Abstract
Benzodiazepine derivatives of the general formula <FORM:0972963/C1/1> (wherein R2 represents an alkyl, alkoxy or trifluoromethyl group or a halogen atom, R3 represents a trifluoromethyl, alkyl, alkoxy, alkylthio, hydroxyalkylthio, nitro, amino, alkylsulphinyl, alkylsulphonyl, hydroxy or acylamino group or a halogen atom and R4, which can only be present if R3 is other than trifluoromethyl, represents any of the values of R3 except the latter value, and wherein the broken lines denote that the alkyl, oxide and R groups or atoms attached thereby are optional) are prepared by hydrolysing a benzodiazepine derivative of the general formula <FORM:0972963/C1/2> with a strong acid under conditions of temperature which avoid the hydrolysis of any acylamino value of R3 and/or R4 or the opening of the 7-membered ring in the product or both as the case may be and, in the case where R3 and R4 were absent in the starting material, subjecting the product to one or more of the steps nitration, reduction of the nitro group and acylation of the amino group where a nitro, amino or acylamino group is required in the product or, in the case where R3 or R4 or both were nitro or amino in the starting material, subjecting the product if desired to reduction and/or acylation where one or two amino or acylamino groups are required in the product.
