This technical sheet provides information about Thiobacillus Broth (B775), which is used for cultivating Thiobacillus species. The broth contains ammonium sulphate, monopotassium phosphate, calcium chloride, ferrous sulphate, magnesium sulphate, and sodium thiosulphate. To prepare, 10.16g of the powder is suspended in 1000ml of distilled water, boiled to dissolve, and autoclaved. Thiobacillus species are identified by turbidity or sulphur precipitation within 7 days of incubating samples in the broth at 25-30°C. Quality control tests confirm the medium supports growth of Thiobacillus thiooxidans

Directions : Suspend 10.16 gms in 1000 ml distilled water. Boil to dissolve the medium completely. Sterilize by autoclaving at 15 lbs pressure (121°C) for 15 minutes & dispense as desired. Principle : Samples are inoculated into Thiobacillus Broth, After incubation at 25-30˚C for about 7 days or more, turbidity or sulphur precipitation on the surface of the liquid or against the walls of the flasks, indicates growth of bacteria.Isolation is subsequently done on Thiobacillus Agar.Thiobacillus forms small sulphur impregnated colonies with clear zones, indicating & formation from thiosulphate oxidation. QC Tests – (I)Dehydrated Medium Colour : Off white Appearance : Homogeneous Free Flowing powder (II)Rehydrated medium pH (post autoclaving/heating) : Self Colour (post autoclaving/heating) : Light amber Clarity (post autoclaving/heating) : Clear to slightly opalescent (III)Q.C. Test Microbiological Cultural characteristics observed after 7 days at 25 - 30°C. MICROORGANISM (ATCC ) GROWTH Thiobacillus thiooxidans Good – luxuriant Thiobacillus thioparus Good – luxuriant
Precautions : 1. For Laboratory Use.
2. Follow proper, established laboratory procedures in handling and disposing of infectious materials. Limitations : 1. Since the nutritional requirements of organisms vary, some strains may be encountered that fail to grow or grow poorly on this medium. Use : B775: For cultivation of Thiobacillus species.
