Description
本出願は、2017年3月20日に出願された米国仮出願第62/473,850号の利益を主張し、その内容は参照により本明細書に組み込まれる。
「真空蒸留槽」という用語は、真空下で蒸留を行うための装置を包含することを意図しており、蒸留される液体はその沸点を下げるために低圧で封入されている。好ましくは、真空蒸留槽は、分離媒体を封入するためのケーシングを含む。好ましくは、蒸留される液体は、生存可能な微生物バイオマスおよび少なくとも1つの生成物を含む発酵ブロスである。そのような発酵ブロスはバイオリアクタから供給されてもよい。バイオリアクタは、C1含有基質の発酵に使用することができる。
真空蒸留は、発酵ブロスに含有される微生物の生存率を確保しながら、発酵ブロスから生成物を効果的に回収することがわかった。真空蒸留槽に供給される発酵ブロスはバイオリアクタから供給される。好ましくは、バイオリアクタは、C1含有ガス状基質の発酵に使用される。発酵プロセスが連続的に操作するためには、ブロスに含有される微生物の少なくとも一部が生存可能なままでなければならない。これらの微生物はある特定の生成物の濃度に対してかなり特異的な耐性を有する。加えて、これらの微生物は温度に対してかなり特異的な耐性を有する。例えば、少なくとも1つの実施形態において、微生物は37℃の最適増殖温度を有する。本発明者らは、真空蒸留を利用することによって、発酵プロセスの連続操作が可能であるような方法で生存率の条件を制御することができることを見出した。

[発明1]
発酵ブロスから少なくとも1つの生成物を回収するための真空蒸留槽であって、前記発酵ブロスがバイオリアクタから搬送され、前記真空蒸留槽が、
a.生存可能な微生物バイオマスおよび少なくとも1つの生成物を含む前記発酵ブロスを受容するための入口、生成物濃縮流を移送するための出口、ならびに生成物枯渇流を移送するための出口を画定する外部ケーシングであって、前記生成物枯渇流が生存可能な微生物バイオマスを含み、前記生成物枯渇流が前記バイオリアクタに移送される、外部ケーシングと、
b.前記ケーシング内に位置する分離部分であって、上部トレイによって上方に、および下部トレイによって下方に結合されており、かつ複数の理論蒸留段を提供するための分離媒体を画定する、分離部分と、を備え、
前記生成物濃縮流を移送するための前記出口が前記発酵ブロスを受容するための前記入口に対して上にあり、前記発酵ブロスを受容するための前記入口が前記上部トレイに対して上にあり、前記生成物枯渇流を移送するための前記出口が前記下部トレイに対して上にある、真空蒸留槽。
[発明2]
前記バイオリアクタが発酵ブロスの体積を定義し、前記発酵ブロスがある供給速度で前記真空蒸留槽に搬送され、前記供給速度が1時間当たりの前記発酵ブロスの体積として定義され、前記供給速度が0.05~0.5である、発明1に記載の真空蒸留槽。
[発明3]
発酵ブロスが滞留時間を定義し、前記滞留時間が前記発酵ブロスが前記真空蒸留槽内にある期間として定義され、前記滞留時間が0.5~15分である、発明1に記載の真空蒸留槽。
[発明4]
前記生成物枯渇流を移送するための前記出口が、配管手段によって前記バイオリアクタに接続されて前記生成物枯渇流を前記バイオリアクタに移送し、前記バイオリアクタが産業プロセスからのC1含有ガスの発酵のための条件下で操作される、発明1に記載の真空蒸留槽。
[発明5]
前記分離媒体が一連の蒸留トレイによって画定されている、発明1に記載の真空蒸留槽。
[発明6]
前記分離媒体が3超の理論蒸留段を提供する、発明1に記載の真空蒸留槽。
[発明7]
前記生成物枯渇流中の前記生存可能な微生物バイオマスが、85パーセントを超える生存率を有する、発明1に記載の真空蒸留槽。
[発明8]
前記生成物枯渇流中の前記生存可能な微生物バイオマスが、前記発酵ブロス中の前記生存可能な微生物バイオマスと実質的に等しい、発明1に記載の真空蒸留槽。
[発明9]
前記生成物枯渇流が0.2重量%未満の生成物を含む、発明1に記載の真空蒸留槽。
[発明10]
前記生成物濃縮流が、エタノール、アセトン、イソプロパノール、またはそれらの混合物を含む、発明1に記載の真空蒸留槽。
[発明11]
前記真空蒸留槽が、前記真空蒸留槽の高さにわたる圧力降下を定義し、前記圧力降下が32mbar未満である、発明1に記載の真空蒸留槽。

前記真空蒸留槽が40mbar(a)~100mbar(a)の圧力で操作される、発明1に記載の真空蒸留槽。

前記生成物枯渇流は、前記生成物枯渇流が前記バイオリアクタに移送される前に、前記生成物枯渇流の温度を下げるために冷却手段に送られる、発明1に記載の真空蒸留槽。
[発明15]
前記生成物枯渇流の前記温度が、35℃~40℃である、発明14に記載の真空蒸留槽。
[発明16]
前記発酵ブロスを受容するための前記入口が配管手段によって脱気槽に接続され、前記脱気槽は、前記発酵ブロスが前記真空蒸留槽に搬送される前に、前記発酵ブロスからガスの少なくとも一部を除去する条件下で操作される、発明1に記載の真空蒸留槽。
[発明17]
前記脱気槽が0.0バール(g)~0.5バール(g)の圧力で操作される、発明16に記載の真空蒸留槽。
[発明18]
前記外部ケーシングが、リボイラーからの蒸気流を受容するための入口と、前記リボイラーに液体流を移送するための出口とをさらに画定し、前記蒸気流を受容するための前記入口が前記下部トレイに対して下方にあり、前記液体流を移送するための前記出口が前記蒸気流を受容するための前記入口に対して下部に位置する、発明1に記載の真空蒸留槽。
[発明19]
前記液体流が実質的に水と最小量の微生物バイオマスとからなる、発明18に記載の真空蒸留槽。
[発明20]
複数のバイオリアクタからの発酵ブロスを混合せずに前記真空蒸留槽に送ることができる様式で、前記真空蒸留槽が複数の区画に分離されている、発明1に記載の真空蒸留槽。
[発明21]
前記産業プロセスが、炭水化物発酵、ガス発酵、セメント製造、パルプ製紙、製鋼、石油精製および関連プロセス、石油化学製造、コークス製造、嫌気性および好気性消化、合成ガス、天然ガス抽出、石油抽出、アルミニウム、銅、および/または合金鉄の製造および/または精製のための冶金プロセス、地質学的貯留層、ならびに触媒プロセスを含む群から選択される、発明4に記載の真空蒸留槽。
[発明22]
真空蒸留槽を用いて発酵ブロスから少なくとも1つの生成物を除去するための方法であって、
a.生存可能な微生物バイオマスおよび少なくとも1つの生成物を含む発酵ブロスをバイオリアクタから真空蒸留槽に送ることと、
b.前記発酵ブロスを部分的に蒸発させて生成物濃縮流および生成物枯渇流を生成することであって、前記生成物枯渇流が生存可能な微生物バイオマスを含む、生成することと、
c.前記生成物枯渇流を前記バイオリアクタに戻すことと、を含む、方法。
[発明23]
前記発酵ブロスを前記真空蒸留槽に送る前に、脱気槽を使用して前記発酵ブロスを脱気して、脱気された発酵ブロスおよび発生したガス流を生成することをさらに含み、前記脱気した発酵ブロスを部分的に蒸発させる、発明22に記載の方法。
[発明24]
前記発生したガス流が少なくとも1つの生成物を回収するために水で洗浄される、発明23に記載の方法。
[発明25]
前記真空蒸留槽が、ケーシング内に位置する分離部分を備え、前記分離部分が上部トレイによって上方に、および下部トレイによって下方に結合されており、前記分離部分が複数の理論蒸留段を提供するための分離媒体を画定する、発明22に記載の方法。

前記バイオリアクタが産業プロセスからのC1含有ガスの発酵のための条件下で操作される、発明22に記載の方法。
Claims (16)
- 発酵ブロスから少なくとも1つの生成物を回収するための真空蒸留槽であって、前記発酵ブロスがバイオリアクタから搬送され、前記真空蒸留槽が、
a.生存可能な微生物バイオマスおよび少なくとも1つの生成物を含む前記発酵ブロスを受容するための入口、生成物濃縮流を移送するための出口、ならびに生成物枯渇流を移送するための出口を画定する外部ケーシングであって、前記生成物枯渇流が生存可能な微生物バイオマスを含み、前記生成物枯渇流が前記バイオリアクタに移送される、外部ケーシングと、
b.前記ケーシング内に位置する分離部分であって、上部トレイによって上方に、および下部トレイによって下方に結合されており、かつ複数の理論蒸留段を提供するための分離媒体を画定する、分離部分と、を備え、
前記生成物濃縮流を移送するための前記出口が前記発酵ブロスを受容するための前記入口に対して上にあり、前記発酵ブロスを受容するための前記入口が前記上部トレイに対して上にあり、前記生成物枯渇流を移送するための前記出口が前記下部トレイに対して上にあり、前記下部トレイと前記生成物枯渇流を移送するための前記出口とが、前記生成物枯渇流中の前記生存可能な微生物バイオマスが85パーセントを超える生存率を有するように配置される、前記真空蒸留槽。 - 前記発酵ブロスは、前記バイオリアクタに含有される前記発酵ブロスの体積に対して1時間当たり0.05~0.5の供給速度で、前記バイオリアクタから前記真空蒸留槽へ搬送される、請求項1に記載の真空蒸留槽。
- 前記真空蒸留槽内での前記発酵ブロスの滞留時間が0.5~15分である、請求項1に記載の真空蒸留槽。
- 前記生成物枯渇流を移送するための前記出口が、配管手段によって前記バイオリアクタに接続されて前記生成物枯渇流を前記バイオリアクタに移送し、前記バイオリアクタが産業プロセスからのC1含有ガスの発酵のための条件下で操作される、請求項1に記載の真空蒸留槽。
- 前記分離媒体が一連の蒸留トレイによって画定されている、請求項1に記載の真空蒸留槽。
- 前記分離媒体が3超の理論蒸留段を提供する、請求項1に記載の真空蒸留槽。
- 前記生成物枯渇流が0.2重量%未満の生成物を含む、請求項1に記載の真空蒸留槽。
- 前記生成物濃縮流が、エタノール、アセトン、イソプロパノール、またはそれらの混合物を含む、請求項1に記載の真空蒸留槽。
- 前記真空蒸留槽における圧力降下が32mbar未満である、請求項1に記載の真空蒸留槽。
- 前記真空蒸留槽が40mbar(a)~100mbar(a)の圧力で操作される、請求項1に記載の真空蒸留槽。
- 前記真空蒸留槽が35℃~50℃の温度で操作される、請求項1に記載の真空蒸留槽。
- 前記生成物枯渇流は、前記生成物枯渇流が前記バイオリアクタに移送される前に、前記生成物枯渇流の温度を下げるために冷却手段に送られる、請求項1に記載の真空蒸留槽。
- 前記発酵ブロスを受容するための前記入口が配管手段によって脱気槽に接続され、前記脱気槽は、前記発酵ブロスが前記真空蒸留槽に搬送される前に、前記発酵ブロスからガスの少なくとも一部を除去する条件下で操作される、請求項1に記載の真空蒸留槽。
- 前記脱気槽が0.0バール(g)~0.5バール(g)の圧力で操作される、請求項13に記載の真空蒸留槽。
- 前記外部ケーシングが、リボイラーからの蒸気流を受容するための入口と、前記リボイラーに液体流を移送するための出口とをさらに画定し、前記蒸気流を受容するための前記入口が前記下部トレイに対して下方にあり、前記液体流を移送するための前記出口が前記蒸気流を受容するための前記入口に対して下部に位置する、請求項1に記載の真空蒸留槽。
- 複数のバイオリアクタからの発酵ブロスを混合せずに前記真空蒸留槽に送ることができる様式で、前記真空蒸留槽が複数の区画に分離されている、請求項1に記載の真空蒸留槽。
