Abstract

发明背景
多年来,α-淀粉酶已经用于多种用途,其中最重要的有淀粉液化,织物脱浆,造纸和纸浆工业中的淀粉改性,以及用于酿造和烘烤。α-淀粉酶的另一用途正变得日益重要,即用于在碱性pH下与洗涤剂一起洗涤时去除淀粉类污渍。
市售α-淀粉酶产品例如有Termamyl,BAN和Fungamyl,这些都得自Novo Nordisk A/S,丹麦。这些产品和其它市售的类似产品的最佳pH在酸性到中性范围,一般在5-7.5之间,它们在碱性pH下的洗涤剂溶液中不能表现出最佳活性。
WO 95/26397披露了一种来自芽孢杆菌菌株的α-淀粉酶,其在pH8时具有最佳活性。WO 96/23873描述了在洗涤条件下性能改进的芽孢杆菌属淀粉酶变体。
US 5147796记述了具有α-淀粉酶活性的碱性支链淀粉酶。此文献的图2b显示其在pH 8-8.5时具有最佳淀粉酶活性。
M.Takagi等,J.Ferment.Bioeng.,vol 81,No.6,557-559(1996)描述了得自芽孢杆菌的碱性α-淀粉酶-支链淀粉酶。此酶的最佳α-淀粉酶活性在pH9,但是此活性随pH升高而快速降低,在pH10的活性低于pH7的活性。
本发明的一个目的是提供改进了在碱性溶液中,特别是在pH约为9-11的碱性洗涤剂溶液中性能的新α-淀粉酶。
发明概述
本发明提供α-淀粉酶,它是:
a)产自芽孢杆菌NCIMB 40916的多肽,或
b)具有SEQ ID NO:2的1-556位所示氨基酸序列的多肽,或
c)由克隆在大肠杆菌DSM 13001的质粒上的DNA序列中该α-淀粉酶编码部分所编码的多肽,或
d)在(a)或(b)中定义的多肽的类似物,其
i)与所述多肽至少60%同源,或
ii)是从所述多肽通过一或几个氨基酸的取代,缺失和/或插入而得的多肽,
另一方面,本发明提供了具有下列一或多个特征的α-淀粉酶:
●37℃检测时,在pH10.5下的活性是pH7.3下的至少2倍。
●37℃检测时,在pH9.5下的活性是pH7.3下的至少4倍。
●37℃检测时,最佳pH约为9.5。
●其具有的热稳定性使得:在3g/l的实验洗涤剂(含有20%STPP,25%Na2SO4,15%Na2CO3,20%LAS,5%C12-C15脂肪醇乙氧基化物,5%Na2Si2O5,0.3%NaCl,pH10.5,German硬度为6度)中25℃下温育20分钟,活性保持90%以上,30℃下在同一溶液中温育20分钟活性保持低于90%。
●SDS-PAGE测定其分子量约为55kDa。
●等电聚焦测定其等电点约为5。
本发明还提供了分离的编码α-淀粉酶的DNA序列,其中该α-淀粉酶如上所述,或其中的DNA序列包含:
a)SEQ ID NO:1的94-1764位所示的DNA序列,或
b)在a)中定义的DNA序列的类似物,其
i)与所述DNA序列至少60%同源,或
ii)与所述DNA序列在至少55℃下杂交。
本发明还提供了含有所述DNA序列的重组表达载体,和用所述DNA序列或重组表达载体转化的细胞。
本发明还涉及提供通过培养细胞生产α-淀粉酶的方法和含有该α-淀粉酶的洗涤剂组合物。

发明详述
微生物来源
本发明的α-淀粉酶可得自芽孢杆菌。优选菌株是芽孢杆菌菌株NCIMB40916。按照布达佩斯条约对用于专利程序目的的微生物保藏的国际认可,本发明人将此菌株于1998年1月28日保藏在国立工业和海洋微生物保藏有限公司(NCIMB),23 St.Machar Drive,Aberdeen AB 21 RY,苏格兰,英国。含有表示为SEQ ID NO:1克隆进质粒pJA 386的α-淀粉酶基因的大肠杆菌菌株NN049489,按照布达佩斯条约于1999年8月7日保藏于德意志微生物和细胞培养物保藏中心(DSMZ),Mascheroder Weg 1b,D-38124Braunschweig DE,保藏号DSM 13001。
α-淀粉酶的生产
可通过在合适的营养培养基中培养合适的生产淀粉酶的芽孢杆菌菌株或本发明的转化的宿主细胞,然后从该培养基中回收该α-淀粉酶,从而生产本发明α-淀粉酶。
用于培养细胞的培养基可以是任何适于目的宿主细胞生长并可以使本发明的α-淀粉酶表达的传统培养基。合适的培养基可得自市售产品或根据出版文献配制(例如美国典型培养物保藏中心目录所述)。
可由已知方法从培养基中方便地回收宿主细胞分泌的α-淀粉酶,包括通过离心或过滤从培养基分离细胞,然后用盐如硫酸铵沉淀培养基中的蛋白成分,再使用层析方法如离子交换层析,亲和层析等。α-淀粉酶的性质
优选得自芽孢杆菌NCIMB 40916的α-淀粉酶。它可按照实施例所述生产。此淀粉酶和编码它的DNA的全长氨基酸表示为SEQ ID NO:1和2。本发明的淀粉酶(NCIMB 40916的α-淀粉酶)具有下列特点:
采用Novex,4-25%梯度凝胶通过SDS-PAGE测定分子量约为55kDa。
通过等电聚焦(Ampholine PAG,pH 3.5-9.5)测定pI约为5。
pH-活性曲线表示在图1中,以pH 7.3的活性为100%。这是通过采用调整至各种pH值的50mM Britten-Robinson缓冲液经Phadebas检测测定的。作为参照,图1也表示了同样条件下两种现有芽孢杆菌淀粉酶(得自地衣芽孢杆菌的Termamyl和按照WO 95/26397生产的SP722)的pH图谱。图1表示在pH 9.5时本发明的淀粉酶比现有的两种淀粉酶活性高约10倍。图1也显示最佳活性在约pH 9.5处。
以调整pH到9.5的50mM Britten-Robinson缓冲液在各种温度下经Phadebas检测测定得到温度-活性曲线。结果显示在图2中。从图2可以看到本发明的淀粉酶在约55℃具有最佳活性。
经30分钟温育在pH 10.5(50Mm CAPS)下于22,37,45,55和65℃测定所述淀粉酶稳定性。所述酶用缓冲液稀释到40NU/ml,在各个温度下取25ml样品温育。30分钟后,将样品于冰上冷却,测定残余活性。图3表示本发明的淀粉酶和现有淀粉酶(SP 722)的稳定性图谱。
通过如上所述在3g/l的A/P型洗涤剂温育40NU/ml淀粉酶于pH 10.5和6°DH(German硬度,Ca∶Mg 2∶1)下测定稳定性。温育后,采用Phadebas法在pH 7.3下测定残余活性。25℃下温育后,残余活性为95%,30℃为87%。
多肽和DNA序列的同源性
氨基酸的同源性可测定为指示第一个序列衍生自第二个序列的两个序列间同一性的程度。同源性优选通过本领域已知的计算机程序测定。例如,可使用GCG version 8提供的FASTA(Needleman,S.B.和Wunsch,C.D.,(1970),Journal of Molecular Biology 48,443-453),采用以下设定:Scoringmatrix GenRunData:blosum50.cmp,所用Variable pamfactor的缺口产生罚分为12,缺口延伸罚分为2。
所述氨基酸序列与SEQ ID NO:2的1-556位表示的氨基酸序列表现出的同一性优选为至少60%,优选至少70%,更优选80%,特别是至少90%或至少95%,更优选至少97%或至少99%。
DNA序列的同源性可测定为指示第一个序列衍生自第二个序列的两个序列间同一性的程度。同源性适合通过本领域已知的计算机程序,例如GCG软件包中提供的GAP(如上所述)测定。Gap GCG version 8采用以下缺省参数设定:GAP的缺口产生罚分为5.0,GAP缺口延伸罚分为0.3,缺省使用Scoring matrix。GAP采用Needleman/Wunsch/Sellers的方法进行比对。
本发明的DNA构建体包含与SEQ ID NO:1的95-1764位核酸序列优选至少60%,优选至少70%,更优选80%,特别是至少90%或至少95%,更优选至少97%或至少99%同一性的DNA序列。
杂交
采用杂交指示特定DNA序列是对应于SEQ ID NO:1的核苷酸探针的类似物。杂交条件如下所述。
确定核酸探针与同源DNA或RNA序列间杂交的合适条件涉及在5×SSC(标准柠檬酸盐)中预浸泡携有待杂交的DNA片段或RNA的滤膜10分钟,在含5×SSC(Sambrook等,1989),5×Denhardt's溶液(Sambrook等,1989),0.5%SDS和100μg/ml超声变性鲑精DNA(Sambrook等,1989)的溶液中预杂交滤膜,然后在含有随机引发的(Feinberg,A.P.和Vogelstein,B.(1983))Anal.Biochem.132:6-13),32P-dCTP标记的(比活>1×109cpm/μg探针的相同溶液中约45℃下杂交12小时。滤膜在2×SSC,0.5%SDS中于温度至少55℃的条件下冲洗2遍,温度更优选至少60℃,更优选至少65℃,更优选至少70℃,特别至少75℃。
采用x-线胶片检测在这些条件下寡核苷酸探针与之杂交的分子。
重组表达载体
本发明的重组表达载体一般包括编码启动子,操纵子,核糖体结合位点,翻译起始信号,和任选地,阻抑基因或各种活化子基因的控制序列。
携带编码本发明α-淀粉酶的DNA序列的重组表达载体可以是任何可方便地进行重组DNA操作的载体,载体的选择经常取决于要引导载体进入其中的宿主细胞。这样,所述载体可以是自主复制载体,即作为染色体外实体存在的载体,其复制独立于染色体的复制,例如质粒,噬菌体或染色体外元件,微型染色体或人工染色体。可选地,所述载体可以是当引入宿主细胞时整合进宿主细胞染色体组并且与整合的染色体一起复制的那些。
载体中,DNA序列应与合适的启动子序列操作性连接。启动子可以是在所选宿主细胞中表现出转录活性的任何DNA序列,并且可能得自编码相对于宿主细胞同源或异源的蛋白的基因。指导编码本发明α-淀粉酶的DNA序列转录,特别是在宿主细胞中转录的适合启动子有大肠杆菌1ac操纵子的启动子,天蓝色链霉菌琼脂糖酶基因dagA启动子,地衣芽孢杆菌α-淀粉酶基因(amyL)的启动子,嗜热脂肪芽孢杆菌产麦芽糖淀粉酶基因(amyM)的启动子,解淀粉芽孢杆菌α-淀粉酶(amyQ)的启动子,枯草芽孢杆菌xy1A和xy1B基因的启动子等。可用于在真菌宿主中转录的启动子有编码米曲霉TAKA淀粉酶,米赫根毛霉(Rhizomucor miehei)天冬氨酸蛋白酶,黑曲霉中性α-淀粉酶,黑曲霉耐酸α-淀粉酶,黑曲霉葡糖淀粉酶,米赫根毛霉脂肪酶,米曲霉碱性蛋白酶,米曲霉三糖磷酸异构酶或构巢曲霉乙酰胺酶的基因的启动子。
本发明的表达载体也可包含合适的转录终止子,并且在真核细胞中,包含聚腺苷酸化序列,它们与编码本发明α-淀粉酶的DNA序列操作性相连。终止序列和聚腺苷酸化序列优选与启动子同源。
载体还可包括使载体在目的宿主细胞中复制的DNA序列。这样的序列例如有质粒pUC 19,pACYC 177,pUB 110,pE 194,pAMB1和pIJ 702的复制起点。
载体还可含有选择性标记,例如其产物能弥补宿主细胞缺陷的那些基因,例如枯草芽孢杆菌或地衣芽孢杆菌的da1基因,或赋予抗生素,如氨苄青霉素,卡那霉素,氯霉素或四环素抗性的那些基因。此外,该载体还含有曲霉选择性标记,如amdS,argB,niaD,sC,产生潮霉素抗性的标记,或可选择通过共转化来实现,例如WO 91/17243所述。
尽管胞内表达在某些方面较为有利,例如当使用特定细菌作宿主细胞时,但优选胞外表达。
本领域技术人员了解适合构建本发明的编码α-淀粉酶的载体(还分别含有启动子,终止子和其它元件)的方法(参见,例如Sambrook等,(1989))。
宿主细胞
本发明的含有如上定义的本发明DNA构建体或表达载体的细胞有利于用作重组生产本发明α-淀粉酶的宿主细胞。该细胞可用编码所述淀粉酶的本发明DNA构建体转化,这可方便地通过将所述DNA构建体(一或多个拷贝)整合入宿主染色体来完成。当所述DNA序列很可能稳定保持在所述细胞中时,通常认为此整合是有利的。可按照传统方法将所述DNA构建体整合入宿主染色体,例如通过同源或异源重组。备选地,所述细胞可根据不同的宿主细胞类型用如上所述的表达载体转化。
本发明的细胞可以是高等生物,如哺乳动物或昆虫的细胞,但优选是微生物细胞,如细菌或真菌(包括酵母)细胞。
在培养时能产生本发明的酶的细菌宿主细胞有革兰氏阳性细菌如芽孢杆菌属,如枯草芽孢杆菌,地衣芽孢杆菌,迟缓芽孢杆菌,B.clausi短芽孢杆菌,嗜热脂肪芽孢杆菌,嗜碱芽孢杆菌,解淀粉芽孢杆菌,凝结芽孢杆菌,环状芽孢杆菌,灿烂芽孢杆菌,巨大芽孢杆菌,或苏云金芽孢杆菌,或链霉菌如浅青紫链霉菌,鼠灰链霉菌,或革兰氏阴性细菌如大肠杆菌。细菌的转化可通过原生质体转化,电穿孔,偶联,或通过使用感受态细胞以其本身已知的方式来实现(参见Sambrook等,出处同上)。
酵母可优先选自糖酵母属或裂殖酵母属,例如酿酒酵母。丝状真菌可优先选自曲霉属,如米曲霉或黑曲霉。转化真菌细胞的方法涉及原生质体的形成及转化,然后使细胞壁以其本身已知的方式再生。EP 238023中记述了适于曲霉属宿主细胞转化的方法。
工业应用
由于本发明的α-淀粉酶在碱性pH值下的活性,很适用于多种工业工艺,特别是所述酶作为洗涤剂,例如洗衣和硬表面洗涤的洗涤剂组合物中一种成分的潜在应用,它也可用于生产甜味剂和由淀粉生产乙醇。这样,它可用于传统的淀粉转化过程,如美国专利3912590和欧洲专利公开号EP252730和63909所述的液化和糖化过程。
本发明的碱性α-淀粉酶也可用于从淀粉增强型废纸和纸板生产木质纤维材料,如纸浆,纸和纸板,特别是当在pH 7以上再浆化时以及当淀粉酶可促进降解增强型淀粉从而分解废料时。本发明的α-淀粉酶在从淀粉包覆的印刷纸张生产造纸用纸浆的工艺中特别有用。此工艺可按照WO95/14807所述进行,包括如下步骤:
a)分解纸产生纸浆,
b)在步骤a)之前,期间,之后用淀粉降解酶处理,
c)在步骤a)和b)之后从纸浆中除去墨点。
本发明的α-淀粉酶在淀粉改性,即淀粉经酶促改性后与碱性填充剂如碳酸钙,高岭土和粘土一起用于造纸时很有用。由于本发明碱性α-淀粉酶,使得在填充剂的存在下能使淀粉改性,这样可以简化整合过程。
本发明的α-淀粉酶在织物脱浆中也非常有用。在织物加工工业中,α-淀粉酶传统上在脱浆工艺中用作辅料,这样有助于去除用作纺织中纺织纱上的保护层的含淀粉的浆。纺织后浆衣的彻底去除对于在随后的工艺中确保最佳效果是重要的,所述后续步骤包括纤维的洗涤,漂白,和染色。优选用酶分解淀粉,因为这不会对纤维材料造成任何损害。为减少处理费用并提高工厂产量,脱浆工艺有时与洗涤和漂白步骤结合。在这种情况下,一般用非酶性辅料如碱或氧化剂分解淀粉,这是因为传统的α-淀粉酶不能与高pH值和漂白剂很好的相容。因为所用化学制剂具有相当的侵蚀性,非酶性分解淀粉浆确实导致一些纤维损害。因此,希望使用在碱性溶液中性能改进的本发明α-淀粉酶。所述α-淀粉酶可以单独使用或当对含纤维素的纤维或织物脱浆时与纤维素酶结合使用。
本发明的α-淀粉酶还在啤酒制造工艺中十分有用;该α-淀粉酶通常在化醪操作期间加入。
洗涤剂组合物
本发明的α-淀粉酶一般用作洗涤组合物例如洗衣洗涤剂组合物的一个成分。这样,它可以无粉尘颗粒,稳定液体,或受保护的酶的形式包含有洗涤剂组合物中。可按照US 4106991和4661452(都属于Novo Industri A/S)公开的方法制造无粉尘颗粒,可任选以本领域已知的任何方法包覆。蜡质包覆材料例如平均分子量1000到20000的聚环氧乙烷产物(聚乙二醇,PEG);具有16到50个氧丙环单位的乙氧基壬基酚;乙氧基脂肪醇,其中所述醇含有12到20个碳原子并且其中含有15到80个氧丙环单位;脂肪醇;脂肪酸;脂肪酸的单-和双-和三甘油酯。GB 1483591中给出了通过流化床技术适合用于形成胶片(film)的包覆材料。例如可以通过已知方法加入多元醇如丙二醇,糖或糖醇,乳酸或硼酸来稳定液态酶制剂。其它的酶稳定剂也为本领域所熟知。可按照EP 238216公开的方法制备受保护的酶。
本发明α-淀粉酶的性质使其特别适于在碱性,例如在pH 9.5-10.5中的洗涤剂中使用,和在低温如20-40℃下洗涤。
本发明的洗涤剂组合物可以采用任何便利的形式,如粉末,颗粒,浆或液体。液体洗涤剂可以是水性的,一般含有多达70%的水和0-30%的有机溶剂,或为非水性的。
所述洗涤剂组合物包含一或多种表面活性剂,它们分别可以是阴离子型的,非离子型的,阳离子型的或两性的。所述洗涤剂通常包含0-50%的阴离子表面活性剂如线性烷基苯磺酸(LAS),α-烯烃磺酸盐(AOS),烷基磺酸酯(脂肪醇磺酸酯)(AS),脂肪醇乙氧基硫酸酯(AEOS或AES),仲烷基磺酸酯(SAS),α-磺基脂肪酸甲酯,烷基或烷烯琥珀酸,或肥皂。它还可包含0-40%的非离子表面活性剂如脂肪醇乙氧基化物(AEO或AE),脂肪醇丙氧基化物,羧基化脂肪醇乙氧基化物,壬基酚乙氧基化物,烷基多聚葡糖苷,烷基二甲胺氧化物,乙氧基化脂肪酸单乙醇胺,脂肪酸单乙醇胺,或多羟基烷基脂肪酸胺(例如WO 92/06154所述)。
所述洗涤剂组合物还可含有一种或多种其它酶,如支链淀粉酶,酯酶,脂肪酶,角质酶(cutinase),蛋白酶,纤维素酶,过氧化物酶,或氧化酶,如漆酶。
通常所述洗涤剂含有1-65%的洗涤剂增洁剂(builder)或复合剂如沸石,二磷酸盐,三磷酸盐,膦酸酯,柠檬酸盐,次氮基三乙酸(NTA),乙二胺四乙酸(EDTA),二亚乙基三氨基五乙酸(DTMPA),烷基或链烯基琥珀酸,可溶性硅酸盐或分层硅酸盐(例如,Hoechst的SKS-6)。
可将洗涤剂增洁剂分为含磷型和不含磷型。含磷型无机碱性洗涤剂增洁剂例如包括水溶性盐,特别是碱金属焦磷酸盐,正磷酸盐,多磷酸盐和膦酸酯。不含磷型无机增洁剂例如包括水溶性碱金属碳酸盐,硼酸盐,硅酸盐以及分层的二硅酸盐,各种类型的水不溶性晶状或无定形硅酸铝,其中沸石已知最具代表性。
合适的有机增洁剂例如有琥珀酸,丙二酸,脂肪酸丙二酸,脂肪酸磺酸,羧甲氧基琥珀酸,聚乙酸,羧酸,聚羧酸,氨基聚羧酸,聚乙酰羧酸的碱金属盐,铵盐或取代的铵盐。洗涤剂也可以是非增强的,即基本上不含洗涤剂增洁剂。
洗涤剂可包含一或多种聚合物。例如有羧甲基纤维素(CMC),聚乙烯吡咯烷酮(PVP),聚乙二醇,聚乙烯醇(PVA),聚羧酸酯如聚乙酸酯,聚马来酸酯,马来酸/丙烯酸共聚物,甲基丙烯酸月桂酸/丙烯酸共聚物。
洗涤剂组合物可能包括氯/溴型或氧型漂白剂。漂白剂可以是包覆型的或胶囊型的。
无机氯/溴型漂白剂例如有次氯酸或次溴酸锂,钠或钙,以及氯化磷酸三钠。有机氯/溴型漂白剂例如有N-溴和N-氯酰亚胺的杂环化物例如三氯异氰尿酸,三溴异氰尿酸,二溴异氰尿酸,二氯异氰尿酸,及它们与水溶性阳离子如钾和钠形成的盐。乙内酰脲化合物也是适合的。漂白系统还可包含如酰胺,酰亚胺或磺酸型的过氧酸。
氧型漂白剂可以是无机过酸盐(优选带有漂白活性剂),或过氧酸化合物。无机过酸盐例如有过硼酸碱金属盐的四水合物和一水合物,过碳酸,过硅酸和过磷酸的碱金属盐。活化剂可以是四乙酰基乙二胺(TAED)或壬酰氧苯磺酸盐(NOBS)。
本发明洗涤剂组合物的酶可采用传统稳定剂,例如多元醇,如丙二醇或甘油,糖或糖醇,乳酸,硼酸或硼酸衍生物如芳香族硼酸酯来稳定,所述组合物可按照,例如WO 92/19709和WO 92/19708进行配制。也可以加入可逆的酶抑制剂,如EP 0544777B1所述的蛋白型,来稳定本发明的酶。
所述洗涤剂还可包含其它传统的洗涤剂成分,例如织物调理剂包括粘土,悬浮剂材料,泡沫促进剂,消泡剂(foam depressor),抑泡剂(sudssuppressor),抗腐蚀剂,污垢悬浮剂,抗污垢再沉积剂(anti-soil-redepositionagent),染料,脱水剂,杀菌剂,光亮剂,或香味剂。
pH值(在所用浓度的水溶液中测量)通常为中性或碱性,例如范围是7-11。
更具体地说,本发明的α-淀粉酶可以加入到WO 96/23873所述的任何一种洗涤剂配方中。
本发明α-淀粉酶可以按照洗涤剂中常规使用的浓度来加入。目前设想,在本发明的洗涤剂组合物中,所述α-淀粉酶的加入量为每升洗液中α-淀粉酶为0.00001-1mg(以纯酶蛋白计算)。
本发明还在下列实施例中予以说明,它们不应视作对本发明的限制。

实施例1
将α-淀粉酶基因克隆进大肠杆菌
采用Pitcher,D.G.,Saunders,N.A.,和Owen,R.J.(1989)Lett.Appl.Microbiol.8,151-156的方法从芽孢杆菌NCIMB 40916分离出DNA。染色体DNA用限制酶Sau3AI部分消化。将这些片段克隆进如图4所示的克隆载体pSJ1678的BamHI位点,并转化进入大肠杆菌SJ2,从而产生芽孢杆菌NCIMB 40916的基因文库。
对此基因文库的α-淀粉酶活性进行筛选,表现出α-淀粉酶活性的菌株命名为大肠杆菌NN049489菌株,它含有本发明的α-淀粉酶。对此菌株进行保藏,保藏号DSM 13001。载有编码α-淀粉酶的名为pJA386的质粒的大肠杆菌NN049489菌株用于DNA测序。
实施例2
DNA和α-淀粉酶测序
采用Taq deoxytermianl cycle测序试剂盒(Perkin Elmer,USA),荧光标记的终止子,以适当的寡核苷酸作引物通过引物步行法进行DNA测序,以此鉴定已克隆在质粒pJA386中的α-淀粉酶基因。
按照Devereux等(1984)Nucleic Acids Res.,12,387-395的方法进行序列数据分析。所述序列对应于表示为SEQ ID NO:1的DNA序列。α-淀粉酶的预计蛋白序列表示为SEQ ID NO:2,它包含信号肽和成熟α-淀粉酶。通过对该蛋白N末端34个氨基酸的测序证实推论的N末端序列。
实施例3
α-淀粉酶的生产
在含有10mg/ml氯霉素的LB培养基中,37℃,以250rpm过夜培养载有质粒pJA386的大肠杆菌NN049489(DSM 13001)。通过6000rpm离心15分钟,从2.71培养液收获细胞。通过下述渗透压休克法从细胞内释放出位于胞内的淀粉酶:
1)将细胞重悬,并用500ml 10mM Tris-HCl,pH7.0(EKV-缓冲液)冲洗,然后离心。
2)将细胞重悬于75ml 20%蔗糖,30mM Tris-HCl pH8,1mM EDTA中,加入溶菌酶至浓度为5mg/ml。
3)将此溶液冰上温育15分钟,然后离心。此时,大部分淀粉酶包含在上清液中。
上清对101 EKV缓冲液透析过液,期间更换一次缓冲液以降低葡萄糖的高浓度。此溶液经过0.45mm膜真空过滤器过滤。
将酶液上柱到预先用EKV缓冲液(pH7)平衡的Pharmacia Q Sepharosecolume FF,用EKV缓冲液冲洗柱。以0-500mM NaCl线性梯度,10倍柱体积洗脱结合的蛋白,汇集含有淀粉酶的级分,对EKV缓冲液透析过夜。
然后将所得溶液上柱到预先用EKV缓冲液(pH8)平衡的Q Sepharosecolume FF,用EKV缓冲液冲洗柱。以0-500mM NaCl线性梯度,10倍柱体积洗脱结合的蛋白,汇集含有淀粉酶的级分。
所得酶液中加入硫酸铵至浓度1M,上样到预先用含有1M硫酸铵的EKV缓冲液(pH8)平衡的Phenyl Superose柱。用硫酸铵缓冲液洗出未结合的物质,以1-0mM NaCl线性梯度的硫酸铵,用20倍柱体积洗脱。汇集含有淀粉酶的级分。
通过SDS-PAGE分析纯化的淀粉酶,考马司亮兰染色后只得到一条带。
实施例4
洗涤实验
将已弄脏的待检布样在含本发明α-淀粉酶的洗涤剂液中25℃下洗涤15分钟,以此进行洗涤效果的评价。
所用洗涤剂是上述的A/P型洗涤剂,用量3g/l,pH10.5,或马来西亚产的一种市售洗涤剂(Colgate的FAB Total),用量3g/l,pH约9.7。将实施例3的纯化淀粉酶以如下所示浓度加入到洗涤剂溶液中。实验样品污渍为orangerice starch得自(CFT(Center for Test Material,荷兰)的CS-28样品)。
洗涤后,测定布样在460nm的污渍减少程度(remission)以对其进行评价。结果表示为DR=用所述α-淀粉酶洗涤的布样的污渍减少程度减去同样条件下没有用该α-淀粉酶而洗涤的布样的污渍减少程度。

Claims (21)
1.一种α-淀粉酶,它是:
a)一种由芽孢杆菌NCIMB 40916菌株产生的多肽,或
b)一种具有SEQ ID NO:2的1-556位所示氨基酸序列的多肽,或
c)一种由克隆在大肠杆菌DSM 13001(NN049489)的质粒上的DNA序列中所述α-淀粉酶编码部分所编码的多肽,或
d)在(a)或(b)中定义的多肽的类似物,其
(i)与所述多肽至少有60%的同源性,或
(ii)是从所述多肽通过一或多个氨基酸的取代,缺失和/或插入而得的多肽。
2.一种α-淀粉酶,在37℃测定时,其在pH10.5下的活性比pH7.3下的高至少2倍。
3.一种α-淀粉酶,在37℃测定时,其在pH9.5下的活性比pH7.3下的高至少4倍。
4.一种α-淀粉酶,在37℃测定时,其最佳pH约为9.5。
5.前述任一项权利要求的α-淀粉酶,其得自芽孢杆菌,优选芽孢杆菌NCIMB 40916菌株。
6.前述任一项权利要求的α-淀粉酶,其中3g/I的实验洗涤剂溶液中25℃下温育20分钟后,其活性还保留了90%以上,所述实验洗涤剂含有20%的STPP,25%Na2SO4,15%Na2CO3,20%LAS,5%C12-C15脂肪醇乙氧基化物,5%Na2Si2O5,0.3%NaCl,pH10.5,German硬度为6,同一种溶液中30℃温育20分钟后其活性保持小于90%。
7.前述任一项权利要求的α-淀粉酶,SDS-PAGE测定其分子量约为55kDa。
8.前述任一项权利要求的α-淀粉酶,等电聚焦测定其等电点约为5。
9.前述任一项权利要求的α-淀粉酶,其在洗涤剂添加剂的形式中为非粉尘状颗粒或稳定液体。
10.一种分离的DNA序列,其编码前述任一项权利要求的α-淀粉酶。
11.一种分离的DNA序列,其编码一种α-淀粉酶并包含:
a)SEQ ID NO:1的94-1764位所示的DNA序列,或
b)在a)中定义的DNA序列的类似物,其
i)与该DNA序列有至少60%的同源性,或
ii)在至少55℃下与该DNA序列杂交。
12.权利要求10或11的DNA序列,其来自细菌,优选来自芽孢杆菌属,最优选来自NCIMB 40916菌株。
13.含有权利要求10-12中任一项的DNA序列的重组表达载体。
14.由权利要求10-12中任一项的DNA序列或权利要求13的重组表达载体转化的细胞。
15.权利要求14的细胞,其是原核细胞,具体是细菌细胞或最初产生该DNA序列的内源细胞。
16.权利要求15的细胞,其中所述细胞属于芽孢杆菌属或埃希氏菌属,优选大肠杆菌。
17.生产α-淀粉酶的方法,其包括在允许所述α-淀粉酶产生的条件下培养权利要求14-16中任一项的细胞,然后从培养液中回收该α-淀粉酶。
18.生产前述任一项权利要求的α-淀粉酶的方法,其包括在合适的营养培养基中培养能生产淀粉酶的芽孢杆菌属菌株,然后从培养液中回收该α-淀粉酶。
19.一种包含权利要求1-9中任一项的α-淀粉酶和一种表面活性剂的洗涤剂组合物。
20.前述权利要求的洗涤剂组合物,其水溶液的pH值为8.5-11,优选pH9-10.5。
21.权利要求19或20的洗涤剂组合物,其是衣物洗涤剂。
