Description
Claims (30)
- (a) 실질적으로 충분한 생물학적 활성을 보유하고, (b) 변형된 탄수화물 구조를 가지며, (c) 변경된 생체내 반감기를 갖고 (d) 기능적으로 변형된 탄수화물 구조를 갖지아니할 경우에는 아미노산 잔기(117)에 관능성 탄수화물 구조를 갖는 인체 조직형 플라스미노겐 활성제 변이체.
- 제1항에 있어서, 상기 탄수화물 구조가 1개 이상의 관능성 탄수화물 구조의 결실에 의해 변형된 것인 증가된 생체내 반감기를 갖는 인체 조직형 플라스미노겐 활성제 변이체.
- 제2항에 있어서, 상기 탄수화물 구조가 아미노산 117 위치에서 관능성 탄수화물 구조가 제거되어 변형된 것인 증가된 생체내 반감기를 갖는 인체 조직형 플라스미노겐 활성제 변이체.
- 제3항에 있어서, 117 위치에서 아스파리긴 이외의 아미노산을 함유하는 인체 조직형 플라스미노겐 활성제 변이체.
- 제3항에 있어서, 119 위치에서 세린 또는 트레오닌 이외의 아미노산을 함유하는 인체 조직형 플라스미노겐 활성제 변이체.
- 제3항에 있어서, 117 위치에서 글루타민을 함유하는 인체 조직형 플라스미노겐 활성제 변이체.
- 제3항에 있어서, 119 위치에서 메티오닌을 함유하는 인체 조직형 플라스미노겐 활성제 변이체.
- 제3항에 있어서, 118 위치에서 프롤린을 함유하는 인체 조직형 플라스미노겐 활성제 변이체.
- 제3항에 있어서, 단일 사슬 인체 조직형 플라스미노겐 활성제인 인체 조직형 플라스미노겐 활성제 변이체.
- 제9항에 있어서 117 위치에서 글루타민을, 275 위치에서 글루탐산을 함유하는 인체 조직형 플라스미노겐 활성체 변이체.
- 제2항에 있어서, 상기 탄수화물 구조가 아미노산 117, 184 및 448 위치에서 관능성 탄수화물 구조가 제거되어 변형되는 것인 증가된 생체내 반감기를 갖는 인체 조직형 플라스미노겐 활성제 변이체.
- 제1항에 있어서, 상기 탄수화물 구조가 아미노산 184 및 448 위치에서 고 만노오스 과당류의 치환에 의해 변형된 것인 감소된 생체내 반감기를 갖는 인체 조직형 플라스미노겐 활성제 변이체.
- 제약학상 허용되는 담체와 함께 혼합시킨 제1항에 따르는 인체 조직형 플라스미노겐 활성제 변이체를 포함하는 제약 조성물.
- 제약학상 허용되는 담체와 함께 혼합시킨 제2항에 따르는 인체 조직형 플라스미노겐 활성제 변이체를 포함하는 제약 조성물.
- 제약학상 허용되는 담체와 함께 혼합시킨 제12항에 따르는 인체 조직형 플라스미노겐 활성제 변이체를 포함하는 제약 조성물.
- 인체 조직형 플라스미노겐 활성제의 탄수화물의 구조를 변형시킴을 특징으로 하는 실질적으로 충분한 생물학적 활성을 보유하고, 변경된 생체내 반감기를 갖는 제1항에 따르는 인체 조직형 플라스미노겐 활성제 변이체의 제조 방법.
- 제16항에 있어서, 상기 변형이 1개 이상의 탄수화물 구조의 제거에 의한 것인 방법.
- 제17항에 있어서, 관능성 탄수화물 구조가 117번 아미노산 잔기에서 제거된 방법.
- 제17항에 있어서, 관능성 탄수화물 구조가 117, 184 및 448번 아미노산 잔기에서 제거된 방법.
- 제16항에 있어서, 상기 변형이 아미노산 184 및 448 위치에서 고 만노스 과당류의 치환에 의한 것인 방법.
- 제16항에 있어서, 인체 조직형 플라스미노겐 활성제 변이체를 재조합 DNA 세포 배양물 중에서 117 위치에 아미노산 아스파라긴을 암호화하는 코돈 이외의 코돈을 갖거나 또는 119 위치에서 아미노산 세린 및 트레오닌을 암호화하는 코돈 이외의 코돈을 갖는 상기 인체 조직형 플라스미노겐 활성제 변이체를 암호화하는 DNA를 발현시킴으로써 제조하는 방법.
- 제21항에 있어서, 상기 코돈이 117 위치에서 글루타민을 암호화하는 것인 방법.
- 제21항에 있어서, 상기 코돈이 119 위치에서 메티오닌을 암호화하는 것인 방법.
- 제21항에 있어서, 상기 코돈이 117 위치에서 글루타민을 암호화하고, 275위치에서 글루탐산을 암호화하는 것인 방법.
- 제16항에 있어서, 인체조직형 플라스미노겐 활성제를 탄수화물의 구조를 변형시키는 시약으로 처리함으로써 탄수화물이 변형되는 방법.
- 제25항에 있어서, 상기 시약이 효소인 방법.
- 제26항에 있어서, 상기 효소가 엔도 H인 방법.
- 제25항에 있어서, 상기 시약이 과요오드산염인 방법.
- 복합 탄수화물 치환체를 갖는 당단백질을 형성할 수 있는 포유동물의 숙주 세포를 인체 조직형 플라스미노겐 활성제를 암호화 하는 DNA로 형질전환시키고, 형질전환된 세포를 배양시킨 후, 세포 배양물로부터 상기 변이체를 회수함을 특징으로 하는 인체 조직형 플라스미노겐 활성제의 제조방법
- 제29항에 있어서, 상기 숙주 세포가 효소 N-아세틸글루코사민 아미노트랜스퍼라제를 가지고 있지 않은 방법.
