Statics) course, we have developed a conceptual instrument, the CUE (Colorado Upper-division Electrostatics) diagnostic. Two central goals of this tool are: to assess impacts of transformed curricula, and to systematically identify and document student learning difficulties. We find persistent issues involving students ’ ability to conceptually approach and visualize E&M, to accurately communicate that understanding, and to appropriately identify and apply upper-level problem-solving strategies. Our work underlines the need for further research on the nature of student learning – and appropriate instructional interventions –at the upper division.

Abstract. As part of an effort to systematically improve our junior-level E&M I course, we are developing a tool to assess student learning of E&M concepts at the upper-division. Along with a faculty working group, we established a list of learning goals for the course that, with student observations and interviews, served as a guide in creating the Colorado Upper-Division Electrostatics (CUE) assessment. The result is a 17-question open-ended post-test (with an optional 7-question pre-test) diagnostic, and accompanying grading rubric. We present the preliminary validation of the instrument and rubric, plus results from 226 students in 4 semesters at the University of Colorado, and 4 additional universities.

FK506 is a candidate drug for acute stroke. For such drugs, any decision to proceed to clinical trial should be based on a full and unbiased assessment of the animal data, and consideration should be given to the limitations of those data. Such an assessment should include not only the efficacy of a drug but also the in vivo characteristics and limits to that efficacy. Here we use systematic review and meta-analysis to assess the evidence for a protective effect of FK506 in animal models of stroke. In all, 29 studies were identified describing procedures involving 1759 animals. The point estimate for the effect of FK506 was a 31.3% (95% confidence interval 27.2% to 35.4%) improvement in outcome. Efficacy was higher with ketamine anaesthesia and temporary ischaemia and was lower in rats, in animals with comorbidities, and where outcome was measured as infarct size alone. Ascomycin and FK506 are powerful calcium-dependent serine/threonine protein phosphatase (calcineurin [CaN], protein phosphatase 2B) inhibitors. Their mechanism of action involves the formation of a molecular complex with the intracellular FK506-binding protein-12 (FKBP12), thereby acquiring the ability to interact with CaN and to interfere with the dephosphorylation of various substrates. Pharmacological studies of ascomycin, FK506, and derivatives have mainly been focused on their action as immunosuppressants and therapeutic use in inflammatory skin diseases, both in animal studies and in humans. CaN inhibitors have been also proposed for the treatment of inflammatory and degenerative brain diseases. Preclinical studies suggest, however, that ascomycin and its derivatives exhibit additional pharmacological activities. Ascomycin has been shown to have anticonvulsant activity when perfused into the rat hippocampus via microdialysis probes, and ascomycin derivatives may be useful in preventing ischemic brain damage and neuronal death. Their pharmacological action in the brain may involve CaN-mediated regulation of gamma aminobutyric acid (GABA) and glutamate receptor channels, neuronal cytoskeleton and dendritic spine morphology, as well as of the inflammatory responses in glial cells. FK506 and ascomycin inhibit signaling pathways in astrocytes and change the pattern of cytokine and neurotrophin gene expression. However, brain-specific mechanisms of action other than CaN inhibition cannot be excluded. CaN is a likely potential target molecule in the treatment of central nervous system (CNS) diseases, so that the therapeutic potential of ascomycin, FK506, and nonimmunosuppressant ascomycin derivatives as CNS drugs should be further explored.

Dual specificity mitogen activated protein kinase phosphatase-1 (MKP-1) inactivates extracellular signal-regulated kinase (ERK), p38 and/or c-jun N-terminal protein kinase (JNK) by dephosphorylation via a negative feed-back loop. The aim of the present study was to assess the role of expression of MKP-1 and phosphorylation status of mitogen-activated protein kinases (MAPKs) in promoting cell survival in PC12 cells. We used FK506 and three different monoperoxovanadium complexes (mpVs) as pharmacological tools for manipulation of MKP-1 expression. Peroxovanadium compounds, known to be insulinomimetic agents and protein tyrosine phosphatase inhibitors, are cytotoxic to the cells, they activate JNK and down-regulate MPK-1. On the other hand, FK 506 has transient effect on ERK activation. However, when the agents are used in combination, ERK phosphorylation is prolonged and intensified, MKP-1 expression is increased, and cell survival is enhanced. Abstract. We describe the implementation and evaluation of an online tool to support student generation of multiple choice assessment questions within two consecutive semesters of introductory physics at the University of Edinburgh. We substituted a weekly homework for an assessment activity in which each student was required to participate in using the system. Engagement with the system was high, with contributions generally going beyond the minimum requirements. The quality of submissions was on average high, with the very best questions being remarkably detailed problems rather than exercises. We explore correlations between use of the online system and end of course examination score. We find that students with higher levels of activity in the system scored significantly higher marks on the exam; this effect was seen for students of lower ability as well as for the highest performing students.

Abstract. The majority of “special access ” students at the University of Cape Town are second language English speakers for whom reading the physics textbook is daunting. As a strategy to encourage meaningful engagement with the text, students wrote textbook summaries due the day material was covered in class. The summaries were returned, and they could bring them or re-write them for use during their examinations. A framework was developed to analyze the summaries based on Waywood, defining three cognitive levels seen in mathematics journaling: recounting, summarizing, and dialoging. This framework was refined, expanded, and tested. Interviews with students were conducted for their views on summary writing and survey questions were included on their final exams. The study was carried out in the 2007 spring semester of the “Foundation Physics Course, ” a component of the special access program.

Neuron-astrocyte interactions are critical for signalling, energy metabolism, extracellular ion and glutamate homeostasis, volume regulation and neuroprotection in the CNS. Glutamate uptake by astrocytes may prevent excitotoxic glutamate elevation and determine neuronal survival. However, an excess of glutamate can cause the death of astrocytes. FK506, an inhibitor of calcineurin, and an immunosuppressive drug, is neuroprotective in animal models of neurologic diseases, including focal and global ischaemia. In the present work, we demonstrate that a single injection of FK506 60 min after a transient middle cerebral artery occlusion (MCAo) significantly decreases the number of terminal deoxynucleotidyl transferase nick-end labelling (TUNEL)-positive cells in the ischaemic cortex and striatum.
