Abstract
本发明涉及一种微生物菌株。本发明的枯草芽孢杆菌BS-3菌株(Bacillus subtilis BS-3)分别经中科院微生物研究所及中国药品生物制品检定所鉴定为枯草芽孢杆菌(Bacillus subtilis)并在中国微生物菌种保藏管理委员会普通微生物的中心保藏，编号为CGMC.NO.0321。该菌株可作为治疗消化不良、急、慢性肠炎及菌群失调腹泻药剂的应用。该菌株体外对肠出血性大肠杆菌(EHE C 0157)有明显的拮抗作用。可制成活菌干粉、胶囊制剂、治疗人、动物的肠炎，腹泻疾病。
Description
本发明涉及一种微生物菌株。
我国50年代开始生产治疗肠道消化不良及肠炎的肠球菌制剂乳酶生和酵母片等。到八十年代由于抗生素大量广泛应用，导致耐药性菌株的大量出现及菌群失调性腹泻增多，此时临床医生更关注生态防治。多年来应用人体的非芽孢菌如乳酸、双歧菌、肠球菌等做成的制剂虽有一定疗效，但由于该类制剂中的细菌成活率低、保存期短、疗效也不令人满意，一直困绕着生物药品业的发展。这样寻找安全、有效、保存期长的微生物菌株及其微生物药品，已成为各科学家探求的方向。
本发明的目的是提供一种枯草芽孢杆菌BS-3菌株及其应用，用该菌株制成的药剂可治疗因各种因素引起的消化不良、急、慢性肠炎及菌群失调性腹泻。尤其是对出血型大肠杆菌感染性肠炎治疗更为有效。
本发明的枯草芽孢杆菌BS-3菌株(Bacillus Subtilis BS-3)的菌体形态，用镜检观察：该菌株呈杆状，直、单个排列，很少成链状。革兰氏染色阳性(G+)大小为0.6～0.7×1.5-2.0um，侧生鞭毛，中生芽孢呈椭圆形，孢囊不膨大，孢内无PHB颗粒。该菌株分别经中科院微生物研究所及中国药品生物制品检定所鉴定为枯草芽孢杆菌(Bacillus subtilis)。并于97年9月3日在中国微生物菌种保藏管理委员会普通微生物的中心保藏，编号为CGMCC.NO.0321。
本发明的枯草芽孢杆菌BS-3菌株(Bacillus Subtilis BS-3)在普通营养肉汤及普通肉汤琼脂平板上均可以培养，在18℃-45℃培养温度下均能生长，最适的生长温度为30-37℃，培养条件：PH值为5-8，最适的生长PH值为6.5-7.4。该菌株在普通营养肉汤及营养肉汤琼脂平板上培养，在培养过程中可产生较强的淀粉酶、蛋白酶、杆菌肽类抗菌物质，且具有抑制和杀灭血型大肠杆菌0157的能力。
本发明的枯草芽孢杆菌BS-3菌株(Bacillus Subtilis BS-3)是采用下述方法分离培养的：
1、菌株的来源：申请人从市场上购买了两袋南效乳品厂生产的鲜牛奶，置于4-10℃冰箱中，次日实验时发现其中一袋牛奶发生变质，即牛奶发生凝固且有乳清液析出，经对该变质牛奶进行初步细菌学直接涂片染色镜检，发现有一种革兰氏阳性杆菌。
2，分离培养：将上述变质牛奶接种于普通肉汤或普通琼脂培养基，采用常规的方法培养出本发明的枯草芽孢杆菌BS-3菌株(Bacillus SubtilisBS-3)。
本发明的枯草芽孢杆菌BS-3菌株(Bacillus Subtilis BS-3)可作为制备治疗消化不良、急、慢性肠炎及菌群失调腹泻药剂的应用。也可作为医药保健品、兽药和饲料添加剂。
本发明的枯草芽孢杆菌BS-3菌株(Bacillus Subtilis BS-3)可制成活菌胶囊制剂，可治疗人体中因各种因素引起的消化不良、急、慢性肠炎及菌群失调性腹泻。该胶囊制剂中各组份的重量比为：
枯草芽孢杆菌BS-3菌株的原菌粉为1.5-2.5份(重量份数)
赋形剂为96-100份(重量份数)，
其中枯草芽孢杆菌BS-3菌株的原菌粉中的活菌数为≥200亿个活菌/克。
所述的赋形剂为乳糖和可溶性淀粉，其中乳糖与可溶性淀粉的重量比为：
乳糖 9-11份(重量份数)
可溶性淀粉 87-89份(重量份数)
还有，所述原菌粉是由湿菌体和无水碳酸钙按1∶3重量比混匀干燥制成的。
本发明的枯草芽孢杆菌BS-3菌株(Bacillus Subtilis BS-3)可制成干粉制剂，治疗人或动物的消化不良、急、慢性肠炎，腹泻等病症。该粉制剂中各组份的重量比为：
枯草芽孢杆菌BS-3菌株的原菌粉为1.5-2.5份(重量份数)
赋形剂为96-100份(重量份数)
其中枯草芽孢杆菌BS-3菌株的原菌粉中的活菌数为≥200亿个活菌/克。
所述的赋形剂为乳糖和可溶性淀粉，其中乳糖与可溶性淀粉的重量比为：

本发明的枯草芽孢杆菌BS-3菌株(Bacillus Subtilis BS-3)经实验体外对肠出血性大肠杆菌(EHEC 0157)有明显的拮抗作用。24-48小时即显示抑杀作用。48小时以后随着培养时间的延长到96小时抑杀作用显著增强。其实验过程如下：
1、所用材料
(1)大肠杆菌菌种：肠出血性大肠杆菌0157(EHEC 0157)，菌号：88-2364。由中国预防医学科学院流行病学微生物学研究所提供。
(2)本发明的枯草芽孢杆菌活菌胶囊：规格0.25克/粒(枯草芽孢杆菌活菌≥1.0×108个/克)
(3)培养基：
a、普通肉汤培养基
b、大肠杆菌选择性培养基(EMB)干燥培养基：每瓶250克。由中国药品生物制品检定所提供。
c、7％NaCl琼脂培养基
2方法：
(1)预备试验测定出EHEC 0157细菌的对数生长曲线。
(2)EHEC 0157菌液的制备：
接种EHEC 0157菌于30毫升的肉汤培养基的三角烧瓶中，37℃水浴恒温振荡，培养15小时(此时为该菌的对数生长期)。用标准细菌浊度管比浊测定培养液中每毫升相当EHEC 0157菌数。然后用肉汤将换算出原培养液中EHEC0157菌数稀释调整为每毫升约含1×108个菌。
(3)枯草芽孢杆菌活菌胶囊悬液的制备：
在无菌操作间称取胶囊菌粉2.0克，加肉汤18毫升。混匀后(每毫升约含枯草芽孢杆菌1×108个菌)。
(4)实验操作步骤：
a、取两个灭菌三角烧瓶，分别为实验瓶及对照瓶。实验瓶中加入EHEC 0157菌液(1×108个菌/毫升)9.0毫升。再加入枯草芽孢杆菌胶囊悬液(摇匀)9.0毫升。混匀后取1.0ml，10倍系列稀释至10-9，测定枯草芽孢杆菌数，作为0小时枯草芽孢杆菌活菌基数，37℃水浴振荡培养。
对照瓶中加入EHEC 0157菌(1×108个菌/毫升)9.0毫升，再加肉汤9.0毫升。充分混匀后取1.0毫升加肉汤9.0毫升，按10倍系列稀释至10-9。每个稀释度各取0.1毫升，另入EMB琼脂平皿上(双份)经“L”棒涂抹后，置37℃温箱内培养24小时，平皿上计数为大肠菌落数，以此作为O小时三角瓶内EHEC 0157基础数。
b、菌数变化观察
分别以下不同培养时间(0、24、48、72、96小时)测定实验组与EHEC0157对照组中EHEC 0157的菌数。(CFU/ml)

从表1中结果可以看出试验组0小时EHECO157菌数对数值为8.3979，作用24小时后即下降为7.1760，对照组由0小时的8.3979，增殖为9.3979。作用至96小时，试验组EHEC 0157菌数降至4.7993，对照组仅下降至8.5682，统计分析有显著差异，说明枯草芽孢杆菌活菌制剂在体外与EHEC0157作用24小时即有拮抗作用，48小时后，这种拮抗作用进一步加强。
本发明的枯草芽孢杆菌BS-3菌株(Bacillus Subtilis BS-3)所制成的药剂，治疗人工实验感染的小型猪，治愈率在80％以上，有效率100％，自然发病仔猪治疗有效率达90％。其实验过程如下：
1、所用材料：
(1)实验感染小型猪：经产毒性大肠杆菌(0149、K88)攻毒的20头小型猪中，48小时内发生腹泻的共有18头，患猪腹泻的主要特征：粪便稀，不成形，少数患猪粪便呈水样，并伴有气泡，经过对18头患猪的粪便采样大肠杆菌培养，18头患猪均分离出大肠杆菌，并与大肠杆菌(0149)O抗原分型血清及K因子血清呈阳性凝集反应，证明患猪为大肠杆菌(0149K88)菌株感染。
(2)枯草芽孢杆菌活菌胶囊：规格0.25克/粒(含活菌2.5亿)。
(3)化药泻痢停：粉剂，50克/袋，江苏泰兴市动物药品厂生产。
2、治疗方法与剂量
方法：采用菌粉或药粉经口腔直接灌饲法，对照组只感染不给药同步观察。
剂量：枯草芽孢杆菌胶囊组：每头每次0.5克菌粉(约5亿活菌)，每日2次，连续灌饲3天。
化药泻痢停组：每头每次1.0克药粉，每日2次，连续灌饲3天。
疗效指标与判定标准：
(1)治愈：用药3天内粪便成形、吃食正常、精神活泼7天无复发。
(2)有效：用药3天内症状改善或减轻，粪便由稀变稠。
(3)无效：用药3天后症状未改善或加重或死亡。
观察期：实验观察治疗期为7天。
3、实验结果
(1)枯草芽孢杆菌活菌胶囊治疗实验感染大肠菌小型猪腹泻结果
在人工实验感染大肠杆菌(0149K88)的18头患猪，经两种不同的药物治疗，结果发现：枯草芽孢杆菌活菌胶囊组治疗6头，治愈5头，有效1头，有效率为100％，化药泻痢停组治疗6头，治愈4头，有效1头，有效率为83.3％；未经治疗的对照组自然好转1头，其余5头病猪中有1头死亡，另1头腹泻症状加重，结果见表2。

从表3可以看出枯草芽孢杆菌活菌量为10亿和5亿时治愈率为100％，活菌量为2.5亿时，治愈率为80％，总有效率为100％。
(3)大肠杆菌(0149 K88ac)实验感染后，经枯草芽孢杆菌胶囊治疗7天小型猪肠道几种主要菌群的改变。

表4结果表明患猪经枯草芽孢杆菌活菌胶囊治愈7天检测粪便中大肠杆菌数为8.02，肠球菌为7.92，枯草芽孢杆菌3.15，正常健康小型猪大肠杆菌为7.19。
本发明的枯草芽孢杆菌BS-3菌株(Bacillus subtilis BS-3)所制成的药剂对初生仔猪腹泻具有良好的预防作用，其保护率高达90％，即预防有效。其实验过程如下：
材料与方法
1、所用材料：
(1)怀孕待产小型猪母猪：2头，中国农业大学小型猪繁育场提供。
(2)枯草芽孢杆菌活菌胶囊：规格0.25克/粒(含活菌2.5亿)。
2、方法
将待产母猪分成实验组及对照组，在实验母猪产仔猪出生后3-5小时内立即灌饲枯草芽孢杆菌胶囊菌粉，每日2次，每次1粒(0.25克)，连续3天，对照组不给药同步观察。
观察指标：仔猪腹泻发生例数
观察期：给药3天观察至第7天。
3、实验结果
应用枯草芽孢杆菌活菌胶囊预防初生仔猪下痢的结果发现：实验组有1例发病，发病率为10％，死亡数为0，保护率为90％，非预防对照组有6例发病，发病率为60％，其中死亡3例，死亡率为30％，结果见表5。

本发明的枯草芽孢杆菌BS-3菌株(Bacillus subtilis BS-3)属于一种需氧芽孢菌，对人体及动物有益无毒付作用。该枯草芽孢杆菌BS-3菌株所制成的制剂经中国药品生物制品检定所检定：未发现由该制剂给动物造成的异常反应和病理改变。其检定报告如下：
检品名称：枯草芽孢杆菌活菌制剂。
规格：0.25克/粒
亚急性毒性试验及一般观察：SPF级小鼠按2.500g/kg体重剂量，每日灌服枯草芽孢杆菌活菌连续灌胃10天。实验组小鼠未见有中毒症状及死亡。小鼠活动自如，毛色光泽，食欲正常，口眼无分泌物，无呕吐症状，粪便未见异常，体重增加。
血液检查：实验小鼠血液中红细孢、白细孢及血红蛋白测定结果与对照组小鼠无明显差异。
剖检和病理学检查：实验组及对照组小鼠经解剖肉眼观察，心、肝、脾、胃、肺、肾、肠未见异常，组织病理学切片镜下观察上述器官或组织也未见因该制剂引起的病变。
结论：未发现由该制剂给动物造成的异常反应和病理改变。
实施例
(一)枯草芽孢杆菌BS-3菌株(Bacillus subtilis BS-3)分离。
1、样本的来源：样本为前述变质袋牛奶。
2、分离培养方法：
(1)分离用的培养基：采用普通肉汤液体培养基，PH值为7.0-7.2；或是普通营养肉汤琼脂培养基，PH值为7.0-7.2，经15磅压力，灭菌20分钟制成。
(2)方法：
采用普通肉汤液体培养法：无菌吸取上述变质袋牛奶的0.1ml，接种于普通肉汤中管两支，在37℃条件下培养18-24小时，观察培养管中的变化。
采用普通营养肉汤琼脂培养基法：用白金耳无菌挑取上述变质袋牛奶一环，划线于普通肉汤琼脂平板两块分离，在37℃条件下培养18-24小时，观察平皿内细菌生长情况。
(3)分离结果
在普通肉汤培养管中，培养液呈微混浊，培养管液面形成完整的一层菌膜，即细菌培养呈阳性生长，涂片培养物革兰氏染色均为G+杆菌，少数菌体有芽孢，位于菌体中央，呈椭圆形。经检定为枯草芽孢杆菌BS-3菌株。
在营养琼脂平板上，两块平板均培养为阳性，菌落圆形不规则，乳脂色不透明，菌落直径为2-4mm，经革兰氏染色为G+杆菌，大小约为0.6-0.7×1.5-2.0孔um芽孢中生，经检定为枯草芽孢杆菌BS-3菌株。
(二)生产菌种活化与检定
将上述枯草芽孢杆菌BS-3菌株接种于10ml普通营养肉汤培养液中管内，在35℃条件下培养18-24小时，然后用白金环挑一环划线转种于营养琼脂平板，在35℃条件下培养18-24小时，挑典型菌落，进行形态学与特性检定，并传代培养。
(三)种子液的制备
将检定合格的纯培养物，无菌吸取0.5ml接种于10ml营养肉汤培养瓶内，35℃条件下培养18-24小时，进行纯菌及菌形特征检查。
(四)生产培养基
将牛肉膏5克、蛋白胨10克、葡萄糖10克、氯化钠5克加入蒸馏水中至1000毫升，将上述混合物加热溶解后，调PH值为7.0-7.2，在15磅压力下灭菌20分钟。
(五)接种与培养
前述种子液经检定合格后，按1-3％比例(体积比)接种在装有上述培养液的发酵罐中，在35℃条件下培养48-72小时，检查芽孢形成的比率。
(六)收集菌体
将培养到期的培养物，要求生孢率达90％以上，再经纯菌试验，确认无杂菌污染。将上述培养物以4000rpm/分转速离心20分钟，弃上清，收集湿菌体。
(七)制备原菌粉
将上述湿菌体与无水碳酸钙按1∶3比例(重量比)混合搅拌成湿菌粉，在70-80℃条件下干燥，并研磨成干粉即原菌粉。该原菌粉的活菌数控制为≥200亿个活菌/克。
(八)拌粉
将上述原菌粉与灭菌过筛(筛孔为80目)的乳糖、可溶性淀粉按下述重量比混匀：原菌粉2份，乳糖10份、可溶性淀粉88份，并充分搅匀，其中上述菌粉混合物的活菌数控制为≥1×108个活菌/克。并按0.25克/袋或50克/袋包装成袋，制成枯草芽孢杆菌BS-3菌株的活菌干粉制剂。
(九)罐装胶囊
上述菌粉混合物也可以制成活菌胶囊制剂，将菌粉混合物置于胶囊罐装机内罐装，压板、包装，制成枯草芽孢杆菌BS-3菌株的活菌胶囊制剂，其中每粒胶囊为0.25克/粒。
综上所述，本发明所提供的枯草芽孢杆菌BS-3菌株可制成活菌干粉制剂、活菌胶囊制剂或其它形式的制剂，可治疗人、动物的因各种因素引起的消化不良，急、慢性肠炎及菌群失调性腹泻。尤其是对出血性大肠杆菌感染性肠炎治疗更为有效。
Claims (5)
1、一种枯草芽孢杆菌BS-3菌株(Bacillus Subtilis BS-3)，其在中国微生物菌种保藏管理委员会普通微生物中心的保藏编号为CGMCC.NO.0321。
2、权利要求1所述的枯草芽孢杆菌BS-3菌株(Bacillus Subtilis BS-3)作为制备治疗消化不良、急、慢性肠炎及菌群失调性腹泻药剂的应用。
3、根据权利要求2所述的枯草芽孢杆菌BS-3菌株(Bacillus SubtilisBS-3)的应用，其特征在于：该药剂为胶囊制剂，该胶囊制剂中各组份的重量份数比为：
枯草芽孢杆菌BS-3菌株的原菌粉为1.5-2.5份
赋形剂为96-100份，
其中枯草芽孢杆菌BS-3菌株的原菌粉中的活菌数为≥200亿个活菌/克。
4、根据权利要求2所述的枯草芽孢杆菌BS-3菌株(Bacillus SubtilisBS-3)的应用，其特征在于：该药剂为粉制剂，该粉制剂中各组份的重量份数比为：
枯草芽孢杆菌BS-3菌株的原菌粉为1.5-2.5份
赋形剂为96-100份，
其中枯草芽孢杆菌BS-3菌株的原菌粉中的活菌数为≥200亿个活菌/克。
5、根据权利要求3或4所述的枯草芽孢杆菌BS-3菌株(BacillusSubtilis BS-3)的应用，其特征在于：所述的赋形剂为乳糖和可溶性淀粉，其中乳糖与可溶性淀粉的重量份数比为：
乳糖 9-11份
可溶性淀粉 87-89份，
还有，所述原菌粉是由湿菌体和无水碳酸钙按1∶3重量比混匀干燥制成的。
