Claims (8)
1. Isoleret og/eller rekombinant β-glucosidase-polypeptidvariant med større aktivitet og/eller termostabilitet end vildtype-C1-protein (Bgl1), og som omfatter en aminosyresekvens, som er mindst ca. 70 % identisk med vildtype-C1-3-glucosidase (rest 20-870 af SEQ ID NO: 2), og som har mindst en substitution i position D369, hvor positionen bestemmes ved optimal alignering med SEQ ID NO: 2. 2. β-glucosidase-polypeptidvariant ifølge krav 1, hvor aminosyren i position 369 er valgt fra gruppen bestående af R, Q, L, Y, C, A, I, P, E, K, F, Μ, H og V, hvor varianten relativt til et nativt C1 -Bgl 1 omfattende aminosyrer 20-870 af SEQ ID NO: 2 eventuelt har: a) mindst 5-fold større termoaktivitet ved ca. pH 5 og ca. 65 °C, b) mindst 3-fold større termostabilitet ved ca. pH 5 og ca. 65 °C eller c) både (a) og (b). 3. β-glucosidase-polypeptidvariant ifølge krav 1 eller krav 2, hvor aminosyresekvensen omfatter Q291W, D369R/H/L/Y og E402N og mindst én substitution af en aminosyrerest valgt fra gruppen bestående af Q258N/H, I106V, E385L, D274Y, K495H/I/N/Q, Q119E, Y135Q, G628W, N437I/V/W/D, S501 R, Q313M, S434P og Y491 F. 4. β-glucosidase-polypeptidvariant ifølge krav 1, som har mindst 70% identitet, mindst 75 % identitet, mindst 80 % identitet, mindst 85 % identitet, mindst 90 % identitet eller mindst 95 % identitet med aminosyrerester 20-870 af SEQ ID NO: 2 og omfatter en aminosyresubstitution i position Q291, en aminosyresubstitution i position D369 og en aminosyresubstitution i position E402, som bestemt med henvisning til SEQ ID NO: 2. 5. β-glucosidase-polypeptidvariant ifølge krav 4, som yderligere omfatter mindst én yderligere aminosyresubstitution ved D47, A79, Q85, 1106, A109, Q258, V260, Q313, F314, A343, S434, A475, K495, G628, A689, Y715 eller A732, hvor substitution eventuelt er D471, A79E/G/M, Q85N/H, I106V, A109/S/T, V260G, Q313M, F314L/V, A343C, S434P, A475L, K495N, G628W, A689I, Y715P eller A732G, for eksempel hvor varianten omfatter mindst én substitution Q291W, D369R/L/H/Y eller E402N. 6. β-glucosidase-polypeptidvariant ifølge krav 4 eller krav 5, hvor varianten relativt til et nativt C1-Bgl1 omfattende aminosyrer 20-870 af SEQ ID NO: 2 har: a) mindst 5-fold større termoaktivitet ved ca. pH 5 og ca. 65 °C, b) mindst 3-fold større termostabilitet ved ca. pH 5 og ca. 65 °C eller c) både (a) og (b). 7. β-glucosidase-polypeptidvariant ifølge et hvilket som helst af de foregående krav, som har mindst 75 % identitet, mindst 80 % identitet, mindst 85 % identitet, mindst 90 % identitet, mindst 91 % identitet, mindst 92 % identitet, mindst 93 % identitet, mindst 94 % identitet, mindst 95 % identitet, mindst 96 % identitet, mindst 97 % identitet, mindst 98 % identitet eller mindst 99 % identitet med aminosyrerester 20-870 af SEQ ID NO: 2. 8. β-glucosidase-polypeptidvariant ifølge krav 1, der omfatter aminosyrerester 20- 870 af en sekvens udvalgt fra SEQ ID NO: 17, SEQ ID NO: 5, SEQ ID NO: 7; SEQ ID NO: 9, SEQ ID NO: 13, SEQ ID NO: 15, SEQ ID NO: 19 og SEQ ID NO: 21.
9. Rekombinant nukleinsyre, der koder for en β-glucosidase- polypeptidvariant ifølge et hvilket som helst af de foregående krav, hvor nukleinsyren eventuelt koder for et præprotein, der omfatter Bg 11-varianten fusioneret til et signalpeptid, eventuelt det naturligt forekommende C1-Bgl1-signalpeptid.
10. Ekspressionsvektor, der omfatter den rekombinante nukleinsyre ifølge krav 9.

Værtscelle, der omfatter den rekombinante nukleinsyre ifølge krav 10, hvor nukleinsyren eventuelt er operativt bundet til en anden promotor end CI-Bg/f-promotoren, for eksempel hvor: a) værtscellen er en art af filamentøs fungus, såsom en Aspergillus-, Trichoderma-, Humicola-, Chrysosporium-, Myceliophthora- eller Thielavia-art, eller b) værtscellen også eksprimerer mindst ét yderligere ikke-naturligt forekommende rekombinant cellulase-polypeptid, udvalgt fra: i) et rekombinant endoglucanase(EG)-polypeptid og ii) et rekombinant cellobiohydrolase-polypeptid (CBH).
12. Fremgangsmåde til fremstilling af et Bgl1 -polypeptid, omfattende dyrkning af en værtscelle ifølge krav 11 under betingelser, hvor Bgl1-varianten eksprimeres, for eksempel hvor varianten secerneres af værtscellen, eventuelt yderligere omfattende et trin til genvinding af Bgl1-varianten fra det medium, hvori cellen dyrkes.
13. Sammensætning, der omfatter en Bgl1-variant ifølge et hvilket som helst af kravene 1-8 og mindst ét yderligere ikke-naturligt forekommende rekombinant cellulase-polypeptid, der er udvalgt fra a) et rekombinant endoglucanase(EG)-polypeptid; b) et rekombinant cellobiohydrolase-polypeptid (CBH); og c) mindst ét rekombinant EG-polypeptid og mindst ét rekombinant CBH-polypeptid eventuelt: i) yderligere omfattende mindst ét polypeptid, der er udvalgt fra en gruppe af polypeptider bestående af en hemicellulase, en pectinase, en lignin-peroxidase, en manganperoxidase, en laccase og en cellobiose-dehydrogenase; ii) hvor cellobiohydrolasen (CBH) og/eller endoglucanasen (EG) er fra Trichoderma reesei, Myceliophthora thermophilia eller Streptomyces avermitilis eller en variant af en T. reesei CBH eller EG, M. thermophilia CBH eller EG eller en S. avermitilis CBH eller EG; eller iii) omfattende en værtscelle, der eksprimerer Bg 11-varianten.
14. Anvendelse af en Bg 11-variant ifølge et hvilket som helst af kravene 1-8 ved fremstilling af opløselige sukre fra cellulose-biomasse.
15. Anvendelse ifølge krav 14, der er en fremgangsmåde til fremstilling af glucose, omfattende kombinering af cellobiose med en β-glucosidase-variant ifølge et hvilket som helst af kravene 1-8 og eventuelt mindst én yderligere cellulase under betingelser, hvor der produceres glucose, for eksempel yderligere omfattende: a) dyrkning af en mikroorganisme under betingelser, hvor den producerede glucose anvendes som en energi- og carbonkilde til fremstilling af et metabolisk produkt, og b) genvinding af det metaboliske produkt, eventuelt hvor: i) mikroorganismen er en gær eller bakterier, eller ii) det metaboliske produkt er en alkohol, en organisk syre eller et carbonhydrid med 1-20 carbonatomer, hvor det metaboliske produkt for eksempel er ethanol.
