Claims (30)
- 숙주 세포에서 발현될 수 있는 유전자 발현 카세트를 포함하는 유전자 발현 조절 카세트로서, 상기 유전자 발현 카세트는ⅰ) 엑디손 수용체 트랜스활성 도메인, VP16 트랜스활성 도메인, B42 산성 활성자 트랜스활성 도메인, 및 p65 트랜스활성 도메인으로 구성된 군으로부터 선택되는 트랜스활성 도메인;ⅱ) 그 발현이 조절되는 유전자와 연관되어 있는 반응 요소를 인식하며, 엑디손 수용체 DNA-결합 도메인, GAL4 DNA-결합 도메인, 및 LexA DNA-결합 도메인으로 구성된 군으로부터 선택되는 DNA-결합 도메인; 및ⅲ) 적어도 하나의 돌연변이를 포함하는 엑디손 수용체 리간드 결합 도메인으로 구성되는 폴리펩티드를 코딩하는 폴리뉴클레오티드를 포함하며,상기 돌연변이는 서열번호 1의 132 아미노산 잔기에 있는 것을 특징으로 하는 유전자 발현 조절 카세트.
- 제1항에 있어서, 척추동물 레티노이드 X 수용체 리간드 결합 도메인, 무척추동물 레티노이드 X 수용체 리간드 결합 도메인, 울트라스피라클 단백질 리간드 결합 도메인 및 두가지 폴리펩티드 절편을 포함하는 키메라 리간드 결합 도메인으로 구성된 군으로부터 선택되는 핵수용체 리간드 결합 도메인을 추가로 포함하며,상기 폴리펩티드 절편에서 제1 폴리펩티드 절편이 척추동물 레티노이드 X 수용체 리간드 결합 도메인, 무척추동물 레티노이드 X 수용체 리간드 결합 도메인, 울트라스피라클 단백질 리간드 결합 도메인 유래이고, 제2 폴리펩티드 절편이 다른 척추동물 레티노이드 X 수용체 리간드 결합 도메인, 무척추동물 레티노이드 X 수용체 리간드 결합 도메인, 또는 울트라스피라클 단백질 리간드 결합 도메인 유래인 것을 특징으로 하는 유전자 발현 조절 카세트.
- 1) ⅰ) 그 발현이 조절되는 유전자와 연관되어 있는 반응 요소를 인식하며, 엑디손 수용체 DNA-결합 도메인, GAL4 DNA-결합 도메인, 및 LexA DNA-결합 도메인으로 구성된 군으로부터 선택되는 DNA-결합 도메인; 및 ⅱ) 핵수용체 리간드 결합 도메인으로 구성되는 제1 폴리펩티드를 코딩하는 폴리뉴클레오티드를 포함하는 숙주 세포에서 발현될 수 있는 제1 유전자 발현 카세트; 및2) ⅰ) 엑디손 수용체 트랜스활성 도메인, VP16 트랜스활성 도메인, B42 산성 활성자 트랜스활성 도메인, 및 p65 트랜스활성 도메인으로 구성된 군으로부터 선택되는 트랜스활성 도메인; 및 ⅱ) 핵수용체 리간드 결합 도메인으로 구성되는 제2 폴리펩티드를 코딩하는 폴리뉴클레오티드를 포함하는 숙주 세포에서 발현될 수 있는 제2 유전자 발현 카세트를 포함하는 유전자 발현 조절 카세트로서,상기 핵수용체 리간드 결합 도메인 중 하나는 적어도 하나의 돌연변이를 포함하는 엑디손 수용체 리간드 결합 도메인이고, 상기 돌연변이는 서열번호 1의 132 아미노산 잔기에 있는 것을 특징으로 하는 유전자 발현 조절 카세트.

- 제12항의 발현 벡터를 포함하는 단리된 숙주 세포로서, 상기 전사 조절 인자가 상기 숙주 세포에서 기능을 하는 것을 특징으로 하는 단리된 숙주 세포.
- 제10항에 따른 단리된 폴리뉴클레오티드에 의해 코딩되는 단리된 폴리펩티드.
- 적어도 하나의 돌연변이로 구성되는 그룹 H 핵수용체 리간드 결합 도메인으로 구성되는 단리된 폴리펩티드로서, 상기 돌연변이는 서열번호 1의 132 아미노산 잔기에 있는 것을 특징으로 하는 단리된 폴리펩티드.
- 삭제
- 1) 제1항 또는 제3항에 따른 유전자 발현 조절 카세트를 숙주 세포 내로 도입하는 단계; 및 2) 상기 숙주 세포에 리간드를 도입하는 단계를 포함하는 숙주 세포에서 유전자의 발현을 조절하는 방법으로서,상기 조절되는 유전자는 ⅰ) 상기 DNA-결합 도메인에 의해 인식되는 반응 요소; ⅱ) 상기 트랜스활성 도메인에 의해 활성화되는 프로모터; 및 ⅲ) 그 발현이 조절되는 유전자로 구성되는 유전자 발현 카세트의 성분인 것을 특징으로 하는 방법.

- 제1항 내지 제3항 중 어느 하나의 항에 있어서, 서열번호 1의 132 아미노산 잔기에서의 돌연변이는 L132M, L132N, L132V 또는 L132E 인 것을 특징으로 하는 유전자 발현 조절 카세트.
- 제1항 내지 제3항 중 어느 하나의 항에 있어서, 엑디손 수용체 리간드 결합 도메인은 서열번호 1의 125 아미노산 잔기에 있는 돌연변이를 더욱 포함하는 것을 특징으로 하는 유전자 발현 조절 카세트.
- 제23항에 있어서, 서열번호 1의 125 아미노산 잔기에서의 돌연변이는 M125P, M125R, M125E, M125L, M125C, M125W, M125G, M125I, M125N, M125S 또는 M125V 인 것을 특징으로 하는 유전자 발현 조절 카세트.
- 제10항에 있어서, 서열번호 1의 132 아미노산 잔기에서의 돌연변이는 L132M, L132N, L132V 또는 L132E 인 것을 특징으로 하는 단리된 폴리뉴클레오티드.
