Claims (9)
- 트라우스토키트리알레스 종 과(Thraustochytriales sp. family)에 속하는 미세조류를 발효시켜 생성된 스쿠알렌을 유기 용매 없이 추출하기 위한 방법으로서,
1) 세포간 가용성 물질의 농도를 낮춰서 발효 배지의 총 건조 중량에 대한 생물체량의 건조 중량으로 표현되는 순도가 30 내지 99% 사이, 또는 95% 초과 내지 99 %를 달성하도록 트라우스토키트리알레스 종 과에 속하는 미세조류의 생물체량을 준비하는 단계;
2) 효소 처리에 의해 생성되는 에멀젼 형성을 방지하면서, 상기 미세조류의 세포벽을 깨뜨리기 위해 중성 또는 염기성 프로테아제들의 군으로부터 선택된 프로테아제 효소, 또는 알칼라아제를 사용하여, 상기 생성된 생물체량을 처리하는 단계;
3) 수상으로부터 오일을 분리하기 위해 상기 생성된 반응 혼합물을 원심분리하는 단계; 및
4) 이렇게 생성된 스쿠알렌이 풍부한 미가공 오일을 회수하는 단계
를 포함하고,
여기서 트라우스토키트리알레스 종 과에 속하는 미세조류가 시조카이트리움 속(Schizochytrium sp.), 오란티오키트리움 속(Aurantiochytrium sp.) 및 트라우스토키트리움 속(Thraustochytrium sp.)으로 이루어진 군으로부터 선택되는 것을 특징으로 하며, 상기 유기 용매는 에탄올을 제외하는 것인, 방법. - 제1항에 있어서,
단계 1)에서 정제된 생물체량은 그 후 6% 내지 12% 사이의 건조 물질 함량으로, 또는 10% 내지 12% 사이의 건조 물질 함량으로 조정되는 것을 특징으로 하는, 방법. - 제1항 또는 제2항에 있어서,
단계 2)의 효소 처리는, 프로펠러 교반기와 배플을 구비한 장치에서 비-전단, 에멀젼화용 약한 교반으로 수행되는 것을 특징으로 하는, 방법. - 제1항 또는 제2항에 있어서,
단계 2)의 효소 처리는 50℃ 초과, 또는 대략 60℃의 온도 및 pH가 7 초과, 또는 대략 8에서 수행되는 것을 특징으로 하며, 여기서 "대략"은 언급된 값의 ±10%를 가리키는 값을 의미하는 것인, 방법. - 제1항 또는 제2항에 있어서,
효소 처리의 종료 시 5%(v/v) 초과, 또는 대략 10%(v/v)로 에탄올을 첨가하는 것을 특징으로 하며, 여기서 "대략"은 언급된 값의 ±10%를 가리키는 값을 의미하는 것인, 방법. - 제5항에 있어서,
에탄올 처리는 10분 초과, 또는 15분 간 교반하면서 수행되는 것을 특징으로 하는, 방법. - 제1항 또는 제2항에 있어서,
원심 분리 전, 반응 혼합물의 온도는 70 내지 90℃ 사이, 또는 80℃이고, 반응 혼합물의 pH는 8 내지 12 사이의 값, 또는 10값이 될 수 있는 것을 특징으로 하는, 방법. - 제1항 또는 제2항에 있어서,
원심 분리는 수상 및 세포 찌꺼기로부터 추출된 가벼운 상부의 상(오일)의 회수를 가능하게 하는 농축기 모드의 3-산출 분리기(3-output separator)에서 수행되는 것을 특징으로 하는, 방법. - 제8항에 있어서,
원심 분리는 4000g 초과, 또는 6000 및 10000g 사이, 또는 대략 10000g의 원심력으로 수행되는 것을 특징으로 하며, 여기서 "대략"은 언급된 값의 ±10%를 가리키는 값을 의미하는 것인, 방법.
