Claims (24)
- 코리네형 세균으로부터 유래되었으며, 서열번호 18에 기재된 아미노산 서열의 311번째 아미노산 잔기가 Thr 이외의 아미노산으로 치환된 아미노산 서열을 가지고, L-리신 및 L-트레오닌에 의한 협동(concerted) 피드백저해를 받지 않는 아스파르토키나아제를 암호화하는 DNA.

- 제 1항 및 제 13항 내지 제 15항의 어느 한 항의 DNA를 벡터에 삽입하여 수득되는 코리네형 세균으로 복제가능한 재조합 DNA.
- 제 1항 및 제 13항 내지 제 15항의 어느 한 항의 DNA를 염색체 상에서 가지는 코리네형 세균.
- 제 16항의 재조합 DNA를 포함하는 코리네형 세균에 속하는 형질전환체.
- 제 18항에 있어서,코리네박테리움 글루타미컴 Tf-5(FERM BP-6689)인 것을 특징으로 하는형질전환체.
- 제 17항의 코리네형 세균, 제 18항 및 제 19항의 형질전환체로부터 선택되는 미생물 또는 형질전환체를 배지에서 배양하고, 배양물 중에서 L-리신을 생성, 축적시키며, 전기 배양물로부터 L-리신을 수득하는 것을 특징으로 하는 L-리신의 제조방법.
- 제 1항, 제 13항 내지 제 15항의 어느 한 항의 DNA로 암호화되는 아스파르토키나아제.
- 제 21항에 있어서,서열번호 18에 기재된 아미노산 서열을 가지는 것을 특징으로 하는아스파르토키나아제.
- (1) 코리네형 세균에 서열번호 18에 기재된 아미노산 서열을 가지는 아스파르토키나아제를 암호화하는 DNA 또는 서열번호 18에 기재된 아미노산 서열의 311번째 아미노산 잔기가 Thr 이외의 아미노산으로 치환된 아미노산 서열을 가지고, L-리신 및 L-트레오닌에 의한 협동(concerted) 피드백저해를 받지 않는 아스파르토키나아제를 암호화하는 DNA를 도입하는 단계;(2) 상기 (1)의 DNA와 코리네형 세균의 염색체 DNA와의 사이에서 상동재조합을 일으키는 단계; 및,(3) 서열번호 18에 기재된 아미노산 서열을 가지는 아스파르토키나아제, 또는 서열번호 18에 기재된 아미노산 서열의 311번째 아미노산 잔기가 Thr 이외의 아미노산으로 치환된 아미노산 서열을 가지고, L-리신 및 L-트레오닌에 의한 협동 피드백저해를 받지 않는 아스파르토키나아제를 암호화하는 DNA를 그 염색체 상에서 가지는 코리네형 세균을 선택하는 단계를 포함하는, 서열번호 18에 기재된 아미노산 서열을 가지는 아스파르토키나아제, 또는 서열번호 18에 기재된 아미노산 서열의 311번째 아미노산 잔기가 Thr 이외의 아미노산으로 치환된 아미노산 서열을 가지고, L-리신 및 L-트레오닌에 의한 협동 피드백저해를 받지 않는 아스파르토키나아제를 암호화하는 DNA를 그 염색체 상에서 가지는 코리네형 세균의 제조방법.
- 제 23항의 방법에 의하여 제조된 코리네형 세균을 배지에 배양하고, 배양물 중에서 L-리신을 생성, 축적시키고, 전기 배양물로부터 L-리신을 수득하는 것을 특징으로 하는 L-리신의 제조방법.
