Claims (1)
- 【特許請求の範囲】 1.アセチルCoAプレグネノロンアセチルトランスフェラーゼ(APAT)活性 が、この活性をコードする遺伝子を変化させることにより除去され、3β−ヒド ロキシステロイドの安定化を生じた改変された酵母株。 2.変化された遺伝子がS.cerevisiaeのATF2遺伝子であるか又は後者の同族 体である、請求項1に記載の改変された酵母株。 3.変化された遺伝子がS.cerevisiaeのATF2遺伝子である、請求項2に記載 の改変された酵母株。 4.ATF2遺伝子が、少なくとも1つのヌクレオチドを有するDNA配列を挿 入することにより変化された、請求項3に記載の改変された酵母株。 5.ATF2遺伝子が、URA3選択遺伝子又は発現ブロックTEF1prom/P GKtermを挿入することにより変化された、請求項4に記載の改変された酵母株 。 6.ATF2遺伝子が、URA3選択遺伝子を挿入することにより変化された、 請求項5に記載の改変された酵母株。 7.請求項6に従って改変された、TGY156及びTGY158と呼ばれるS. cerevisiaeの株。 8.ATF2遺伝子が、発現ブロックTEF1prom/PGKtermを挿入すること により変化された、請求項5に記載の改変された酵母株。 9.請求項8に従って改変された、TGY186と呼ばれるS.cerevisiaeの株。 10.トランスフォームされた酵母株であって、アセチルCoAプレグネノロン アセチルトランスフェラーゼ(APAT)活性が、この活性をコードする遺伝子を 変化させ且つ下記より選択するコレステロールからのヒドロコーチゾンの生合成 の経路の酵素の少なくとも1つを発現させることにより除去されている当該トラ ンスフォームされた酵母株: − コレステロールの側鎖開裂酵素(P450SCC)、 − 3β−ヒドロキシ−デルタ5−ステロイドデヒドロゲナーゼ/デルタ5−デル タ4−ステロイドイソメラーゼ(3β−HSD)及び − 17α−ステロイドヒドロキシラーセ(P45017α)。 11.変化された遺伝子が、S.cerevisiaeのATF2遺伝子又は後者の同族体で ある、請求項10に記載のトランスフォームされた酵母株。 12.変化された遺伝子が、S.cerevisiaeのATF2遺伝子である、請求項11 に記載のトランスフォームされた酵母株。 13.ATF2遺伝子が、少なくとも1つのヌクレオチドを有するDNA配列を 挿入することにより変化された、請求項12に記載のトランスフォームされた酵 母株。 14.ATF2遺伝子が、URA3選択遺伝子を挿入することにより変化された 、請求項12に記載のトランスフォームされた酵母株。 15.請求項14に記載の3β−HSDを発現するトランスフォームされた酵母 株。 16.請求項15に従ってトランスフォームされたTGY158/pTG108 62と呼ばれるS.cerevisiaeの株。 17.ATF2遺伝子が、発現ブロックTEF1prom/PGKtermを挿入するこ とにより変化された、請求項12に記載のトランスフォームされた酵母株。 18.請求項17に記載のP45017αを発現するトランスフォームされた酵母 株。 19.請求項18に従ってトランスフォームされたTGY186/pTG104 35と呼ばれるS.cerevisiaeの株。 20.請求項17に記載され、3β−HSD及びP45017αを発現するトラン スフォームされた酵母株。 21.請求項20に従ってトランスフォームされたTGY186/pTG104 17と呼ばれるS.cerevisiaeの株。 22.内因性ステロール、外因性ステロール又は外因性ステロイドから選択する 基質のイン・ビボ酸化の方法であって、該方法では、請求項10〜21の何れか 1つに記載のトランスフォームされた酵母株を用い、該酵母株を、この株が内因 性ステロールを生成する場合には単独で培養し、或いは外因性ステロール又はス テロイドと共にインキュベートし、必要であれば得られた酸化された化合物を単 離する、上記の方法。 23.基質が3β−ヒドロキシステロイドであり、請求項15に記載のトランス フォームされた酵母株を用い、必要であれば得られた3−オキソ−デルタ4−ス テロイドを単離する、請求項22に記載のイン・ビボ酸化の方法。 24.3β−ヒドロキシステロイドをプレグネノロン又は17α−ヒドロキシプ レグネノロンから選択する、請求項23に記載のイン・ビボ酸化の方法。 25.基質がステロイドであり、請求項18に記載のトランスフォームされた酵 母株を用い、必要であれば得られた17α−ヒドロキシステロイドを単離する、 請求項22に記載のイン・ビボ酸化の方法。 26.ステロイド基質がプレグネノロン又はプロゲステロンである、請求項25 に記載のイン・ビボ酸化の方法。 27.基質が3β−ヒドロキシステロイドであり、請求項20に記載のトランス フォームされた酵母株を用い、得られた17α−ヒドロキシ3−オキソ−デルタ4 −ステロイドを必要であれば単離する、請求項22に記載のイン・ビボ酸化の 方法。 28.ステロイド基質がプレグネノロンである、請求項27に記載のイン・ビボ 酸化の方法。
