Claims (14)
- 반응 배지 중에서 카르니틴아미드를 (A) 카르니틴아미드를 가수분해하여 카르니틴을 생성할 수 있는 아미다제 또는 (B) 아미다제를 함유하는 미생물과 접촉시켜 카르니틴아미드를 가수분해시킴을 특징으로 하는 카르니틴 제조방법.
- 제1항에 있어서, 언급한 카르니틴아미드가 DL-카르니틴아미드며 언급한 카르니틴은 L-카르니틴인 방법.
- 제1항에 있어서, 언급한 카르니틴아미드가 L-카르니틴아미드이며 언급한 카르니틴은 L-카르니틴인 방법.
- 제1항에 있어서, 언급한 카르니틴아미드가 D-카르니틴아미드이며 언급한 카르니틴은 D-카르니틴인 방법.
- 제1항에 있어서, 언급한 미생물이 슈도모나스(Pseudomonas)속에 속하는 미생물인 방법.
- 제1항에 있어서, 언급한 미생물이 CA 32-C(FERM BP-1379), CA10-1-5(FERM BP-1376), CA27B1(FERM BP-1380), CA30-11B(FERM BP-1378), CA 28-50A(FERM BP-1377) 및 CA30-35(FERM BP-1375)로 이루어진 그룹 중에서 선택된 균주 중의 분류학적 특성을 갖는 균주의 방법.
- 제1항에 있어서, 카르니틴을 생성하기 위한 카르니틴아미드의 가수분해반응을, 언급한 카르니틴아미드의 농도가 반응 배지의 중량을 기준으로 하여 0.1 내지 30중량%가 되도록 하면서 5 내지 60℃ 및 pH 4 내지 10에서 1 내지 100시간동안 수행하는 방법.
- 제2항에 있어서, 언급한 아미다제가 다음의 물리화학적 특징을 갖는 방법; (1) 작용 및 특이성 : L-카르니틴아미드를 가수분해하여 L-카르니틴 및 암모니아를 생성함. (2) 최적 pH 및 안정한 pH 범위 : 최적 pH : 6내지 7, 안정한 pH 범위 : 7 내지 8, (3) 최적 온도 : 26 내지 40℃에서 활동이 양호하며 그의 최적온도는 40℃이다. (4) 불활성화 조건 : 저온일수록 더욱 안정하다. 4℃에서 7일동안 90% 또는 그 이상의 화렁도를 보유한다 45℃ 또는 그 이상에서 활성도는 급격히 감소된다. (5) 분자량 : 겔 여과법(셀룰로핀 GC-700m을 사용)에 의해 수득된 효소의 분자량은 약 36,000이다. (6) 억제제 : 고농도(10mM)에서 Ag이온에 의해 억제된다 EDTA 및 2-머캅토에탄올에 의해서는 100mM에서도 억제되지 않는다.
- L-카르니틴아미드를 가수분해하여 L-카르니틴을 생성하는 가수분해 반응을 촉매할 수 있는 카르니틴 아미드 가수분해요소.
- 제9항에 있어서, 다음의 물리화학적 특성을 갖는 카르니틴아미드 가수분해효소 : (1) 작용 및 특이성 : L-카르니틴아미드를 가수분해하여 L-카르니틴 및 암모니아를 생성한다. (2) 최적 pH 및 안정한 pH 범위 : 최적 pH : 6 내지 7, 안정한 pH 범위 : 7 내지 8, (3) 최적 온도 : 26 내지 40℃에서 활동이 양호하며 그의 최적 온도는 40℃이다. (4) 불황성화 조건 : 저온일수록 더욱 안정하다. 4℃에서 7일동안 90% 또는 그 이상의 활성도를 보유한다. 45℃또는 그 이상에서 활성도는 급격히 감소된다. (5) 분자량 : 겔여과법(셀룰로핀 GC-700m을 사용)에 의해 수득한 효소의 분자량은 약 36,000이다. (6) 억제제 : 고농도(10mM)에서 Ag이온에 의해 억제된다. EDTA 및 2-머캅토에탄올에 의해서는 10mM에서도 억제되지 않는다.
- L-카르니틴아미드를 가수분해하여 L-카르니틴을 생성하는 가수분해 반응을 촉매할 수 있는 L-카르니틴아미드 가수분해효소 생성가능한 미생물을 카르니틴아미드, 카르니틴 및 γ-부티로베타인으로 이루어진 그룹 중에서 선택되는 적어도 하나의 화합물을 함유하는 배지 중에서 배양시킴을 특징으로 하는 L-카르니틴아미드 가수분해효소의 생성방법.
- 제11항에 있어서, 언급한 미생물이 슈도모나스 속에 속하는 미생물인 방법.
- 제12항에 있어서, 언급한 미생물을 탄소 및 질소공급원 및 무기염을 함유하는 영양배지중, pH 4 내지 9 및 10 내지 40℃에서 성장시키는 방법.
- 제12항에 있어서, 언급한 미생물이 CA 32-C(FERM BP-1379), CA10-1-5(FERM BP-1376), CA27B1(FERM BP-1380), CA30-11B(FERM BP-1378), CA28-50A(FERM BP-1377) 및 CA30-35(FERM BP-1375)로 이루어진 그룹중에서 선택된 균주 분류학적 특성을 갖는 균주인 방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
