Description
도 1은 본 발명에 이용된 분화 프로토콜의 개요를 나타낸다.

도 2는 본 발명에 이용된 분화 프로토콜의 다양한 시기와 관련된 마커의 발현을 나타낸다. 패널 a)는 제1기로 분화된 인간 배아 줄기 세포주 H1 세포에 있어서 FACS에 의해 결정할 때 CXCR4의 발현을 나타낸다. 패널 b)는 제1기로 분화된 인간 배아 줄기 세포주 H1 세포에 있어서 실시간 PCR에 의해 결정할 때 제1기와 관련된 마커의 발현을 나타낸다.

도 3은 본 발명에서 이용된 분화 프로토콜의 제6기로 분화된 그리고 후속적으로 액티빈 A로 처리된 (제7기) 인간 배아 줄기 세포주 H1 세포의 디티존 염색을 나타낸다. 패널 a)는 40㎛의 세포 여과기(strainer)에 통과시키기 전에 디티존으로 염색된 세포의 위상차를 나타낸다. 패널 b)는 40㎛의 세포 여과기를 통과할 수 있었던, 디티존으로 염색된 세포의 위상차를 나타낸다. 패널 c는 40㎛의 세포 여과기 통과가 가능하지 않았던, 디티존으로 염색된 세포의 위상차를 나타낸다.

도 4는 실시예 2에서 설명된 방법에 따라 본 발명에서 이용된 분화 프로토콜의 제6기로 분화된 그리고 제6기로의 분화 이후 액티빈 A로 처리된 인간 배아 줄기 세포주 H1 세포에 있어서 실시간 PCR에 의해 결정할 때 pdx-1 (패널 a), nkx6-1 (패널 b), Pax4 (패널 c), nkx2.2 (패널 d), 인슐린 (패널 e) 및 글루카곤 (패널 f)의 발현을 나타낸다.
Claims (7)
- 췌장 내분비 계통과 관련된 마커의 발현을 증가시키기에 충분한 양의 TGF-β 수용체 작용제를 포함하는 배지로 췌장 내분비 계통과 관련된 마커를 발현하는 세포를 처리하는 단계를 포함하는, 췌장 내분비 계통과 관련된 마커의 발현을 증가시키는 방법.
- 제1항에 있어서, TGF-β 수용체 작용제가 액티빈(activin) A, 액티빈 B, 액티빈 C, 액티빈 A의 펩티드 변이체, GDF-8, GDF-11 및 GDF-15로 이루어진 군으로부터 선택되는 방법.
- 제1항에 있어서, 췌장 내분비 계통과 관련된 마커를 발현하는 세포를 농축시키는 방법.
- a. 만능 줄기 세포를 배양하는 단계,
b. 만능 줄기 세포를 완성 내배엽 계통의 특징적인 마커를 발현하는 세포로 분화시키는 단계,
c. 완성 내배엽 계통의 특징적인 마커를 발현하는 세포를 췌장 내배엽 계통의 특징적인 마커를 발현하는 세포로 분화시키는 단계, 및
d. 췌장 내배엽 계통의 특징적인 마커를 발현하는 세포를 췌장 내분비 계통의 특징적인 마커를 발현하는 세포로 분화시키는 단계를 포함하는, 만능 줄기 세포를 분화시키는 방법. - 제4항에 있어서, 췌장 내분비 계통과 관련된 마커의 발현을 증가시키기에 충분한 양의 TGF-β 수용체 작용제를 포함하는 배지로 췌장 내분비 계통의 특징적인 마커를 발현하는 세포를 처리함으로써 췌장 내분비 계통의 특징적인 마커의 발현을 증가시키는 방법.
- 제4항에 있어서, TGF-β 수용체 작용제가 액티빈 A, 액티빈 B, 액티빈 C, 액티빈 A의 펩티드 변이체, GDF-8, GDF-11 및 GDF-15로 이루어진 군으로부터 선택되는 방법.
- 제4항에 있어서, 췌장 내분비 계통과 관련된 마커를 발현하는 세포를 농축시키는 방법.
