Claims (27)
1. Способ определения последовательности полинуклеотида, предусматривающий стадии
(i) взаимодействия фермента, осуществляющего процессинг полинуклеотида, иммобилизованного в фиксированном положении, с полинуклеотидом-мишенью при условиях, обеспечивающих ферментативную активность;
(ii) обнаружение эффекта, являющегося результатом взаимодействия фермента с полинуклеотидом,
причем эффект обнаруживают путем измерения нелинейного оптического сигнала или линейного сигнала, связанного с нелинейным сигналом.
2. Способ по п.1, при котором эффект обнаруживают путем измерения нелинейного сигнала.
3. Способ по п.1 или 2, при котором нелинейное оптическое определение представляет собой визуализацию генерации второго или третьего колебания.
4. Способ по п.1 или 2, при котором нелинейное оптическое определение представляет собой спектроскопию комбинационного светорассеяния или поверхностную усиленную спектроскопию комбинационного светорассеяния.
5. Способ по п.1 или 2, при котором диполярную молекулу помещают на фермент или вблизи фермента.
6. Способ по п.5, при котором молекула представляет собой молекулу стирилового красителя.
7. Способ по п.5, при котором молекула представляет собой зеленый флуоресцентный белок.
8. Способ по п.5, при котором диполярная молекула присоединена к отдельным основаниям полинуклеотида.
9. Способ по п.1 или 2, при котором фермент является полимеразой.
10. Способ по п.1 или п.2, при котором фермент является геликазой или праймазой.
11. Способ по п.10, при котором стадия (i) предусматривает присоединение нуклеозидтрифосфатов dATP, dTTP, dGTP и dCTP.
12. Способ по п.11, при котором нуклеозидтрифосфаты содержат одну или несколько блокирующих групп, которые могут быть избирательно удалены импульсным монохроматическим светом.
13. Способ по п.1 или 2, при котором наночастицу металла помещают на ферменте или вблизи фермента.
14. Способ по п.13, при котором наночастица является наночастицей золота или серебра.
15. Способ по п.13, при котором наночастицу включают в одно или несколько отдельных оснований полинуклеотида.
16. Способ по п.1 или 2, при котором фермент иммобилизован на твердом носителе.
17. Способ по п.16, при котором имеется множество ферментов, иммобилизованных на твердом носителе.
18. Способ по п.16, при котором твердый носитель имеет шероховатую металлическую поверхность.
19. Способ по п.17, при котором твердый носитель имеет шероховатую металлическую поверхность.
20. Способ по п.16, при котором носитель является серебром или золотом.
21. Способ по п.17, при котором носитель является серебром или золотом.
22. Способ по п.1 или 2, при котором определение проводят в сочетании с атомной силовой микроскопией или ближнепольной сканирующей оптической микроскопией.
23. Способ по п.1 или 2, который дополнительно предусматривает применение локализованного поверхностного плазмонового резонанса.
24. Способ по п.1 или 2, при котором фермент иммобилизован в фиксированном положении в клетке.
25. Вещество твердого носителя, которое содержит, по крайней мере, одну иммобилизованную полимеразу и, по крайней мере, одну диполярную молекулу, помещенную на полимеразу или вблизи полимеразы.
26. Вещество твердого носителя, которое содержит клетку, иммобилизованную на нем, причем клетка содержит полимеразный фермент, поддерживаемый в фиксированном положении в клетке.
27. Устройство системы визуализации для определения нелинейного оптического сигнала, содержащее твердый носитель, содержащий иммобилизованный на нем фермент, который взаимодействует с полинуклеотидом, и диполярную молекулу, помещенную на фермент или вблизи фермента.
