Claims (1)
- 다음 일반식(Ⅱ)의 s-설포네이트를 수용성 매질 ml당 약 10mg까지의 농도로 수용성 매질중에서 각 -SSO3-잔기당 약 1내지 5-SH 잔기를 얻을 수 있는 량의 메르캅탄과 pH 약 7 내지 11.5에서 반응시킴을 특징으로 하는 상기 일반식(Ⅰ)의 인슐린 전구체의 제조방법.상기 일반식에서R은 수소, 화학적 또는 효소적으로 분해 가능한 아미노산 잔기 또는 2이상의 아미노산 잔기를 가진 화학적 또는 효소적으로 분해 가능한 펩타이드 부분이고Y는 ―Lys―Z―이고(여기에서 Z는 Ala, Thr 또는 Ser임).(B-29) (B-30)A-1 내지 A-21의 부분은 인슐린 A쇄이고B-1 내지 B-30의 부분은 인슐린 B쇄이고X는 A-1의 아미노 그룹에서 인슐린 A-쇄와, B-29의 ε-아미노그룹 또는 B-30의 카복실 그룹에서 인슐린 B-쇄에 결합된 부위이며 이 부위는 분열이 없이 A-쇄 및 B-쇄로 부터 효소적 또는 화학적으로 분리될 수 있다.
