インビトロにおけるHIV患者に対する抗CD40.LIPO5 HIVペプチドワクチン試験。αCD40.LIPO5 HIVペプチド融合組換え抗体(αCD40.LIPO5 rAb)が抗原提示を媒介する能力を研究するために、融合rAbは、HIVに感染した個人に由来する血液細胞に追加し、末梢血単核細胞(PBMC)からのサイトカイン産生を測定した。
図9は、DCによる、ターゲティングによる取り込み及びそれらの表面MHC複合体上へのペプチドエピトープの提示から結果として生じる抗原特異的T細胞の増大を指示するためのその能力についてαCD40.LIPO5 HIVペプチドワクチンを試験するためのプロトコールを示す図である。手短に言えば、HIV患者単球は、IFNα及びGM−CSFを用いる2日間の培養によってDCに分化する。次いで、異なる用量のαCD40.LIPO5 HIVペプチドワクチン又は5つのペプチドのミックスを18時間追加する。自己由来T細胞を3日目に共培養物(1:20の比で)に追加した。5日目に、100U/ml IL−2を培養物に追加し、次いで、培地は、2日ごとに100U/ml IL−2を用いてリフレッシュする。10日目に、増大した細胞に、αCD40.LIPO5 HIVペプチド融合rAb中に組み込まれる5つのHIVペプチド配列に対応する個々の長いペプチドを48時間再度曝露する。次いで、培養上清は、収集し、Luminexを使用してサイトカイン産生について評価する。

抗CD40 mAbの内部移行。1×106 IL−4DCは、非特異的結合をブロックするために3%BSAを含有するPBS中で3mg/mlヒトガンマグロブリンと共に氷中で1時間インキュベートした。細胞に、Alexa 568標識抗CD40 mAbを氷上で30分間適用した(すべて、非特異的ブロック中20ng/ml最終濃度で)。次いで、細胞は、洗浄し、37℃で、0〜90分間の異なる時間、表面結合抗体を内部移行させた。内部移行の後に、細胞は、1%BSA及び0.05%アジ化ナトリウムを含有する氷冷PBS(PBA)を用いて2度洗浄し、4℃で一晩、PBS中、氷冷1%メタノールフリーホルムアルデヒド(MFF)中で固定した。細胞は、4℃で20分間、0.5%サポニンを含有するPBS 3%BSA(PBAS)中で浸透性にし、96ウェル丸底ポリプロピレンマイクロタイタープレートに移入した。氷冷PBASを用いて2度洗浄した後に、細胞は、PBAS中3mg/mlヒトガンマグロブリンと共に氷上で1時間インキュベートした。BODIPY−ファロイジンをPBAS中で希釈し、氷中で1時間、細胞と共にインキュベートした。細胞は、さらに、核対比染色として、TOPRO−IIを用いて染色した。スライドは、Leica SP1共焦点顕微鏡上で画像処理した。
(配列番号29)をコードするイタリック体の残基は、免疫優性HLA−A2拘束性ペプチドであり、ペプチドを囲む下線を引いた残基は、MART−1に由来する)は、上記のベクターのNhe I及びXho I部位の間に挿入した。タンパク質は、大腸菌(E. coli)株BL21(DE3)(Novagen社製)又はT7 Express(NEB社製)中で発現させ、120mg/L IPTGを追加したら、カナマイシン抵抗性(40μg/ml)について選択し、200回/分で振盪しながら、対数増殖期中期まで37℃でLB中で成長させた。3時間後に、細胞は、遠心分離によって収集し、−80℃で保存した。それぞれの1Lの発酵に由来する大腸菌細胞は、0.1mlのプロテアーゼ阻害剤Cocktail II(Calbiochem社製、CA)と共に、30ml 氷冷50mM Tris、1mM EDTA pH8.0(緩衝液B)中で再懸濁した。細胞は、5分間の休み時間と共に、設定18(Fisher Sonic Dismembrator 60)で2×5分間、氷上で超音波処理し、次いで、4℃で20分間、17,000r.p.m.(Sorvall SA-600)で回転させた。His.Flu M1精製については、50ml細胞溶解物上清画分は、5ml Q Sepharoseビーズを通過させ、6.25ml 160mM Tris、40mMイミダゾール、4M NaCl pH7.9を、Q Sepharose通過画分に追加した。これは、Ni++を用いて荷電した5ml HiTrapキレート化HPカラム上に4ml/分でロードした。カラムに結合したタンパク質は、20mM NaPO4、300mM NaCl pH7.6(緩衝液D)を用いて洗浄し、100mM H3COONa pH4.0を用いてさらに洗浄した。結合したタンパク質は、100mM H3COONa pH4.0を用いて溶出した。ピーク画分は、プールし、100mM H3COONa pH5.5を用いて平衡化した5ml HiTrap Sカラム上に4ml/分でロードし、平衡緩衝液を用いて洗浄し、その後、50mM NaPO4 pH5.5中0〜1M NaClの勾配を用いて溶出した。約500mM NaClで溶出するピーク画分をプールした。Coh.Flu M1.His精製については、2Lの培養物に由来する細胞を上記のように溶解した。遠心分離後に、2.5ml Triton X114は、5分間氷上でインキュベーションしながら上清に追加した。5分間の25℃でのさらなるインキュベーションの後に、上清は、25℃での遠心分離後にTriton X114から分離した。抽出は、反復し、上清は、5mlのQ Sepharoseビーズを通過させ、6.25ml 160mM Tris、40mMイミダゾール、4M NaCl pH7.9を、Q Sepharose通過画分に追加した。次いで、タンパク質は、上記に記載されるようにNi++キレート化クロマトグラフィーによって精製し、緩衝液D中0〜500mMイミダゾールを用いて溶出した。

Claims (14)
- 配列番号41、42、及び43で示されるアミノ酸配列を含む少なくとも1つの抗体軽鎖可変領域と、配列番号44、45及び46で示されるアミノ酸配列を含む少なくとも1つの抗体重鎖可変領域とを含み、CD40に結合する組換え抗体又はその抗原結合断片であって、単独で、樹状細胞に、前記樹状細胞を事前に活性化することなく、IL−6、MIP−1a、IL−12p40及びTNFアルファのうちの少なくとも1つを分泌させることができる、組換え抗体又はその抗原結合断片。
- ガンマ−1、ガンマ−2、ガンマ−3若しくはガンマ−4ヒト重鎖定常領域又は前記ヒト重鎖定常領域のバリアントを含む重鎖定常領域と、ラムダ又はカッパヒト軽鎖定常領域を含む軽鎖定常領域とをさらに含む、請求項1に記載の抗体。
