Description
Technical Field
The invention belongs to the technical field of traditional Chinese medicines, and particularly relates to a traditional Chinese medicine formula for treating rheumatoid arthritis and a preparation method thereof.
Background
Rheumatoid arthritis is a chronic systemic autoimmune disease mainly caused by joint diseases, and the pathological features of the rheumatoid arthritis are chronic inflammation of joint synovium, cell infiltration, pannus formation and invasion of cartilage and bone tissues, which cause damage of joint structures, and the rheumatoid arthritis is still one of major diseases threatening human health and life at present. At present, the clinical western medicines mostly adopt nonspecific symptomatic treatment, including glucocorticoid and nonsteroidal anti-inflammatory drug, and drugs for improving the disease condition and resisting rheumatism, and although the symptomatic treatment drugs can improve the symptoms of arthritis of patients, the modern medicine has no specific treatment for the middle and late stages of rheumatoid arthritis. The history of the traditional Chinese medicine for treating rheumatoid arthritis is long, a unique theoretical system is gradually formed, and the traditional Chinese medicine has detailed description on the expression of the arthralgia syndrome and records the application of the prescription and the medicine in the aspect of dialectical treatment of the arthralgia syndrome. In recent years, with the wide and deep development of traditional Chinese medicine research on the treatment of rheumatoid arthritis, the traditional Chinese medicine and western medicine combined treatment or the combined administration of oral administration and external application of the traditional Chinese medicine are adopted clinically, so that the good curative effect is obtained, and meanwhile, the application of the traditional Chinese medicine has the advantages of stable curative effect, safety, small adverse reaction and the like, and becomes one of indispensable effective methods for treating the rheumatoid arthritis.

Rheumatic diseases are diseases which are different in pain but involve joints and surrounding soft tissues including muscles, ligaments, bursa of slipping and fascia, and are caused by joint pain, stiffness in the morning, swelling and tenderness, limited activity and a chronic process of alternating attack and relief, and some patients can suffer from whole body muscle weakness, pain, joint disability and viscera function weakness. The diseases are in the scope of arthralgia and flaccidity symptoms of traditional Chinese medicine, and are caused by liver and kidney deficiency, external pathogenic factors such as wind, cold and dampness attacking human body, blocking channels and collaterals and unsmooth circulation of qi and blood. The book of emperor's internal channel, questions and Bi Lun says that the combination of three qi of wind-cold-dampness is also arthralgia and that the qi must be deficient. Based on this theory, the recipe uses the actions of dispelling wind and removing dampness, nourishing blood and dispelling cold, and strengthening healthy qi to eliminate pathogenic factors, so called "self-extinguishing of blood-moving wind", "internal storage of healthy qi and no drying of pathogenic factors". The medicines used in the prescription have the effects of expelling wind and removing cold, and the traditional Chinese medicine theory considers that the arthralgia syndrome is 'wind-cold-dampness, three qi mixed and even arthralgia'. Wind is the first to treat all kinds of diseases, so the main treatment for arthralgia syndrome is wind. The angelica and the rhizoma gastrodiae in the formula enrich the blood and promote blood circulation, and remove blood stasis and relieve pain; radix Cyathulae has effects of removing blood stasis, dredging channels, dispelling pathogenic wind, removing dampness, and dredging joint; radix Paeoniae alba can nourish blood and soften liver, and relieve epigastric distention and pain; the angelica dahurica dispels wind and dampness, and reduces swelling and relieves pain; it is combined with Bai Fu Zi and quan Xie to strengthen the actions of removing toxicity, dissipating nodulation and alleviating pain; ru Xiang and Mo Yao are the essential herbs for dredging the zang-fu organs and circulating meridians and collaterals when used together. The whole formula has the effects of expelling wind and clearing away cold, and removing dampness and removing arthralgia.
The modern pharmacological research of the efficacy is summarized as follows:

Can be used for treating infantile convulsion, epilepsy, convulsion, tetanus, headache, vertigo, limb paralysis, numbness of limbs, and rheumatalgia. The medicine mainly contains phenols, glycosides, polysaccharide compounds and the like, and modern pharmacological research shows that the medicine has the effects of tranquilizing and resisting convulsion, relieving pain, resisting inflammation, improving immunity and protecting the cardiovascular system.
Ru Xiang and Mo Yao can activate blood and resolve stasis, move qi and alleviate pain. But frankincense is pungent and fragrant, and is better at relieving pain mainly by promoting qi and blood circulation; mo Yao is bitter in flavor and good at promoting blood circulation and dissipating blood stasis, so it is good at relieving swelling. Compendium of materia Medica: "dispel blood and relieve swelling, relieve pain and promote tissue regeneration". "Ru Xiang is used for activating blood and Mo Yao for dispersing blood, so it can alleviate pain, resolve swelling and promote granulation, so they are used together. "
Radix angelicae: has effects in expelling pathogenic wind, relieving exterior syndrome, expelling cold, relieving pain, eliminating dampness, dredging orifice, and relieving swelling and pus. Compared with the prior art, the invention has the following advantages:
1. in the prior art, the original formula is a capsule formulation, but the preparation process is simple and crude, and scientific extraction and preparation processes are lacked, the content of the capsule is that the medicinal materials are directly filled into the capsule after being crushed, the crude medicines are directly taken without extraction, the content of the effective components of the medicine is low, and the dosage is large. The invention improves the existing medicine preparation method, classifies the properties of various active ingredients contained in the medicinal materials of the prescription, selects different extraction methods to respectively extract, and changes the solubility of a solvent by changing the concentration of ethanol to remove impurities such as tannin, resin, fat-soluble pigment, protein and the like, thereby achieving the purpose of coarse extraction and fine extraction and improving the content of the active ingredients of the medicine contained in unit weight. The effective content of the medicine taken every day of the medicine prepared by the process is 15-18 times of that of the medicine processed by the prior art, so that the curative effect of clinical medicine application is improved by 10-13%.
2. Because the angelica, the angelica dahurica, the frankincense, the myrrh and the like in the formula all contain volatile substances, the main effective components of the medicinal materials are volatile oil and fat-soluble substances, the volatile oil is easy to volatilize when the temperature is higher than 60 ℃, the water decoction temperature reaches 100 ℃, the volatile components are greatly dissipated, and the lipid components can not be completely extracted in water, so that the original effect of the formula is lost, and the two substances are easy to dissolve in a low-polarity organic solvent and have low solubility in water, therefore, the extraction process firstly adopts a steam distillation method to extract the volatile components, and then mother liquor is reserved, so that the volatile oil and the fat-soluble substances are effectively obtained, and the water-soluble substances which are not completely extracted during extraction are reserved.
3. The invention adopts β -cyclodextrin inclusion technology to carry out inclusion on the volatile oil, the medicine is included in the molecular capsule, the release can be controlled, the irritation and the side effect are reduced, the volatilization is reduced, the liquid medicine is powdered, and β -cyclodextrin is carbohydrate which can be absorbed and utilized by human body without accumulation.
