Abstract
본 발명은 디리트로마이신의 신규한 다형제를 함유하는 개선된 정제를 개시한다.
Description
본 발명은 약학 제형, 보다 구체적으로 매크롤라이드 항생 물질의 시규한 다형체를 함유하는 정제에 관한 것이다.디리트로마이신은 에리트로마이신으로부터 유도된 매크롤라이드 항생 물질이다. 항생물질은 미합중국 특허 제4,048,306호에 개시되어 있으며, 화학명, 에리트로마이신, 9-데옥소-11-데옥시-9,11-[이미노[2-메톡시에톡시)에틸리덴]옥시-[9S, 16R]를 갖는다. 미합중국 특허 제4,048,306호에 개시된 방법으로 제조된 디리트로마이신(이후로 형태 I로 지칭)은 다형체의 형태로 생성된다. 상기 디리트로마이신은 하기 X-선 분말 회절 패턴터(여기서, D는 결정면간 거리를 나타내고, I/Io는 상대 강도를 나타낸다)을 갖는다.
본 발명에 이르러, 본 출원인은 형태 I이 준안정하여 정제와 같은 약학 제형에 사용하기에 별로 적합하지 않음을 밝혀 내었다. 그러나, 놀랍게도, 본 발명에 따라, 디리트로마이신의 제2다형체 (이후로는 형태 Ⅱ로서 지칭)가 안정하며 따라서 정제와 같은 약학 제형에 사용하기 매우 적합함을 이제 밝혀내게 되었다. 그리하여, 본 발명의 한 태양으로, 활성 성분으로서 디리트로마이신 형태 Ⅱ를 함유하는 정제가 제공된다. 이 정제는 또한 하나 이상의 약학적으로 허용되는 부형제, 예를 들면 결합제, 윤활제, 붕해제, 충전제 등을 함유할 것임은 물론이다. 본 발명의 신규한 다형체는 필수적으로 하기 X-선 분말 회절 패턴(여기서, D는 결정면간 거리를 나타내고 I/Io는 상대 강도를 나타낸다)을 갖는다.
상기 X-선 분말 회절 패턴은 1=1.5418/Å의 구리 조사를 갖는 흑연 단색광기가 장착된 니콜렛(Nicolet) 12V 분말 회절계를 사용하여 얻었다. 당해 분야에 숙련된 자는 강도 값이 샘플 제법 및 기기 편차와 같은 요인에 영향을 받음을 인지할 것이다. 그러므로, D-거리는 회절 패턴의 기본이다. 정제(형태 Ⅱ)의 활성 성분은 약제로서 사용될 수 있으므로, 약학적 상태의 순도를 가져야 한다. 안정성의 목적을 위해, 형태 Ⅱ는 많은 양의 형태 Ⅰ을 함유하지 않아야 한다, 바람직하게, 형태 Ⅱ는 10%미만, 가장 바람직하게는 5%미만의 형태 Ⅰ과 혼합되어야 한다. 디리트로마이신은 매우 효과적인 항생물질이며, 총 500mg이하의 투여량으로 하루에 1회만 투여될 뿐이므로 다른 매크롤라이드 항생물질에 비해 상당한 잇점을 갖는다. 그러나, 상기 화합물은 위에 존재하는 것과 같은 산성 매질에 민감하므로 경구루여 가능한 제형의 고안에 있어서 다소 주의할 필요가 있다. 이점에 있어서, 미합중국 특허 제4,755,385호를 참조할 수 있으며, 본 발명의 정제는 위액에 내성을 갖는 층, 즉 장용성 제피(enteric coating)로 코팅되는 것이 바람직하다. 상기 제피의 성질은 당해분야에 숙련된 자에게 공지되어 있다. 흔히, 래커를 사용하여 제피를 생성한다.
하기 실시예 1내지 4는 형태 Ⅱ의 제법을 예시한다.
[실시예 1]
[아세토네이트로부터 디리트로마이신 형태 Ⅱ의 단리]
20 g 분량의 비-용매화된 디리트로마이신을 81 ml의 아세톤과 9ml의 물을 포함하는 용매 혼합물에 가하였다. 용액을 약 60℃로가열하였다. 용액을 반응 부피가 35ml가 될 때까지 60℃ 유지시켰다, 100ml 분량의 물을 1시간에 걸쳐 서서히 추가로 가하였다. 혼합물을 실온으로 냉각시켰다. 냉각시킨 혼합물을 빙욕에 넣고 1시간 동안 교반시켰다. 고형물을 여과에 의해 단리하고, 67% 물과 33%아세톤으로 이루어진 냉각 용매 25ml로 헹구었다. 고형물을 주위 온도에서 40℃물로 헹구었다. 디리트로마이신의 아세톤 용매화물의 습윤 케이크를 주변 조건에서 밤새 방치하였다. 180 ml 분량의 물을 아세톤 용매화물의 습윤 케이크에 가하였다. 혼합물을 70℃ 가열하고, 질소 퍼지하에 4시간 동안 70℃에서 교반시켰다. 고형물을 여과에 의해 즉시 단리하고, 70℃ 가온시킨 30 ml 분량의 물로 헹구었다. 단리된 고형물을 40℃ 진공하에 밤새 건조시켰다. 고형물은 X-선 분말 회절법에 의해 형태 Ⅱ 디리트로마이신으로 확인되었다.
총 수율 :90.8%.
[실시예 2]
[형태 Ⅱ 디리트로마이신의 단리]
디리트로마이신의 아세톤 용매화물 샘플 10.0g을 교반하면서 질소퍼지하에 100 ml의 afn에 가하였다. 반응 혼합물의 온도를 74℃로 증가시키고 약 72 내지 75℃에서 4시간 동안 교반시켰다. 따뜻한 혼합믈을 진공 여과시키고 60*의 물 약 35 ml로 세척하였다. 고형물을 50℃에서 진공하에 밤새 건조시켰다. 고형물은 X-선분말 회절법에 의해 형태 Ⅱ 디리트로마이신으로 확인되었다. 반응의 총 수율은 8.74g(87.4%)이었다.
[실시예 3]
[형태 Ⅱ 디리트로마이신의 단리]
15 g 분량의 형태 Ⅰ디리트로마이신을 150 ml의 물에 가하였다. 반응 혼합물을 74℃로 가열하고 74℃에서 4시간 동안 교반시켰다. 고형물을 여과에 의해 단리하고 각 70℃물 40ml로 2회 세척하였다. 샘플을 25℃의 진공 오븐 내에서 약 68시간 동안 건조시켰다. 생성물은 X-선 분말 회절법에 의해 형태 Ⅱ 디리트로마이신으로 확인되었다.
수율 : 97.5 %.
[실시예 4]
[형태 Ⅱ 디리트로마이신의 단리]
3.01g 분량의 형태 Ⅰ 디리트로마이신을 15 ml의 에틸 아세테이트에 가하였다. 반응 혼합물을 약 76℃로 가열하고, 혼합물을 반응 부피가 약 10 ml가 될 때까지 비등시켰다. 20 ml 분량의 n-옥탄을 가하였다. 혼합물을 실온으로 냉각시켰다. 고형물을 여과에 의해 단리하였다. 샘플을 실온에서 건조시켰다. 생성물은 X-선 분말 회절법에 의해 형태 Ⅱ 디리트로마이신으로 확인되었다. 총 수율 : 95%.
디리트로마이신을 함유하는 약학 제형을 제조하는 일반적인 방법은 미합중국 특허 제4,755,385호에 개시되어 있다. 사실상, 상기 제조 방법은 형태 Ⅱ 디리트로마이신을 필수적인 약학적으로 허용되는 담체와 혼합한 후 정제로 압착시키고, 임의로 위액에 내성을 갖는 층으로 코팅시킴을 포함한다. 그러나, 하기 특정 실시예는 본발명에 따른 코팅된 정제의 제법을 예시한다.
[실시예 5]

Claims (2)
