본 발명은 젤라틴, 글리세롤과 같은 가소제, 상기 가소제를 위한 2차 메트릭스를 형성할 수 있는 추가성분으로 이루어지는 식용 캡슐 껍질용 조성물에 관한 것이다. 이 제2메트릭스는 가소제에 대한 젤라틴의 상대적인 양을 감소시키며 입에서의 분해시간을 줄인다. 상기 추가성분은 전형적으로는 표백되지 않은 감자녹말 아세테이트이다.
Description
본 발명은 약물 전달 시스템이나 식품 또는 식품 첨가물에 사용될 수 있는, 먹을 수 있으며 기타 연 젤라틴(soft gelatin) 캡슐용 조성물에 관한 것이다.
그 자체가 식용인 캡슐을 사용하여 의약 및 식품을 캡슐화하는 것이 알려져 있다. 이러한 캡슐을 위한 껍질 재료는 보통 잴라틴을 기초로 하므로 풍미나 나쁜 냄새, 색이 생기게 된다. 이러한 캡슐이 매우 성공적으로 사용되고 있기는 하나, 젤라틴-기초 껍질 재료는 상대적으로 분해가 느리다. 특히 경구 투여를 위한 캡슐화 제품은 입에서 해리되어야 하므로 분해 속도가 더욱 빠른 것이 바람직하다.
젤라틴-기초 캡슐의 예는 EP-A-0 233 231, 미국특허 4,804,542, EP-A-0 199 034, EP-A-0 120 248 및 미국특허 4,744,988, 미국특허 2,580,863에 기재되어 있다.
위에서 언급된 껍질 재료는 보통 젤라틴과, 글리세롤과 같이 생리적으로 허용되는 가소제로 이루어지며, 상기 젤라틴은 상기 가소제를 위한 메트릭스를 형성한다. 이들 구성성분의 상대적인 양은 캡슐화 및 저장 특성을 확보하기 위해 중요하다.
그러나 본 발명자들은 가소제를 위해 2차 메트릭스를 형성하는 추가성분을 함유하는 경우 젤라틴의 양을 감소시킬 수 있다는 것을 알게 되었다. 따라서 본 발명에 따라, 가소제, 상기 가소제를 위한 메트릭스를 형성하는 젤라틴 및 상기 젤라틴과 함께 사용할 수 있으며 상기 가소제를 위한 2차 메트릭스를 형성하는 추가성분으로 이루어지는 조성물이 제공된다. 전형적으로는, 본 발명의 조성물은 젤라틴 18-30중량%와 가소제 30-45%를 함유한다. 추가성분은 보통 감자녹말 아세테이트, 다른 녹말 유도체, 녹말 자체 또는 이들의 혼합물이다.
또한 본 발명은 위의 기준에 따라 제조되는 씹을 수 있는 조성물, 그 조성물로 이루어지는 식용 캡슐도 제공한다. 본 명세서에는 부가적으로 본 발명에 따른 조성물의 제조방법도 기재되어 있으며, 이 제조방법은 추가성분을 물과 가소제와 혼합하고, 이 혼합물에 젤라틴을 첨가하고, 이 혼합물을 가루로 만들고, 이 혼합물을 가열하고, 가열된 혼합물을 정치하고, 수분 손실을 최소화하면서 이 혼합물을 탈기하는 것으로 이루어진다.
본 발명에 따른 바람직한 조성물에서 추가성분의 양은 25중량%를 넘지 않으며, 보통은 12중량%를 넘지 않는다. 보다 바람직한 추가성분은 표백되지 않은 녹말 아세테이트이며, 가장 바람직하게는 감자에서 얻은 것이다. 상표명 PERFECTAMYL GEL MB(Avebe BA사)로 판매되는 것이 적당하다. 상표명 CLEARAM(Roquette Freres사)도 감자녹말 아세테이트로 적당하다.
추가성분의 전형적인 양은 12중량% 이하이다. 양이 많을수록 씹히는 제품을 얻을 수 있으나 용해성이 나빠진다. 부가적으로, 보다 많은 양의 녹말 아세테이트를 추가성분으로 사용하여 제조된 조성물을 검토한 결과, 녹말 엉김이 일어났다. 최대량은 바람직하게는 10%이고, 8%가 특히 바람직하다.
앞에서 특정된 젤라틴의 양은 종래의 젤라틴-기초 캡슐 껍질 조성물에서의 젤라틴의 양보다 실질적으로 적다. 따라서 가소제의 양은 상대적으로 늘어난다. 이것은 추가성분이 존재하는 경우, 캡슐화와 저장에 충분한 강도의 구조를 이루지 못한다고 하는, 젤라틴에 대한 가소제의 영향이 적어지므로 가능하게 된다. 효과면에서 보면, 추가성분은 전형적으로 20-60℃ 범위에서 1차 젤라틴 메트릭스 안에 있으면서 가소제의 역기능을 발휘하지는 않으나 제조물의 부착 표면을 형성하는 경향을 감소시키는 가소제를 위해, 양립할 수 있는 2차 메트릭스를 형성한다. 조성물에 추가성분은 항상 어느 정도 필요하며, 전형적인 최소량은 3중량%이다.
가소제로는 주로 글리세롤이 사용되며, 자일리톨, 솔비톨, 폴리글리세롤, 솔비톨의 비결정 용액, 글루코스, 프락토스 및 기타 동등물의 글루코스 시립 등도 사용된다. 바람직한 대체물 중 하나가 ANIDRISORB(특허된 솔비톨, 솔비탄, 말티톨 및 만니톨의 혼합물, Roquette Freres사)이다. 이들은 단독으로 또는 함께 사용될 수 있다. 글리세롤을 함유하는 가소제 조성물에서 글리세롤은 전형적으로 그 조성물의 최소한 30중량%, 보통은 30-70중량% 차지한다. 글리세롤을 함유하면 “씹는 질감의(chewable)” 제품을 얻을 수 있다. 다른 가소제 또는 가소제 성분을 사용하여 덜 “씹는 질감의” 제품을 얻을 수 있으며, 이들은 공지된 제제보다 더 빨리 분해된다.
본 발명에 따른 조성물의 씹는 질감은 분별된(fractionated) 코코넛유와 같은 오일을 함유함에 의해 촉진될 수 있다. 최고 15%, 바람직하게는 10% 이하를 함유할 수 있으며, 양이 많아지면 제품이 뿌옇게 된다. 바람직한 양은 3-7중량%이고, 전형적으로 5중량%이다. 껍질구조에서 오일은 미세구조의 방울로 분산되며, 이것은 일부 젤라틴이 결합을 형성하여 가소제처럼 행동하는 것을 방지한다. 그러나 가소제는 젤 메트릭스 사이의 공간을 가로질러 수소결합을 하고, 이 공간에 있는 액체의 일부는 젤을 건조시킬 때 제거된다. 젤이 링크를 더 많이 형성할수록 메트릭스는 더 딱딱해진다. 오일은 더욱 씹는 질감인 제품에서 젤라틴에 의한 압력에 의해 방울 중 일부가 뭉치게 되기는 하나, 이와 같이 젤라틴이 더 많이 링크를 형성하는 것을 저해한다.
본 발명의 바람직한 구체예에서는 보통 감자에서 얻은, 표백된 녹말 아세테이트를 보통 12중량% 이하, 바람직하게는 6-10중량% 함유한다. 이러한 감자녹말 유도체는 상표명 PERFECTAMYL GEL 45(Avebe BA사)로 판매되는 것도 적당하다. 이 녹말 유도체는 본 발명에 따른 조성물의 제조에서 용해되며, 따라서 그 조성물을 건조시킬 때까지 용액에 남아 있는다. 이 단계에서 표백된 녹말 아세테이트는 필름을 형성하고, 따라서 벌킹제로 작용한다. 이것은 제조된 조성물에 딱딱한 정도의 원인이 되며, 젤라틴과 바람직한 추가성분; 표백되지 않은 녹말 아세테이트는 이와 반대 작용을 한다. 이 두 녹말의 조합은 표백되지 않은 아세테이트만을 사용하는 경우에 비해 젤라틴을 더 많이 대체할 수 있다. 다양한 용해성 재료를 사용함에 의해서도 비슷한 효과를 얻을 수 있다.
본 발명의 조성물을 사용하여 적당한 방법에 의해 캡슐을 제조할 수 있다. 그 중 두 예가 농축 실린더(concentric cylinder)와 로터리 다이(rotrary dye) 방법이다. 로터리 다이 방법은 R.P. Scherer Corporation와 그 협력사에 의해 수년간 사용되고 있으며, Pharmaceutical Technology의 1985년 9월호에 기재되어 있다. 이 문헌은 본 발명의 명세서 내용에 참고가 된다. 요약하면, 액체 상태의 조성물을 적당하게 준비하여 냉각시키고 그 위에 젤 덩어리를 비 부착성 필름에 놓아 둔 드럼에 바른다. 두개의 비슷한 필름이 캡슐화로 병합되고, 밀봉이 형성되며 이 밀봉은 덩어리가 건조될수록 더 강해진다. 본 발명의 추가성분은 이 단계에서 필름의 강도에 중요한 역할을 하는데, 보다 바람직한 성분은 감자녹말 아세테이트로, 이것은 젤라틴 메트릭스 내에서 녹말 젤을 형성하도록 녹말 분자를 연관시킴에 의해 젤 메트릭스에 구조를 더해준다.
표백되지 않은 녹말에 더하여 표백된 녹말을 사용하면 이 둘이 실질적으로 상호 결합을 하지 않으면서 다른 속도로 젤라틴으로 팽윤되므로 씹는 질감을 더욱 개선할 수 있다. 결과적으로, 씹을 때 쉽게 분리된다.
본 발명을 구체화하는 조성물과 공지의 조성물은 아래의 예에서 설명될 것이다. 조성물의 자세한 내용은 다음과 같다.
[실시예 S1(제제 1 사용)]
감자녹말 아세테이트를 물과 글리세롤과 섞어 슬러지를 형성하여 젤라틴 달인 것(gelatin decoction)을 준비하였다. 저어주면서 젤라틴을 첨가하고 진공하에서 10분동안 혼합물이 가루가 되도록 한 후, 알려진 방법에 따라 순환기가 장치된 수조를 사용하여 혼합물의 온도가 90℃로 유지되도록 용기를 가열하고 35분 동안 방치하여 달인 것을 만들었다. 수분 손실을 최소화하면서 진공 펌프로 이 젤 덩어리를 탈기하였다.
향이 있는 위약(placebo) 페이스트 제제로 캡슐이 쉽게 만들어졌으며, 이것은 잘 씹혀졌다.
[실시예 S2(제제 2 사용)]
슬러지를 형성하도록 녹말 유도체를 물과 글리세롤과 섞어 젤라틴 달인 것을 준비하였다. 젤라틴을 첨가한 후, 알려진 방법에 따라 달인 것을 만들었다.
향이 있는 위약 페이스트 제제로 캡슐이 쉽게 만들어졌으며, 이것은 잘 씹혀졌다.
[실시예 S3(제제 3 사용)]
실시예 S2의 방법에 따라 젤라틴 달인 것을 만들었다. 여기에 색과 향을 첨가하고 고속 블렌더를 이용하여 6.0% 젤라틴 덩이를 만들었다.
향이 있는 위약 페이스트 제제로 캡슐이 쉽게 만들어졌으며, 이것은 잘 씹혀졌다.
[실시예 S4(제제 3 사용)]
젤라틴 달인 것에 색과 향을 첨가하여 섞고 기름 충진 재료와 함께 캡슐 제조에 사용하였다. 70℃의 증류수에서의 분해 실험 결과, 캡슐은 붕괴되었으며 전체 껍질의 분해는 본 발명의 캡슐이 공지의 껍질 제제(6)을 사용한 것보다 약 40-60% 빨랐다.
[실시예 S5(제제 4 사용)]
기름을 제외하고는 제제 4와 같은 성분을 사용하여 실시예 S2의 방법으로 젤라틴 달인 것을 준비하였다. 이 달인 것에 실시예 S3에서 사용한 색과 향을 첨가하고 식물성 기름 3%을 첨가하였다(기름의 총 농도를 5%로 하기 위하여 향유를 2%로 사용하였다).
향이 있는 위약 페이스트 제제로 캡슐이 쉽게 만들어졌으며, 이것은 부드럽고 잘 씹혀졌다.
[실시예 S6, S7, S8(제제 5 사용)]
실시예 S2의 방법에 따라 젤라틴 달인 것을 만들었다. 사용된 젤라틴 달인 것의 양에 대해 3-7.2% 범위로 다양한 색과 향을 첨가하였다.
활성 비타민을 함유하는 향 페이스트 충진제로 캡슐이 쉽게 만들어졌으며, 모든 캡슐은 껍질 기초 제제로 제제 6을 사용한 경우와 비교할 때 씹는 감이 매우 현저하게 개선되어 쉽게 씹혀졌다.
[실시예 S9(제제 5 사용)]
2단계로 젤라틴 달인 것을 만들었다. 먼저, 감자녹말 유도체를 1.5배 무게의 글리세롤과 혼합하고 이 슬러리를 50-60℃로 가열하였다. 이것을, 다른 나머지 성분을 사용하여 알려진 방법으로 제조한 슬러리에 첨가하였다. 60-65℃가 될 때까지 이 혼합물을 고속 블렌더로 혼합하고, 여기에 전체 혼합물에 대하여 모두 6.25% 되도록 색과 향을 첨가하고 더 혼합하였다.
활성 비타민 성분을 함유하며 향이 첨가된 페이스트 충진제로 캡슐이 쉽게 만들어졌으며, 씹히는 특성은 실시예 S7에서 제조된 제제의 캡슐과 유사하였다.
[실시예 S10(종래기술, 제제 6 사용)]
알려진 기술에 따라 같은 색과 향을 혼합하고 같은 충진 제제를 사용하여 표준 껍질을 만들었다. 이 캡슐은 팽윤 전에 씹기 위해 약 2배의 시간(60-65초)이 필요할 정도로 씹는 특성이 매우 질겼다. 또한 입안에서 미끄럽고 끈적거리는 느낌을 주었다.
제제 7부터 11을 사용하여, 먼저 녹말 유도체를 물과 가소제 성분(글리세롤, 솔비톨, 안드리솔브, 폴리글리세롤)과 섞어 슬러리를 만들고, 실시예 S2에서와 같이 여기에 젤라틴을 첨가하였다. 제조된 캡슐들은 물에 접촉시켰을 때 분해 시간이 짧았으나, 씹는 특성은 실시예 S1 내지 S9의 캡슐과 비교할 때 덜 씹혔다.
위에서 실시한 실험으로부터, 일반적으로 연 젤라틴 캡슐에 사용되는 어떠한 가소제라도 본 발명의 조성물에 사용될 수 있음을 알 수 있다. 씹는 특성은 가소제로 글리세를 사용하고 기름을 사용하거나 사용하지 않은 제제에서 가장 현저하게 개선되었으나, 저 젤라틴 함량, 변형 녹말 성분 및 고 가소제 함량으로부터 제조된 메트릭스는 종래의 제제들보다 더 빠르게 분해되었다.
S1부터 S10의 실시예에서 알 수 있듯이, 웰에서 본 발명의 캡슐에서의 재료는 그 자체가 전체 특성에 영향을 주는 중요한 성분을 함유할 수 있다. 이것은 특히 두 성분이 사용전에 분리되어 있을 수 있으며 특히 캡슐 재료와 캡슐화된 제품 사이에 분리되어 유지될 수 있으면 더욱 가치가 있다.
본 발명의 캡슐로 캡슐화 하기 위해 액체 상태로 제공되는 제품은 소수성 담체, 친수성 담체 또는 이들의 조합과 함께 사용된다. 친수성 용매 또는 담체 매질의 예에는 폴리에틸렌 글리콜(PFGs), 특히 PFG 400, PFG 600, 글리콜푸롤, 폴리글리세롤, 프로필렌 글리콜, 에탄올, 물, 글리세롤, 트랜스큐톨, 폴리솔베이트 및 프로필렌 카보네이트 등이 있다.
소수성 용매/담체 매질에는 수소화 천연유, 폴리메틸실록산(디메티콘)과 같은 합성유, 분획화된 코코낫유, 광유, 트리아세틴, 에틸 올레이트와 같은 중성유, 두유, 아라키스유, 옥수수유, 참기름, 올리브유, 겨자씨유, 해바라기유 및 잇꽃유와 같은 기타 종성유가 있다. 점도가 큰 두껍게 한 충진 제품은 느리게 퍼지고 좋은 맛을 개선하므로 바람직하다. 이러한 두꺼운 제품은 껍질과 충진재료 사이의 대비를 줄인다.
본 발명에 따라 구체화된 캡슐은 캡슐화된 내용물에 또는 캡슐 껍질 그 자체에 풍미와 방향 성분을 함유할 수 있다. 적당한 성분에는 레몬, 오렌지, 페퍼민트유와 같은 에센스유, 과일향, 아니스 열매, 감초, 카라멜, 벌꿀, 크림, 기타 향료 및 이들과 다른 풍미 물질의 조합이 있다. 이러한 성분은 International Flavour ＆ Fragrances, IFF(GB) Ltd.(Haverhill, Suffolk, CB9 8LG England)에서 구할 수 있다. 아스파탐, 사카린, 아세설팜 K, 네오헤스페리딘 하이드로클로라이드, 만니톨, 자일리콜, 말티톨과 같은 천연 또는 인용 감미료도 사용할 수 있으며, 소디움 바이카보네이트, 이온교환수지, 사이클로덱스트린과 앱솔베이트와 같은 맛-차단제(taste-masking agent)도 사용될 수 있다. 또 밀랍, 수소화 식물유, 글리세롤 모노스테아레이트 또는 글리세롤 팔미테이트, 고분자, 예를 들어 분자량 1500-6000인 PEGs와 같은 현탁제도 사용할 수 있다.
캡슐화되는 내용물이 현택액내에 입자를 함유할 때, 이 입자는 개별적으로 코팅될 수 있으며, 전형적으로는 위에서 언급된 물질로 적당하게 감미 또는 풍미화된 코팅으로 코팅될 수 있다. 그러한 코팅은 현탁액에 맛 차단제 및/또는 안정제를 제공할 수 있다.

Claims (16)
- 가소제, 상기 가소제를 위한 메트릭스를 형성하는 젤라틴 및 상기 젤라틴과 함께 사용할 수 있으며 상기 가소제를 위한 2차 메트릭스를 형성하는 녹말 아세테이트로 이루어지는 캡슐 껍질용 조성물.
- 제1항에 있어서, 상기 조성물이 젤라틴 18-30중량%와 가소제 30-45%를 함유하는 조성물.
- 제1항에 있어서, 상기 가소제가 글리세롤, 자일리톨, 솔비톨, 폴리글리세롤, 솔비톨의 비결정 용액, 글루코스, 프락토스, 글루코스 시럽 및 이들의 조합물로 구성되는 군에서 선택되는 조성물.
- 제3항에 있어서, 상기 가소제가 글리세롤인 조성물.
- 제1항에 있어서, 상기 녹말 아세테이트가 표백되지 않은 녹말 아세테이트인 조성물.
- 제5항에 있어서, 상기 표백되지 않은 녹말 아세테이트가 표백되지 않은 감자녹말 아세테이트인 조성물.
- 제1항에 있어서, 상기 녹말 아세테이트를 최고 12중량%으로 함유하는 조성물.
- 제7항에 있어서, 상기 녹말 아세테이트를 최고 3-10중량%으로 함유하는 조성물.
- 제1항에 있어서, 상기 오일을 최고 10중량% 함유하는 조성물.
- 제9항에 있어서, 상기 오일이 분별된 코코낫유인 조성물.
- 제1항에 있어서, 상기 표백된 녹말 아세테이트를 최고 12중량%으로 함유하는 조성물.
- 제11항에 있어서, 상기 표백된 녹말 아세테이트가 녹말에서 유래된 조성물.
- 제1항에 따른 씹을 수 있는 조성물.
- 제1항 내지 4항 및 5항 내지 13항중 어느 한 항에 따른 조성물로 이루어지는 껍질을 가지는 식용 캡슐.
- 녹말 아세테이트를 물 및 가소제와 혼합하고, 이 혼합물에 젤라틴을 첨가하고, 이 혼합물을 가루로 만들고, 이 혼합물을 가열하고, 가열된 혼합물을 정치하고, 수분손실을 최소화하면서 이 혼합물을 탈기하는 것으로 이루어지는 제1항 내지 4항 및 5항 내지 제13항 중 어느 한 항에 따른 조성물을 제조하는 방법.
