Claims (1)
- 【特許請求の範囲】 1.核酸送達システムであって、 (1)標的細胞上の部位に特異的に結合する標的部分と、 (2)核酸セグメントに結合する結合部分と、 を含む融合タンパク質、および 目的の核酸配列を含む該核酸セグメント を含む、核酸送達システム。 2.前記標的部分が抗体である、請求項1に記載の核酸送達システム。 3.前記抗体がウイルスエンベロープタンパク質、細胞レセプター、または活性 型レセプターの細胞外ドメインに対する抗体である、請求項2に記載の核酸送達 システム。 4.前記抗体が単鎖抗体、抗体のFab部分、または(Fab')2セグメントである、請 求項2に記載の核酸送達システム。 5.前記結合部分がタンパク質またはタンパク質の核酸結合ドメインであり、そ して該結合部分が前記標的部分のカルボキシ部分に融合されている、請求項1に 記載の核酸送達システム。 6.前記結合部分が、GCN4、Fos、Jun、TFIIS、FMRI、酵母タンパク質HX、ビギ リン、Mer1、細菌ポリヌクレオチドホスホリラーゼ、リボソームタンパク質S3、 および熱ショックタンパク質からなる群より選択されたタンパク質に存在する核 酸結合ドメインの群から選択される、タンパク質の核酸ドメインである、請求項 5に記載の核酸送達システム。 7.前記結合部分がタンパク質プロタミンである、請求項5に記載の核酸送達シ ステム。 8.前記目的の核酸配列が、抗体、優性ネガティブ変異体、アンチセンスRNA、 リボザイム、または細胞傷害性因子をコードする、請求項1に記載の核酸送達シ ステム。 9.前記核酸セグメントが、前記目的の核酸配列中に、隣接する5’および3’ 末端反復配列(LTR)領域または逆位末端反復(ITR)領域、所望の遺伝子に作動可能 に連結したプロモーターを含む、請求項1に記載の核酸送達システム。 10.融合タンパク質と核酸セグメントとを含む核酸送達システムであって、こ こで、該融合タンパク質の一部が細胞上の所望の部位に選択的に結合する抗体を 含み、そして該融合タンパク質の他の部分が該核酸セグメントに結合し得るプロ タミンタンパク質を含む、核酸送達システム。 11.前記核酸セグメントがサイトトキシン遺伝子または細胞傷害性タンパク質 をコードするそのフラグメントに対応するDNA配列である、請求項10に記載の 核酸送達システム。 12.前記核酸セグメントが少なくともシュードモナスエキソトキシンAのドメ インIIIをコードする、請求項11に記載の核酸送達システム。 13.標的細胞を形質転換する方法であって、該方法が、有効量の請求項1に記 載の核酸送達システムを標的細胞を含む培地に添加する工程、および該核酸送達 システムの核酸配列が該細胞を形質転換するまで待つ工程を含む、方法。 14.核酸送達システムを調製する方法であって、該方法が、細胞を、作動可能 にプロモーターに連結された請求項1に記載の融合タンパク質をコードするDNA セグメントを含むベクターで形質転換する工程、該細胞をインキュベートする工 程、そして発現された該融合タンパク質を収集する工程を含む、方法。 15.核酸送達システムを使用する方法であって、該方法が、有効量の請求項1 に記載の核酸送達システムを標的細胞を含有する血清に投与する工程、そして該 核酸送達システムが該標的細胞と接触するまで待つ工程を含む、方法。 16.核酸送達システムを使用する方法であって、該方法が、有効量の請求項1 0に記載の核酸送達システムを標的細胞を含有する血清に投与する工程、そして 該核酸送達システムが該標的細胞と接触するまで待つ工程を含む、方法。
