이를 위하여 Detroit 세포를 12-well plate에 분주한 후, 세포 배양 조건에서 24시간 배양하였다. 이후 정제수를 첨가한 대조군과 시험 물질인 조성물을 최종 0.1, 1, 5, 10 %인 농도가 되도록 세포에 처리하고, 2시간 후 Topview 5450 VRI SMD LED를 이용하여 블루라이트를 조사(114 mW/cm2, 30분)하였다. 이후 24시간 동안 세포를 추가 배양하였다. 상기 과정을 통해 배양한 세포의 용해물을 희석한 일부 50 μL를 취해 ATP assay kit(Abcam, Cat. ab83355) 내 ATP 표준물질 50 μL와 함께 각각 96-well plate에 넣고 50 μL의 ATP reaction mix 용액을 또한 각 well에 첨가하여 차광한 상태에서 30분간 실온 반응시켰다. 이후 570 nm에서 흡광도를 측정한 후 ATP 표준농도곡선을 작성하여 시료 내 ATP 양을 산정하였다. 본 발명의 조성물에 의한 열노화 방지 효과를 평가하였다. 즉, 본 발명에 의한 조성물을 농도별로 포함되도록 화장수 제형화한 후 피부에 적용하여 열노화 방지 효과를 확인하였다. 20 ~ 30대 건강한 피험자 25명의 전완부에 실시하였으며, 시험 전 20분간 피험자의 실험 부위(전완부)를 직사광선에 노출시켰다. 시험 시료와 대조 시료를 전완부의 선정된 시험부위 (2 cm * 2 cm)에 Micro Pipette을 사용하여 각각 50 μL 양으로 도포하였으며, 시료 도포 전, 도포 직후, 도포 5분 후, 도포 20분 후의 온도를 측정하였다. 열노화 방지 효능 비교를 위한 음성 대조군으로 물을 사용하였고, 측정 후 각 시료의 온도 감소율은 아래의 수학식에 따라 도출하였다.The effect of preventing heat aging by the composition of the present invention was evaluated. 한련 추출물, 케일잎 추출물, 서양민들레잎 추출물, 순무잎 추출물, 당근전초 추출물, 서양호박 추출물, 고구마뿌리 추출물, 수박 추출물, 및 울금뿌리줄기 추출물을 함유한 제1항 내지 제6항 및 제8항 중 어느 한 항에 따른 화장료 조성물의 제조방법.(a) preparing nasturtium extract, kale leaf extract, dandelion leaf extract, turnip leaf extract, carrot spring extract, pumpkin extract, sweet potato root extract, watermelon extract, and turmeric rhizome extract; and
(b) mixing the extract prepared in step (a) with purified water and heating to obtain a composite extract after hot water extraction,
Claims 1 to 6 and 8 containing nasturtium extract, kale leaf extract, dandelion leaf extract, turnip leaf extract, carrot starch extract, pumpkin extract, sweet potato root extract, watermelon extract, and turmeric rhizome extract. Method for producing a cosmetic composition according to any one of the preceding.
