Abstract

BIOPOSTUPCI ZA DOBIJANJE 7-ACK I 7-ADKC, važni intermedijeri za dobijanje cefalosporinskih antibiotika, 7-amino-cefalosporanska kiselina (7-ACK) i 7-aminodeacetilcefalosporanska kiselina (7-ADKC), dobijaju se novim biopostupkom u kome se transformisani Penicillium chrysogenum soj gaji u prisustvu adipatne sirovine za napajanje da bi se dobila adipoil-6-APK (6-amino penicilanska kiselina), a zatim in situ ekspresijom sledećih gena sa kojima je P. chrysogenum bio transformisan: 1) ekspandaza gena na pr., iz Cephalosporium acremonium, čiji ekspresioni proizvod konvertuje adipoil-6-APK ekspanzijom prstena u adipoil-7-ADCK; 2) hidroksilaza gena čiji ekspresioni proizvod konvertuje 3-metil bočni lanac adipoil-7-ADCK u 3-hidroksimetil, pri čemu daje prvi proizvod, 7-amino-deacetilcefalosporansku kiselinu (7-ADKC); i 3) acetiltransferaza gena čiji ekspresioni proizvod konvertuje 3-hidroksimetil bočni lanac u 3-acetiloksimetil bočni lanac 7-ACK. Finalni proizvod, 7-ACK, se tada dobija cepanjem adipoil bočnog lanca koristeći adipoil acilazu. Prema tome celokupna sinteza se izvodi koristeći biopostupke, i efikasna je i ekonomična.
