. coli) 고유의 크실로스 자화 경로 결손주를 숙주로 한 ccrNXA 오페론 발현주에 의한 L-글루탐산 생산 배양의 결과를 도시한 도면이다.
도 4는, 중카피(medium copy)형 벡터를 사용한 ccrNXA 오페론 발현주에 의한 L-글루탐산 생산 배양의 결과를 도시한 도면이다.
도 5는, 각종 미생물 유래의 xylD 호모로그 유전자 발현주에 의한 L-글루탐산 생산 배양의 결과를 도시한 도면이다. Atu, Hse, Amis, Aor은 각각 Agrobacterium tumefaciens, Herbaspirillum seropedicae, Actinoplanes missouriensis, Aspergillus oryzae를 나타낸다.

Claims (16)
- 목적 물질을 생산하는 능력을 갖는 세균을, 탄소원으로서 크실로스를 함유하는 배지에서 배양하여, 목적 물질을 당해 배지 중에 생성 축적시키고, 당해 배지로부터 목적 물질을 채취하는, 목적 물질의 제조 방법으로서,
상기 목적 물질은 2-케토글루타르산 또는 이의 유도체이고,
상기 세균은, 크실로스로부터 크실론산을 생성하는 능력을 갖고, 또한, 크실로네이트데히드라타제 활성, 2-케토-3-데옥시크실로네이트데히드라타제 활성, 및 2-케토글루타르산 세미알데히드데히드로게나제 활성이 부여 또는 증강된 것을 특징으로 하는 방법. - 제1항에 있어서, 상기 세균은, 크실로네이트데히드라타제, 2-케토-3-데옥시크실로네이트데히드라타제, 및 2-케토글루타르산 세미알데히드데히드로게나제를 각각 코드화하는 유전자가 발현 가능한 형태로 도입됨으로써, 이들 효소 활성이 부여 또는 증강된 것을 특징으로 하는, 방법.
- 제1항 또는 제2항에 있어서, 크실로네이트데히드라타제, 2-케토-3-데옥시크실로네이트데히드라타제, 및 2-케토글루타르산 세미알데히드데히드로게나제를 코드화하는 유전자가 카울로박터(Caulobacter)속, 에스케리키아(Escherichia)속, 아그로박테리움(Agrobacterium)속, 헤르바스피릴룸(Herbaspirillum)속, 악티노플라네스(Actinoplanes)속, 쿠프리아비두스(Cupriavidus)속, 슈도모나스(Pseudomonas)속, 조벨리아(Zobellia), 써모바실루스(Thermobacillus)속, 아스로박터(Arthrobacter)속, 아조스피릴리움(Azospirillum)속, 할로모나스(Halomonas)속, 바실루스(Bacillus)속, 또는 아스페르길루스(Aspergillus)속에 속하는 미생물 유래인, 방법.
- 제1항 내지 제3항 중 어느 한 항에 있어서, 상기 세균은, 하기 중 어느 하나에 의해 크실로스로부터 크실론산을 생성할 수 있는 것을 특징으로 하는, 방법:
(A) 크실로스데히드로게나제 활성, 또는 크실로스데히드로게나제 및 크실로노락토나제 활성을 부여 또는 증강시킴, 또는,
(B) 크실로스로부터 크실론산을 생성하는 반응을 촉매할 수 있는 글루코스데히드로게나제 활성을 가짐. - 제4항에 있어서, 상기 글루코스데히드로게나제는 피롤로퀴놀린퀴논을 조효소로 하고, 또한, 상기 세균은, 피롤로퀴놀린퀴논 생산능을 갖거나, 또는 피롤로퀴놀린퀴논을 함유하는 배지에서 배양됨으로써 글루코스데히드로게나제 활성을 갖는, 방법.
- 제4항에 있어서, 상기 세균은, 크실로스데히드로게나제, 또는 크실로스데히드로게나제 및 크실로노락토나제를 각각 코드화하는 유전자가 발현 가능한 형태로 도입됨으로써, 크실로스로부터 크실론산을 생성할 수 있는, 방법.
- 제1항 내지 제6항 중 어느 한 항에 있어서, 상기 세균은, 2-케토글루타르산데히드로게나제의 활성이 저하되도록 추가로 개변(改變)된 것을 특징으로 하는, 방법.
- 제1항 내지 제7항 중 어느 한 항에 있어서, 상기 세균은, 석신산데히드로게나제의 활성이 저하되도록 추가로 개변된 것을 특징으로 하는, 방법.
- 제1항 내지 제8항 중 어느 한 항에 있어서, 상기 세균이 장내 세균 또는 코리네형(coryneform) 세균인 것을 특징으로 하는, 방법.
- 제9항에 있어서, 상기 세균이 판토에아(Pantoea)속 세균인 것을 특징으로 하는, 방법.
- 제10항에 있어서, 상기 세균이 판토에아·아나나티스(Pantoea ananatis)인 것을 특징으로 하는, 방법.
- 제9항에 있어서, 상기 세균이 에스케리키아(Escherichia)속 세균인 것을 특징으로 하는, 방법.
- 제12항에 있어서, 상기 세균이 에스케리키아·콜라이(Escherichia coli)인 것을 특징으로 하는, 방법.
- 제9항에 있어서, 상기 세균이 코리네박테리움(Corynebacterium)속 세균인 것을 특징으로 하는, 방법.
- 제14항에 있어서, 상기 세균이 코리네박테리움·글루타미쿰(Corynebacterium glutamicum)인 것을 특징으로 하는, 방법.
- 제1항 내지 제15항 중 어느 한 항에 있어서, 상기 2-케토글루타르산 유도체가, L-글루탐산, L-글루타민, L-아르기닌, L-시트룰린, L-오르니틴, L-프롤린, 푸트레신, 및 γ-아미노부티르산으로 이루어지는 그룹으로부터 선택되는 물질인, 방법.
