Abstract
Compounds of the general formula <FORM:0972966/C1/1> (wherein R represents an alkyl, alkenyl, hydroxyalkyl or aralkyl group, R1 represents an alkyl or aryl group, R2 represents an alkyl, alkoxy or trifluoromethyl group or a halogen atom, R3 represents a trifluoromethyl, alkyl, alkoxy, alkylthio, hydroxyalkylthio, nitro, amino, alkylsulphinyl, alkylsulphonyl, hydroxy or acylamino group or a halogen atom and R4, which can only be present if R3 is other than a trifluoromethyl group, represents any of the values of R3 except the latter value, and wherein the broken lines denote optional substituents) are prepared by reacting a benzophenone derivative of the general formula <FORM:0972966/C1/2> (wherein X represents a halogen atom) with ammonia, heating any uncyclized product obtained to effect ring closure and, in the case where R3 and R4 were absent in the benzophenone derivative, subjecting the benzodiazepine formed to one or more of the steps nitration, reduction of the nitro group and acylation of the amino group where it is desired to introduce a nitro, amino or acylamino group or, where either or both of the groups R3 and R4 in the benzophenone derivative were nitro or amino, converting them, if desired, into amino or acylamino by reduction and/or acylation. The reaction with ammonia proceeds through an a -aminoacylamido compound which can be isolated, if desired, when liquid ammonia is used. The benzophenone derivatives used as starting materials are prepared from 2-aminobenzophenone or a 2-(R-substituted amino) benzophenone or the appropriate nuclear-substituted derivative thereof by reaction with an a -haloacyl halide.
