Abstract
Compounds of the general formula <FORM:0972965/C1/1> (wherein R1 represents an alkyl, hydroxyalkyl or aralkyl group, R2 represents an alkyl, alkoxy or trifluoromethyl group or a halogen atom, R3 represents a trifluoromethyl, alkyl, alkoxy, alkylthio, hydroxyalkylthio, nitro (in cases where a 4, 5 double bond is present), amino, alkylsulphinyl (in cases where a 4, 5 double bond is present), alkylsulphonyl, hydroxy or acylamino group or a halogen atom and R4, which can only be present when R3 is other than a trifluoromethyl group, represents any of the values of R3 except the latter value and wherein the broken lines denote that the substituents attached thereto are optional and the double headed arrow signifies that, in the absence of the hydroxyl substituent on the 4-aza atom, the optional bond indicated thereby can exist as an additional bond in the 4, 5-position), and acid addition salts of those compounds which are basic, are prepared by either treating a compound of the general formula <FORM:0972965/C1/2> with a phosphorus trihalide (except in cases where a hydroxyalkyl or hydroxyalkylthio substituent is present) or with hydrogen in the presence of Raney nickel (where R3 and/or R4 is absent or is other than nitro or alkylsulphinyl or where R3 and/or R4 is nitro or alkylsulphinyl and an amino or alkylsulphonyl group is required in the product) and if desired where R3 and R4 are absent subjecting the product to one or more of the steps nitration, reduction of the nitro group and acylation of the amino group and, where R3 and R4 are either absent or have any of the values other than nitro or alkylsulphinyl accorded to them hereinbefore and a tetrahydro product is required, catalytically hydrogenating the double bond in the 4,5-position of the resulting 2,3-dihydro-1H-1, 4-benzodiazepinone-(2) with hydrogen in the presence of a platinum or other noble metal catalyst or, where a product is required in which R3 and R4 are either absent or have any of the values other than nitro or alkylsulphinyl accorded to them hereinbefore, catalytically hydrogenating the compound of said formula with hydrogen in the presence of a platinum catalyst so as to reduce the double bond in the 4,5-position and convert the oxide component into a hydroxy substituent and, in either case if desired, converting the product (if a base) into a salt by treatment with an acid.
