Abstract
Un dispositivo de membrana de afinidad utilizado para la remociónselectiva de moléculas objetivo contenidas en el plasma de sangre comprende: una caja alargada que tiene una puerta de entrada y una puerta de salida para la entrada y salida de la sangre de ella; fibras huecas encajadas en la cavidad interna, las fibras huecas tienen poros con tamaño de poro adecuados para separar la sangre de plasma y componentes celulares, los poros tienen ligandos inmovilizados a una superficie interna de los poros, los ligandos tienen una afinidad por las moléculas objetivo en el plasma, en donde los componentes celulares de la sangre no fluyen dentro de los poros de las fibras huecas y el plasma es transportado dentro de los poros por medio de filtración positiva e inversa en la ausencia de una bomba externa para generar un flujo de plasma a través de las fibras huecas. 1Un método para la remoción selectiva de moléculas objetivo presentes en el plasma comprende las etapas de: Suministrar un dispositivo de membrana de fibra hueca que tiene (a) una caja alargada con una puerta de entrada única y una puerta de salida única para entrada y salida de la sangre desde ella; y (b) fibras huecas encajadas dentro de la caja, las fibras huecas tienen poros con tamaños de poro adecuados para separar la sangre en plasma y componentes celulares, los poros tienen ligandos ligados a una superficie interna de los poros, los ligandos tienen una afinidad por las moléculas objetivo en el plasma; transportar la sangre dentro de la puerta de entrada de la caja; causar que el plasma de la sangre fluya dentro de los poros de las fibras huecas por medio de filtración positiva e inversa mientras que no le permite que los componentes celulares fluyan dentro de la misma; poner en contacto las moléculas objetivo en el plasma con los ligandos durante un período clínicamente significativo de tiempo para permitir que haya ligación entre las moléculas objetivo a los ligandos; y causar que las moléculas no objetivo del plasma se reúnan con los componentes celulares de la sangre.
