Abstract
Description
도 2는 실시예 2에서 유가 배양 세포의 제2 세트에 대해 사용된 세포주에 대한 계대 동안 배가시간에서의 클론 생존도를 도시한 도면. 이 실시예에서, MAb B를 발현시키는 세포주로부터의 세포를 형질감염시켜 Mgat1, Mgat2 및/또는 Slc35A2를 과발현시켰다. Claims (9)
- 포유류 세포 배양 방법 동안 관심 대상의 재조합 단백질의 고수준 만노스(high mannose) 글리코폼(glycoform) 함량을 조절하도록 형질감염된 단리된 포유류 숙주 세포이며,
상기 포유류 숙주 세포는 N-글리코실화 경로에 수반되는 단백질을 과발현하도록 형질감염된 것이고, 여기서 상기 N-글리코실화 경로에 수반되는 단백질은 N-아세틸-글루코사미닐트랜스퍼라제-1 (Mgat1에 의해 암호화됨) 및 N-아세틸-글루코사미닐트랜스퍼라제-2 (Mgat2에 의해 암호화됨), 및 이들의 조합물로 이루어진 군으로부터 선택되고,
