Claims (12)
- 외래 유전자의 발현을 가능케 하기 위해 상기 외래 유전자를 발현벡터에 삽입시켜 구축되는 조환체플라스미드를 도입한 대장균의 균체에서 외래 유전자 발현을 제어하는 방법에 있어서, 상기 외래 유전자 발현 제어를 위해 대장균을 배양하기 위한 배지중의 탄소원인 당분농도를 0.3% 이상으로 유지하는 공정을 포함하는 것을 특징으로 하는 외래 유전자 발현 제어방법.
- 제1항에 있어서, 상기 외래 유전자가 L-페닐알라닌 암모니아리아제를 코우딩하는 구조 유전자임을 특징으로 하는 외래 유전자 발현 제어방법.
- 제2항에 있어서, 상기 L-페닐알라닌 암모니아리아제를 코우딩하는 구조 유전자임을 특징으로 하는 외래 유전자 발현 제어방법.
- 제1항에 있어서, 상기 대장균이 조환체 플라스미드 pSYPL-3을 도입한 대장균, 수탁번호 FERMP-9024(MT-10424)임을 특징으로 하는 외래 유전자 발현 제어방법.
- 제1항에 있어서, 상기 대장균이 조환체 플라스미드 pYtrp 6을 도입한 대장균, 수탁번호 FERMP-8876(MT-10414)임을 특징으로 하는 외래 유전자 발현 제어방법.
- 제1항에 있어서, 상기 대장균이 조환체 플라스미드 pSW 115을 도입한 대장균, 수탁번호 FERMP-9023(MT-10423)임을 특징으로 하는 외래 유전자 발현 제어방법.
- 소망의 외래 유전자의 발현을 가능케 하기 위해 상기 외래 유전자를 발현 벡터에 삽입시켜 구축되는 조환체 플라스미드를 도입한 대장균을 배양하여 소망의 외래 유전자 산물을 생산하는 과정에 있어서, (a) 배지중의 탄소원으로서 당분 농도가 0.3%이상으로 유지되는, 상기 대장균을 배양하기 위한 제1배양과정과 (b) 배지중의 상기 당분 농도가 0.3% 미만으로 유지되는, 제1배양 과정에서 증식된 상기 대장균을 배양하기 위한 제2배양과정의 단계들로 이루어지는 것을 특징으로 하는 외래 유전자 산물의 생산방법.
- 제7항에 있어서, 상기 외래 유전자 산물이 L-페닐알라닌 암모니아리아제임을 특징으로 하는 외래 유전자 산물의 생산방법.
- 제8항에 있어서, 상기 L-페닐알리닌 암모니아리아제가 제3항에 지시된 아미노산 배열을 가짐을 특징으로 하는 외래 유전자 산물의 생산방법.
- 제7항에 있어서, 상기 대장균이 조환체 플라스미드 pSYPL-3을 도입한 대장균, 수탁번호 FERMP-9024(MT-210424)임을 특징으로 하는 외래 유전자 산물의 생산방법.
- 제7항에 있어서, 상기 대장균이 조환체 플라스미드 pYtrp 6을 도입한 대장균, 수탁번호 FERMP-8876(MT-10414)임을 특징으로 하는 외래 유전자 산물의 생산방법.
