단메밀의 경우, 4~5cm 발아한 실험군의 함량이 91.93mg/g으로 가장 높았으며 발아하지 않은 실험군에 비해 약 1.2배 많았다(도 2A). 쓴메밀의 경우, 5~6cm 발아한 실험군의 함량이 196.67mg/g으로 가장 높았으며 발아하지 않은 실험군에 비해 약 1.2배 많았다(도 2B). 이러한 결과는 DPPH 자유 라디컬 소거능과 비슷한 경향을 보였는데, 메밀이 함유하고 있는 클로로겐산(Chlorogenic acid), 토코페롤 등의 페놀계 화합물들의 증가에 따른 것으로 사료된다.In the case of buckwheat, the content of the experimental group germinated 4 to 5 cm was the highest at 91.93 mg / g, which was about 1.2 times that of the ungerminated experimental group (Fig. 2A). 단메밀의 경우, 4~5cm 발아할 때까지 티로시나제 억제 활성이 증가하는 것을 확인할 수 있었다(도 3A). 4~5cm 발아한 실험군에서 44.17%의 활성을 보인 후, 다시 천천히 억제 활성이 감소하는 경향을 보였다. 발아하지 않은 실험군은 76.49%의 활성으로 가장 낮은 티로시나제 활성을 보인 실험군 보다 약 1.7배 높은 활성을 보였다. 실시예Example 4. 4. HPLCHPLC 분석 analysis
단메밀의 경우, 발아하지 않았을 때 검출되지 않았던 오리엔틴(orientin)과 이소오리엔틴(isoorientin)이 발아가 시작되면서 검출되는 것을 확인할 수 있었다(도 4a 및 도 4b). 루틴을 비롯한 5개 플라보노이드 화합물이 발아가 4~5cm 진행될 때까지 꾸준히 증가하는 것을 확인할 수 있었으며, 이후 감소하는 경향을 보였다. 루틴, 비텍신, 이소비텍신은 발아하지 않았을 때, 각각 16.96mg/mL, 5.49mg/mL, 5.44mg/mL의 함량을 보였으며, 가장 높은 함량을 보인 4~5cm 발아한 실험군에서 120.49mg/mL, 126.67mg/mL, 118.53mg/mL로 약 7.1배, 23배, 21.8배로 크게 증가하였다. 오리엔틴과 이소오리엔틴은 발아하지 않은 실험군에서 검출되지 않다가 발아가 시작되면서 검출되어 4~5cm 발아한 실험군에서 각각 187.56mg/mL, 161.75mg/mL로 크게 증가한 함량을 보였다(표 2).In the case of buckwheat, it was confirmed that orientin and isoorientin, which were not detected when not germinated, were detected at the beginning of germination (Figs. 4A and 4B). 쓴메밀의 경우, 발아하지 않은 실험군에서 루틴 밖에 검출되지 않았으나 발아가 시작되자 오리엔틴을 비롯한 4개 화합물들이 검출되는 것을 확인할 수 있었다(도 5a 및 도 5b). 쓴메밀은 발아가 5~6cm 진행될 때까지 꾸준히 증가하다가 감소하는 경향을 나타냈다. 가장 높은 함량을 보인 5~6cm 발아한 실험군의 루틴 함량은 1334.67mg/mL로 발아하지 않은 실험군의 함량인 947.65mg/mL보다 약 1.4배 증가하였다. 오리엔틴, 이소오리엔틴, 비텍신, 이소비텍신은 발아하지 않았을 때 나타나지 않았다가 5~6cm까지 꾸준히 증가하였고, 이후 감소하였다. 5~6cm 발아한 실험군에서 오리엔틴, 이소오리엔틴, 비텍신, 이소비텍신의 함량은 각각 6.94mg/mL, 5.79mg/mL, 14.83mg/mL, 19.64mg/mL였다(표 2).In the case of bitter buckwheat, only the routine was detected in the ungerminated experimental group, but when the germination started, it was confirmed that four compounds including orientin were detected (FIGS. 5A and 5B).
