技术领域
本发明涉及真菌提取物,特别涉及一种牛樟芝提取物及其制备方法和应用。
背景技术
牛樟芝(Antrodia camphorata,原先命名为Antrodiacinnamomea)是一种寄生于牛樟树(CinnamomumkanchiraiHayata,Lauraceae)中空树干内的真菌,为台湾本地特有药用真菌。在台湾民俗医学上,樟芝是良好的解毒剂,可用于食物中毒或农药中毒,民间亦常用以治疗肝炎或肝脏相关疾病。因为其具有高度医药价值,但栽培不易且生长速度缓慢,因此现已有针对牛樟芝基因体学及代谢体学的研究(Lu et al.2014 PNAS、Lin et al.2011J Agr Food Chem),意图对牛樟芝的医药用途及栽培方法有进一步认识。
牛樟芝内含多种生物活性物质可用于医药用途,包含大分子的多糖和小分子的萜类。其中多糖以不同单糖单元组成存在,经光谱分析后具有大量1,3-β-D-葡聚糖(1,3-β-D-glucan)的成分。牛樟芝多糖已被报导具有多种医疗用途,包括抑制血管新生(Yang etal.2009 J Ethnopharmacol、Cheng et al.2005 Life Sci)、抑制免疫(Menget al.2012Nutrition)、调节免疫反应并抑制气喘(Liu et al.2010 Immunology)、抑制B型肝炎病毒(Lee et al.2002 FEMS Microbiol Lett)、抑制癌细胞生长(Liu et al.2004ToxicolApplPharmacol、Lee et al.2014 Food Funct)等;另已知安卓糖(antrodan),一牛樟芝萃取的糖蛋白(glycoprotein、protein-bound polysaccharide),具有避免肝脏损伤的功效(Ker et al.2014 PLoS ONE、TW500628、US 7763723)。
此外,三萜类(Triterpenoids)亦为牛樟芝主要医药用化合物,也是牛樟芝食用时苦味主要来源。早期Chernget al.发表以麦角甾烷(ergostane)类的3种新颖三萜化合物:antcins A-C(Chernget al.1995 J Nat Prod),以及另有4种新颖三萜化合物:antcins E-F、methyl antcinate G、methyl antcinate H(Chernget al.1996 Phytochemistry);之后数十种牛樟芝三萜化合物已被报导具有的医疗功效,包括抑制癌细胞生长(Wu et al.2010J Nat Prod)、抑制免疫反应(Liawet al.2013 J Nat Prod)、治疗肝癌或肝炎(Lien etal.2014 Molecules)、抵抗疲劳(Huang et al.2012 Evid Based Complement AlternMed)等。
美国专利US7109232揭示自牛樟芝纯化的5种新颖化合物以及其用途,包括抗发炎以及抗肿瘤;US7732482则揭示前述化合物具有抑制器官纤维化的功效。US7342137揭示一牛樟芝新颖化合物群组,其具有抑制多种癌细胞生长之功效。US7745647揭示自子实体分离的新颖二萜类化合物,其具有保护神经细胞之功效。US7994158揭示自牛樟芝纯化之去氢硫色多孔菌酸(dehydrosulphurenic acid)对于肿瘤细胞,特别是血癌或是胰脏癌具有抑制效果。US7531627揭示一分子量为29kDa的牛樟芝新颖蛋白质ACA1,其具有促进免疫功效以及治疗癌症的潜力。
牛樟芝抑制癌症的机制依据化合物的化学结构有所不同。Yehet al.报导倍半萜内酯安卓幸(sesquiterpene lactone antrocin)可抑制JAK2/STAT3的讯息传递路径,进而诱发细胞凋亡(apoptosis)(Yehet al.2013Carcinogenesis);Yang et al.报导可使用牛樟芝可抑制HER-2/neu讯息传导,并抑制卵巢癌(Yang et al.2013 J Ethnopharmacol);Wang et al.则报导安卓奎诺尔D(antroquinonol D)可导致DNA去甲基化,并且具有抑制癌症的潜力(Wang et al.2014 J Agric Food Chem)。
综上所述,牛樟芝含有具有抗癌潜力的物质,并且已经证实对于多种癌症有效果,但对于是否尚存在新颖化合物,以及哪些化合物具有抗癌效果所知甚少。若能自牛樟芝中纯化新颖化合物,并且分析其结构及功效,则可用于癌症的治疗,对于开发新药有所帮助。
发明内容
基于此,有必要提供一种牛樟芝提取物。

所述化合物选自AC006、AC007、AC009、AC011、AC012、AC007-H1、AC009-H1、以及AC012-H1中的一种或至少二种的任意组合:
在其中一个实施例中,所述载剂包含赋形剂、稀释剂、增稠剂、填充剂、黏结剂、崩解剂、润滑剂、油性或非油性的基质、表面活性剂、悬浮剂、胶凝剂、佐剂、防腐剂、抗氧化剂、稳定剂、色素或香料。
在其中一个实施例中,所述治疗癌症的医药组合物是通过抑制癌细胞增生以达到治疗的目的。
在其中一个实施例中,所述治疗癌症的医药组合物是以静脉注射、皮下注射、口服、或涂抹方式施予患者。
在其中一个实施例中,所述治疗癌症的医药组合物的剂型包含溶液、乳剂、悬浮液、粉末、锭剂、油剂、软膏、口含锭、胶囊。
本发明还提供所述的治疗癌症的医药组合物在制备治疗癌症的药物中的应用,所述药物包含治疗有效量的所述的治疗癌症的医药组合物。
在其中一个实施例中,所述癌症选自前列腺癌、肝癌、黑色素瘤、脑癌、直肠结肠癌中的一种或多种。
在其中一个实施例中,所述癌症为直肠结肠癌和/或肝癌。
在其中一个实施例中,所述药物是以静脉注射、皮下注射、口服、或涂抹方式施予患者。
本发明另提供一种制备牛樟芝提取物的方法,包括以下步骤:
取70-230孔洞的硅胶(silica gel)(70-230mesh)与菌丝体重量1:1(w/w)填充管柱,以n-Hexane/乙酸乙酯(Ethyl acetate)梯度冲提,冲出物分成分层(fraction)F1、F2与F3,梯度分别为17-22%%Ethyl acetate、23-27%Ethyl acetate及28-33%Ethylacetate,将F3依停留时间(retention time)顺序分成三个区段F3-1~F3-3;
取F3-1使用移动相为二氯甲烷/丙酮(CH2Cl2/Acetone)以硅胶管柱层析分离(极性由50:1到20:1梯度冲提),取40:1-15:1的区段,进一步使用正相MPLC半制备管柱以n-Hexane/Ethyl acetate(4:1)分离纯化得到化合物AC006;
将F3-2以正相MPLC硅胶管柱,使用移动相CH2Cl2/Acetone梯度分离(100%:0%至70%:30%),取95%:5%至85%:15%区段分成3个区段F3-2-1至F3-2-3;
将F3-2-3进一步分离纯化,使用移动相n-hexane/Acetone(100%:0%至0%:100%)以硅胶管柱层析,取n-hexane/Acetone=90/10至70/30的区段继续以移动相n-hexane/Ethyl acetate(100%:0%至0%:100%)以硅胶管柱层析于n-hexane/Ethylacetate=60/40获得AC007;
F-3-3以正相MPLC硅胶管柱,使用移动相CH2Cl2/Acetone(100%:0%至0%:100%)梯度冲提分离,取90%:10%至70%:30%分成5个区段F3-3-1至F3-3-5;将F-3-3-5(CH2Cl2/Acetone=73/27的区段)进一步分离纯化,使用移动相n-hexane/Acetone(95%:5%至50%:50%)以硅胶管柱取85%:15%至70%:30%的区段层析分成5个区段F3-3-5-1至F3-3-5-5;
取F3-3-5-1(n-hexane/Acetone=90/10-80/20)进一步以逆相MPLC C-18管柱分离纯化,移动相以1%甲酸水溶液/Methanol=35%/65%至20%/80%为梯度,取28%/72%-22%/78%的区段进一步使用移动相n-hexane/Ethyl acetate=80%:20%至50%:50%以硅胶管柱层析,于n-hexane/Ethyl acetate=75%:25%-65%:35%得到AC012;
取F3-3-5-3以逆相MPLC C-18硅胶管柱分离纯化,移动相为1%甲酸水溶液/Methanol=25/75等梯度以15毫升/分钟流度冲提,取以AC009为主的区段(停留时间130-170分钟)进一步纯化,使用移动相CH2Cl2/Ethyl acetate以silica gel梯度冲提(90%:0%至0%:100%)管柱层析,于CH2Cl2/Ethyl acetate=80%/20%-60%/40%得到AC009;
取F3-3-5-4(n-hexane/Acetone=76/24)以逆相HPLC C18硅胶管柱分离纯化,移动相为1%甲酸水溶液/Methanol=25%:75%等梯度冲提,纯化产物为AC011;其中该AC006、AC007、AC009、AC011、AC012即具生物活性的牛樟芝提取物。
在其中一个实施例中,所述牛樟芝提取物可进一步用于制备其C4为羟基取代基的衍生物,包含以下步骤:
以1mol当量的甲醇去乙酰基所述牛樟芝提取物;
去乙酰基反应完成后加入安柏莱特树脂并用滤纸过滤后得到中间产物;
所述中间产物利用硅胶管柱层析,以正己烷与乙酸乙酯为移动相,冲提比例为正己烷/乙酸乙酯4:1至1:1,约于冲提比例1:1时收集产物;
所述产物以逆相HPLC以及C18半制备管柱纯化,以甲醇/甲酸缓冲液冲提梯度75:25等梯度冲提,即得所述C4为羟基取代基的衍生物。
与现有技术相比,本发明具有以下有益效果:
本发明所述牛樟芝提取物及取代基改造的化合物具有抑制血管新生的功效以及抑制多种癌细胞增生的功效,包括肝癌、脑癌、前列腺癌、乳癌、直肠结肠癌、和黑色素瘤,其抑制癌细胞增生的功效对于肝癌以及直肠结肠癌有更佳的效果,且经取代基改造的化合物其抑制效果较牛樟芝萃取的化合物有显著地提升。

术语「治疗」、「用于治疗」以及其类用语是指称推迟、改善、减少、或逆转患者所罹患的可诊断病症以及该病症造成的相关症状的方法以及预防该病症或任何其所属的相关症状的方法。
术语「药学上可接受」是指称物质或组合物必须与其药学上调配物的其他成分兼容,且不加剧患者的症状。

术语「治疗有效量」系指称产生、造成预期的生物体反应所必须的剂量,且非以治疗痊愈所需为定量。本发明所属技术领域具通常知识者可理解,医药组合物的治疗有效量可视诸如下列等因素而变化:期望生物终点、拟递送生物活性剂、囊封基质(encapsulatingmatrix)之组成、目标组织等等。
实施例1牛樟芝提取物的制备
牛樟芝液态发酵菌丝体以正己烷(n-hexane)回流萃取(reflux)2次,每次1-3小时;抽气过滤后,合并两次n-hexane萃取液;取70-230目之硅胶(silica gel)(70-230mesh)与菌丝体重量1:1(w/w)填充管柱,以n-Hexane/乙酸乙酯(Ethyl acetate)梯度冲提,冲出物分成分层(fraction)F1、F2与F3,梯度分别为17-22%Ethyl acetate、23-27%Ethylacetate及28-33%Ethyl acetate,将F3依停留时间(retention time)顺序分成三个区段F3-1~F3-3。
取F3-1使用移动相为二氯甲烷/丙酮(CH2Cl2/Acetone)以硅胶管柱层析分离(极性由50:1到20:1梯度冲提),取40:1-15:1的区段,进一步使用正相HPLC(Medium pressureliquid chromatography)半制备管柱以n-Hexane/Ethyl acetate(4:1)分离纯化得到化合物AC006。
将F3-2以正相MPLC(Medium pressure liquid chromatography)硅胶管柱,使用移动相CH2Cl2/Acetone梯度分离(100%:0%至70%:30%),取95%:5%至85%:15%区段分成3个区段;将F3-2-3进一步分离纯化,使用移动相n-hexane/Acetone(100%:0%至0%:100%)以硅胶管柱层析,取n-hexane/Acetone=90/10至70/30的区段继续以移动相n-hexane/Ethyl acetate(100%:0%至0%:100%)以硅胶管柱层析于n-hexane/Ethylacetate=60/40获得AC007。
F-3-3以正相MPLC硅胶管柱,使用移动相CH2Cl2/Acetone(100%:0%至0%:100%)梯度冲提分离,取90%:10%至70%:30%分成5个区段;将F-3-3-5(CH2Cl2/Acetone=73/27的区段)进一步分离纯化,使用移动相n-hexane/Acetone(95%:5%至50%:50%)以硅胶管柱取85%:15%至70%:30%的区段层析分成5个区段;取F3-3-5-1(n-hexane/Acetone=90/10-80/20)进一步以逆相MPLC C-18管柱分离纯化,移动相以1%甲酸水溶液/Methanol=35%/65%至20%/80%为梯度,取28%/72%-22%/78%的区段进一步使用移动相n-hexane/Ethyl acetate=80%:20%至50%:50%以硅胶管柱层析,于n-hexane/Ethylacetate=75%:25%-65%:35%得到AC012。
取F3-3-5-3以逆相(reverse phase)MPLC C-18硅胶管柱分离纯化,移动相为1%甲酸水溶液/Methanol=25/75等梯度(isocratic)以15毫升/分钟流度冲提,取以AC009为主的区段(停留时间130-170分钟)进一步纯化,使用移动相CH2Cl2/Ethyl acetate以silicagel梯度冲提(90%:0%至0%:100%)管柱层析,于CH2Cl2/Ethyl acetate=80%/20%-60%/40%得到AC009。
取F3-1并以逆相MPLC C-18硅胶管柱分离纯化,移动相为1%甲酸水溶液/Methanol=25%/75%为移动相等梯度冲提,纯化产物为AC-05-01。
取F3-3-5-4(n-hexane/Acetone=76/24)以逆相HPLC C18硅胶管柱分离纯化,移动相为1%甲酸水溶液/Methanol=25%:75%等梯度冲提,纯化产物为AC011。
其中该萃取物AC006、AC007、AC009、AC011、AC012即为牛樟芝具有抗癌活性的提取物。

实施例3牛樟芝提取物取代基去乙酰基
前述牛樟芝提取物分析其化学结构后,将其C3以及C4的取代基进行去乙酰基改造;其取代基改造的产物若于C4以羟基(-OH)取代者,以H1标示;若其取代基改造的产物若于C4以羟基取代且于C3以二甲氧基(dimethoxy)取代者,以H2标示。该去乙酰基方法详述如下:
AC012以1莫耳(mol)当量甲氧基钠(Sodium methoxide)以及无水甲醇作为溶剂进行去乙酰基反应。去乙酰基过程以TLC(thin layer chromatography)进行监测,直到完全反应完再加入安柏莱特树脂中和反应,再用滤纸过滤后得到中间产物。
该中间产物利用正相系统进行玻璃管柱层析,分离树酯使用硅胶,以hexane/ethyl acetate为移动相,冲提比例从hexane:ethyl acetate=4:1至1:1,冲提过程以TLC进行监测,AC012-H1和AC012-H2之混合物约在hexane:ethyl acetate=1:1时被冲提下来,收集并浓缩,得浓缩的产物。
将该浓缩的产物使用逆相HPLC进行最后的纯化,管柱使用C18半制备不锈钢管柱,移动相以甲醇:0.1%磷酸盐缓冲溶液(phosphate buffered saline)=75:25单一比例进行冲提。AC012-H1之停留时间(retention time)约是31至25分钟;AC012-H2为39至43分钟。

实施例4牛樟芝提取物抑制血管新生的功效
本发明利用SRB试验以及血管形成试验(matrigel capillary tube formationassay),分析自牛樟芝纯化所得之化合物的抑制血管新生的功效。
SRB试验系将内皮前驱细胞(EPC,endothelial progenitor cells)以5x103细胞/培养皿孔洞(cell/well)的数量置于96孔培养皿,并令其处于无血清(serum free)的培养基饥饿48小时,再置于10%FBS培养基以及不同浓度AC012中培养48小时,AC012浓度分别为0.1,0.3,1,3,10and 30微克/毫升(μg/ml)。经处理药物培养后,取出培养基并于培养皿孔洞加入100微升(μl)10%三氯乙酸(trichloroacetic acid)(w/v),于4℃放置1小时以固定细胞。再将培养皿以去离子水洗净晾干。于各孔洞加入50微升0.4%Sulforhodamine B(SRB)(w/v)(另含1%乙酸)并于室温放置5分钟,再以1%乙酸清洗未与细胞结合的SRB,之后晾干。与细胞结合的SRB以100微升10mMTris缓冲液(pH 10.5)溶解,并于亮度计以495nm定量。
Matrigel capillary tube formation试验包含以下步骤:将Matrigel以10μl/well加入15孔培养皿中,于37℃放置30分钟以形成胶膜(gel layer)。形成胶膜后,5×103EPC细胞、VEGF(vascular endothelia growth factor)、以及不同浓度的AC012(0,0.1,0.3,1μg/ml)置入培养皿孔洞中,于37℃、5%二氧化碳的条件下放置16小时。16小时之后,使用反向相位差显微镜(inverted contrast phase microscope(Nikon,Japan))观察细胞生成。
AC012抑制血管新生的功效:IC50为29μg/ml;1μg/ml AC012可抑制31.89%的血管新生。
实施例5牛樟芝提取物抑制癌细胞增生的功效
利用SRB试验分析自牛樟芝纯化的化合物及其衍生物抑制癌细胞增生的功效;此实施例即分析AC006、AC007、AC009、AC011、以及AC012,以及其水解改造取代基的产物,抑制多种癌细胞株增生的功效。
各细胞株分别以5×103cells/well置于96孔培养皿,并令其处于无血清(serumfree)的培养基饥饿48小时,再置于10%FBS培养基以及0.1、0.3、1、3、10、30μg/ml等不同浓度AC006、AC007、AC009、AC012中培养48小时。经处理药物培养后,取出培养基并于培养皿孔洞加入100微升(μl)10%三氯乙酸(trichloroacetic acid)(w/v),于4℃放置1小时以固定细胞。再将培养皿以去离子水洗净晾干。于各孔洞加入50微升0.4%Sulforhodamine B(SRB)(w/v)(另含1%乙酸)并于室温放置5分钟,再以1%乙酸清洗未与细胞结合的SRB,之后晾干。与细胞结合的SRB以100微升10mMTris缓冲液(pH 10.5)溶解,并于亮度计以495nm定量。

以AC006测试5种不同癌症,包括8种不同癌细胞株,其抑制半数癌细胞生长有效浓度,即IC50效果,其结果揭示于表1:

综实施例所揭示,本发明提供牛樟芝提取物及其萃取方法,并且分析其化学结构、揭示其于低浓度之下具有抑制血管新生的功效;基于该牛樟芝化合物,本发明亦提供化学合成新颖化合物的方法。本发明所揭示的新颖萃取方法可大量纯化该化合物、降低培养牛樟芝子实体的成本。
本发明亦揭示该牛樟芝提取物具有抑制癌细胞增生的功效,可用于制备治疗癌症的药物。此外,藉由水解牛樟芝提取物,改造其取代基所得的新颖化合物H1,可有效地提升化合物抑制癌细胞增生的效果,尤其是针对大肠癌细胞株;其中HCT116具有特征为低表现量的Bax(Wang et al.2012)、以及growth factor(TGFαand EGFR)-independent(Howellet al.1998)。此外,本发明提供的牛樟芝提取物对于肝癌细胞亦有高度抑制其增生的功效,特别是针对Huh-7,取自亚裔人种的肝癌上皮细胞株(epithelial-like tumorigeniccells),具有HFE突变之特征(Vecchi et al.2009)。
综上所述,本发明提供纯化牛樟芝提取物的方法、其化学结构、以及其抑制血管新生以及抑制癌细胞增生的功效。

1.一种治疗癌症的医药组合物,其特征在于,包含治疗有效量的化合物,以及至少一种药学上可接受的载剂、盐类、或前驱药物;
所述化合物选自AC007-H1、AC009-H1、以及AC012-H1中的一种或至少二种的任意组合:
所述化合物AC007-H1、AC009-H1、以及AC012-H1的制备方法包括如下步骤:
获取牛樟芝提取物AC007、AC009、AC012;
以1mol当量的甲氧基钠以及甲醇作为溶剂去乙酰基所述牛樟芝提取物;
去乙酰基反应完成后加入安柏莱特树脂并用滤纸过滤后,得到中间产物;
所述中间产物利用硅胶管柱层析,以正己烷与乙酸乙酯为移动相,冲提比例为正己烷/乙酸乙酯4:1至1:1,约于冲提比例1:1时收集产物;

7.根据权利要求6所述的应用,其特征在于,所述癌症选自前列腺癌、肝癌、黑色素瘤、脑癌、直肠结肠癌中的一种或多种。
8.根据权利要求7所述的应用,其特征在于,所述癌症为直肠结肠癌和/或肝癌。
9.根据权利要求6所述的应用,其特征在于,所述药物是以静脉注射、皮下注射、口服、或涂抹方式施予患者。
10.一种制备牛樟芝提取物的方法,其特征在于,包括以下步骤:
将牛樟芝菌丝体以正己烷回流萃取二次,每次一至三小时,再将二次萃取液混合并过滤;
取70-230目的硅胶与菌丝体重量比1:1填充管柱,以正己烷/乙酸乙酯梯度冲提,冲出物分成分层F1、F2与F3,梯度分别为17-22%乙酸乙酯、23-27%乙酸乙酯及28-33%乙酸乙酯,将F3依停留时间顺序分成三个区段F3-1~F3-3;
取F3-1使用移动相为二氯甲烷/丙酮以硅胶管柱层析分离,极性由50:1到20:1梯度冲提,取40:1-15:1的区段,进一步使用正相MPLC半制备管柱以正己烷/乙酸乙酯4:1分离纯化得到化合物AC006;
将F3-2以正相MPLC硅胶管柱,使用移动相CH2Cl2/丙酮梯度分离,由100%:0%到70%:30%梯度冲提,取95%:5%至85%:15%区段分成3个区段F3-2-1至F3-2-3;

F-3-3以正相MPLC硅胶管柱,使用移动相CH2Cl2/丙酮,由100%:0%到0%:100%梯度冲提分离,取90%:10%到70%:30%分成5个区段F3-3-1至F3-3-5;将F-3-3-5,即CH2Cl2/丙酮=73/27的区段进一步分离纯化,使用移动相正己烷/丙酮,由95%:5%到50%:50%梯度冲提分离,以硅胶管柱取85%:15%到70%:30%的区段层析分成5个区段F3-3-5-1至F3-3-5-5;
取F3-3-5-1,即正己烷/丙酮=90/10-80/20的区段进一步以逆相MPLC C-18管柱分离纯化,移动相以1%甲酸水溶液/甲醇=35%/65%至20%/80%为梯度,取28%/72%-22%/78%的区段进一步使用移动相正己烷/乙酸乙酯=80%:20%至50%:50%以硅胶管柱层析,于正己烷/乙酸乙酯=75%:25%-65%:35%得到AC012;
取F3-3-5-3以逆相MPLC C-18硅胶管柱分离纯化,移动相为1%甲酸水溶液/甲醇=25/75等梯度以15毫升/分钟流度冲提,取以AC009为主的区段,即停留时间130-170分钟的区段,进一步纯化,使用移动相CH2Cl2/乙酸乙酯以硅胶梯度冲提,由90%:0%到0%:100%管柱层析,于CH2Cl2/乙酸乙酯=80%/20%-60%/40%得到AC009;
取F3-3-5-4,即正己烷/丙酮=76/24的区段,以逆相HPLC C-18硅胶管柱分离纯化,移动相为1%甲酸水溶液/甲醇=25%:75%等梯度冲提,纯化产物为AC011;

11.根据权利要求10所述的方法,其特征在于,所述牛樟芝提取物可进一步用于制备其C4为羟基取代基的衍生物,包含以下步骤:
以1mol当量的甲氧基钠以及甲醇作为溶剂去乙酰基所述牛樟芝提取物;
去乙酰基反应完成后加入安柏莱特树脂并用滤纸过滤后得到中间产物;所述中间产物利用硅胶管柱层析,以正己烷与乙酸乙酯为移动相,冲提比例为正己烷/乙酸乙酯4:1至1:1,约于冲提比例1:1时收集产物;
所述产物以逆相HPLC以及C18半制备管柱纯化,以甲醇/甲酸缓冲液冲提梯度75:25等梯度冲提,即得所述C4为羟基取代基的衍生物。
13.根据权利要求12所述的治疗癌症的医药组合物,其特征在于,所述载剂包含赋形剂、稀释剂、增稠剂、填充剂、黏结剂、崩解剂、润滑剂、油性或非油性的基质、表面活性剂、悬浮剂、胶凝剂、佐剂、防腐剂、抗氧化剂、稳定剂、色素或香料。
14.根据权利要求12所述的治疗癌症的医药组合物,其特征在于,其是通过抑制癌细胞增生以达到治疗的目的。
15.根据权利要求12所述的治疗癌症的医药组合物,其特征在于,其是以静脉注射、皮下注射、口服、或涂抹方式施予患者。
16.根据权利要求12所述的治疗癌症的医药组合物,其特征在于,其剂型为溶液、乳剂、悬浮液、粉末、锭剂、油剂、软膏、口含锭或胶囊。
17.权利要求12-16任一项所述的治疗癌症的医药组合物在制备治疗癌症的药物中的应用,其特征在于,所述药物包含治疗有效量的所述的治疗癌症的医药组合物。
18.根据权利要求17所述的应用,其特征在于,所述癌症选自前列腺癌、肝癌、黑色素瘤、脑癌、直肠结肠癌中的一种或多种。
19.根据权利要求18所述的应用,其特征在于,所述癌症为直肠结肠癌。
20.根据权利要求17所述的应用,其特征在于,所述药物是以静脉注射、皮下注射、口服、或涂抹方式施予患者。
