まず、セロトニンを検出するために、1μg/mlのセロトニンが添加された血液に対して、図1aに示している装置を用いて、電気化学的検出方法を行った。セロトニンについて、0〜1.0V vs. Ag/AgClまで単位時間当りの一定の速度(10mV/min)で電極反応を調査したところ、図2のような一般形態の循環電圧電流曲線(Cyclic voltammogram)が導出された。特に、0.3V vs. Ag/AgClでピーク値が得られたことは、放出された電子が伝達されて、電気化学的に酸化反応が起きたことを意味する。よって、この時の電流値を測定することで、セロトニン濃度の定量が可能である。すなわち、セロトニンの濃度を定量的に測定するためには、濃度に対する電流値との相関関係を導出することで可能である。このような測定は、図1bの器具を用いて行うことができる。
に記載されたセロトニン測定用電気化学センサの作業電極(B)を、ナフィオンでコートした。具体的に、5重量%のナフィオン溶液5mlを電極表面に滴下した後、1時間の間、室温で乾燥させ、作業電極がナフィオンでコートされた作業電極(A)を備えた電気化学センサを製造した。

Claims (18)
- セロトニンの酸化反応が起きる作業電極と、電圧が印加される基準電極とを含み、セロトニンの酸化電位の印加時に流れる酸化電流を測定して、セロトニンの有無又は濃度を確認する電気化学センサにおいて、
前記作業電極は、(−)電荷を帯びるコート層を含み、コート層の静電気力で、(−)電荷を帯びる妨害物質の接近を抑制することを特徴とするセロトニン検出用電気化学センサ。 - 前記セロトニンの酸化反応は、0.3〜0.4Vで起きることを特徴とする請求項1に記載のセンサ。
- 前記コート層は、陽電荷イオン交換高分子を含有することを特徴とする請求項1に記載のセンサ。
- 前記陽電荷イオン交換高分子は、スルホン化テトラフルオロエチレン基盤フッ素−共重合体(sulfonated tetrafluoroethylene based fluoropolymer-copolymer)であることを特徴とする請求項3に記載のセンサ。
- 前記コート層は、セロトニンが作業電極に接触できるように気孔又はパターンが形成されたことを特徴とする請求項1に記載のセンサ。
- 前記妨害物質は、アスコルビン酸、尿素、及び尿酸を含む群より選ばれるいずれか1つ以上であることを特徴とする請求項1に記載のセンサ。
- 前記作業電極及び基準電極は、金(Au)又は炭素(C)からなることを特徴とする請求項1に記載のセンサ。
- 電流変化を測定する対照電極を含むことを特徴とする請求項1に記載のセンサ。
- サンプルを収容する反応容器を備え、
反応容器にサンプルが導入されると、作業電極と基準電極が通電して、回路が形成されることを特徴とする請求項1に記載のセンサ。 - 前記反応容器は、アレルギー誘発物質が収容又はコートされていることを特徴とする請求項9に記載のセンサ。
- 前記センサは、基準電極に所望する電位の電圧を印加する自動電位調節装置を含むことを特徴とする請求項1に記載のセンサ。
- 前記自動電位調節装置は、0〜1.0Vの電位範囲内で自動に調節することを特徴とする請求項11に記載のセンサ。
- 更に、セロトニンの酸化反応が起きる電位の印加時に測定された電流値を表示する表示部を備えることを特徴とする請求項1に記載のセンサ。
- セロトニンの酸化電位の印加時にセロトニンの酸化反応が起きる作業電極と、電圧が印加される基準電極とを含むセロトニン測定用ストリップにおいて、
前記作業電極は、静電気力で(−)電荷を帯びる妨害物質の接近を抑制するように、(−)電荷を帯びるコート層を含むことを特徴とするセロトニン検出用ストリップ。 - 前記ストリップは、分離交換され、一回又は複数の使用が可能であることを特徴とする請求項14に記載のセロトニン検出用ストリップ。
- 請求項14又は請求項15に記載のストリップを1つ以上備えることを特徴とするセロトニン検出用センサ。
- 請求項1乃至請求項13のいずれか一項に記載のセンサを備えることを特徴とするセロトニン測定キット。
- 分離された生体サンプルを、請求項1乃至請求項13のいずれか一項に記載のセンサで処理するステップと、
セロトニンの酸化電位の印加時に流れる酸化電流を測定するステップとを含むことを特徴とするアレルゲン確認方法。
