Description
− 皮膚:アレルギー、アトピー性湿疹、乾癬、
− 肺:嚢胞性線維症、喘息、COPD、
− 血管:冠状動脈性心疾患、動脈硬化症、アテローム性動脈硬化症、
− 内分泌系:糖尿病、脂肪症。
本発明の教示を有利に発展させ、さらに発展させる種々の可能性がある。このため、代表して本発明を説明する以下の実施例について記載する。
グループ1:0.25mLのビヒクル(滅菌水)の毎日強制経口投与。
グループ2:0.25mLのニコチンアミド(Cat.No.4488;Caelo)の滅菌水溶液(6mg/mL、最終用量:約60mg/kg体重)の毎日強制経口投与。
グループ3:食餌中に均質に分散した制御放出NAMミニ錠剤(最終用量:マウス1匹につき1日2.5gの飼料摂取を基準にして、約60mg/kg体重)。
グループ4:0.5%メチルセルロース(Cat.No.M7140、Sigma−Aldrich)中に濃度15mg/mLで懸濁した0.25mLの5−アミノサリチル酸(5−ASA;Cat.No.A3537、Sigma−Aldrich、Brondby、Denmark)の毎日強制経口投与(最終用量:約150mg/kg体重)。
投与溶液は、毎日投与直前に同じストック溶液から新たに調製した。
完全な結腸粘膜の盲検化した組織学的評価を、特許請求したNAM製剤の治療効果のハードエンドポイントとして選んだ。
グループ1(水対照):6.25±1.39(n=8)、
グループ2(NAM水溶液):5.14±1.07(n=7)、
グループ3(NAM制御放出錠剤):3.38±0.92(n=8)、
グループ4(5−ASA):6.50±1.60(n=8)。
グループ1:0.1mLのビヒクル(滅菌水)の毎日強制経口投与。
グループ2:0.1mLのNAM(Sigma−Aldrich、Brondby、Denmark)滅菌水溶液(18mg/mL、最終用量:約60mg/kg体重)の毎日強制経口投与。
グループ3:食餌中対照顆粒剤(顆粒剤含量はグループ6に相当)。
グループ4:食餌中NAM顆粒剤(最終用量:30mg/kg体重)。
グループ5:食餌中NAM顆粒剤(最終用量:60mg/kg体重)。
グループ6:食餌中NAM顆粒剤(最終用量:120mg/kg体重)。
グループ7:食餌中5−ASA顆粒剤(最終用量:150mg/kg体重)。
Claims (9)
- ニコチン酸;ニコチンアミド;ニコチン酸エステル;またはこれらの組合せから選択される活性物質を含み、
腸内微生物叢内の病原細菌の数の低減、および/または、病原細菌対有益な細菌の比の低減に用いるための;ここで、トリプトファン、ニコチン酸、ニコチンアミドまたはニコチン酸エステルの不足により、病原細菌の数は増加している、および/または、病原細菌対有益な細菌の比は増加している;
回腸終末部および/または結腸において前記活性物質を遅延放出または制御放出する、経口投与用に製剤化され、
改変しようとする前記腸内微生物叢が位置する回腸終末部、結腸またはその両方において局部的に有効であるように前記活性物質を選択的に放出する、
組成物。 - ニコチン酸、ニコチンアミド、またはこれらの組合せから選択される活性物質を含み、
腸内微生物叢内の病原細菌の数の低減、および/または、病原細菌対有益な細菌の比の低減に用いるための;ここで、トリプトファン、ニコチン酸、ニコチンアミドまたはニコチン酸エステルの不足により、病原細菌の数は増加している、および/または、病原細菌対有益な細菌の比は増加している;
回腸終末部および/または結腸において前記活性物質を遅延放出または制御放出する、経口投与用に製剤化され、
改変しようとする前記腸内微生物叢が位置する回腸終末部、結腸またはその両方において局部的に有効であるように前記活性物質を選択的に放出する、
組成物。 - 回腸終末部および/または結腸において前記活性物質を遅延放出する、経口投与用に製剤化された、請求項1または2に記載の組成物。
- 回腸終末部および/または結腸において前記活性物質を制御放出する、経口投与用に製剤化された、請求項1または2に記載の組成物。
- ニコチンアミドを含む、請求項1から4のいずれか一項に記載の組成物。
- 経口適用が、最終剤形あたり1〜3000mgの活性物質含量で実施されることを特徴とする、請求項1−5のいずれか一項に記載の組成物。
- 経口適用が、最終剤形あたり100〜1000mgの活性物質含量で実施されることを特徴とする、請求項6に記載の組成物。
- ニコチン酸および/またはニコチンアミドに加えて、アセチルサリチル酸および/またはプロスタグランジンD2拮抗薬を含有することを特徴とする、請求項1から7のいずれか一項に記載の組成物。
- 腸内微生物叢内の病原細菌の数の低減、および/または、病原細菌対有益な細菌の比の低減に用いるための;ここで、トリプトファン、ニコチン酸、ニコチンアミドまたはニコチン酸エステルの不足により、病原細菌の数は増加している、および/または、病原細菌対有益な細菌の比は増加している;
回腸終末部および/または結腸において前記活性物質を遅延放出または制御放出する、経口投与用に製剤化され、
改変しようとする前記腸内微生物叢が位置する回腸終末部、結腸またはその両方において局部的に有効であるように前記活性物質を選択的に放出する、
組成物の製造における、ニコチン酸、ニコチンアミド、ニコチン酸エステル、またはこれらの組合せの使用。
