Description
技术领域
本发明涉及基因工程领域,特别是一种家蚕乙酰胆碱酯酶基因核苷酸序列,以该核苷酸序列编码的多肽及其制备方法和应用。
背景技术
乙酰胆碱酯酶(acetylcholinesterase,AChE;Ace;EC3.1.1.7)是神经传导中的一种关键酶,在胆碱能突触间,该酶降解乙酰胆碱,终止神经递质对突触后膜的兴奋作用,保证神经信号在生物体内的正常传递。它是有机磷和氨基甲酸酯类杀虫剂的作用靶标。杀虫剂与酶分子结合后,使酶磷酰化或氨基甲酰化,钝化酶的活性而阻断正常的神经传导,使昆虫致死。
1986年Hall和Spierer从黑腹果蝇中克隆到第一个昆虫的乙酰胆碱酯酶基因,迄今为止已经报道了23种昆虫和螨类的乙酰胆碱酯酶的基因序列。家蚕是重要的经济昆虫,是人类最早驯化的动物之一。也是继果蝇和冈比亚按蚊之后第三个完成基因组测序的昆虫。在长期的驯化过程中与农药的接触相对较少,关于家蚕乙酰胆碱酯酶基因的克隆和序列分析在国内外还未见报道。
杆状病毒表达系统(Baculovirus expression vector system,BEVS)是20世纪80年代发展起来的真核表达系统,Smith等人在“Production ofhuman belta interferon in insect cells infected with baculovirus expressionvector”(《Mol Cell Biol》.19833,183-192.)公开的技术,利用AcMNPV,在草地贪夜蛾(Spodoptera frugiperda)Sf-9细胞中成功地表达了β-干扰素。又有人利用BmNPV在家蚕幼虫体内高水平表达了人的α-干扰素,目前日臻完善,已有数百种外源基因在该系统中进行了表达。Bac to Bac杆状病毒表达系统是利用杆状病毒穿梭载体Bacmid,它含有完整的AcMNPV基因组,既能在大肠杆菌中低拷贝复制和稳定分离,又可以直接转染昆虫细胞,得到病毒粒子。外源基因通过位点特异性转座作用整合到Bacmid中,经过筛选重组Bacmid。直接转染昆虫细胞,得到纯的重组病毒,同时高效表达外源基因,该系统具有快速稳定高效等特点。
发明内容
本发明提供了家蚕两种类型乙酰胆碱酯酶的核苷酸序列及编码的多肽序列及其表达和应用。
一种编码家蚕乙酰胆碱酯酶基因的序列,具有序列表中SEQ ID No.1所述的核苷酸序列。
一种编码家蚕乙酰胆碱酯酶基因的序列,具有序列表中SEQ ID No.2所述的核苷酸序列。
本发明首次克隆到编码家蚕乙酰胆碱酯酶的两个基因,分别为Bmace1和Bmace2。(具有序列表中SEQ ID No.1、SEQ ID No.2所述的核苷酸序列)。Bmace1基因编码区全长为2052bp,预计编码683个氨基酸残基,Signal P预测到一个17氨基酸残基的信号肽。Bmace2编码区长度为1917bp,编码一个638个残基的蛋白,预测有一个23个氨基酸残基的信号肽。同时将两个基因与电鳐,果蝇等其它昆虫序列联配的结果表明,克隆的两个基因具有保守的催化三联体,6个半胱氨酸残基形成三对二硫键,保守的氧阴离子洞,保守的FGESAG基序。
本发明还提供了所述基因(Bmace1)编码的多肽,该多肽具有序列表中SEQ ID No.3所述的氨基酸序列。
本发明还提供了所述基因(Bmace2)编码的多肽,该多肽具有序列表中SEQ ID No.4所述的氨基酸序列。
本发明还提供了基因(Bmace1)编码的多肽的制备方法,包括如下步骤:
(a)构建带有目的基因的杆状病毒转移载体;
(b)将杆状病毒转移载体转化含有穿梭载体Bacmid的大肠杆菌DH10Bac,提取重组基因;
(c)将重组基因转染粉纹夜蛾细胞,培养后收获细胞,分离出多肽。
本发明还提供了基因(Bmace2)编码的多肽的制备方法,包括如下步骤:
(a)构建带有目的基因的杆状病毒转移载体;
(b)将杆状病毒转移载体转化含有穿梭载体Bacmid的大肠杆菌DH10Bac,提取重组基因;
(c)将重组基因转染粉纹夜蛾细胞,培养后收获细胞,分离出多肽。
本发明还提供了基因(Bmace1)和基因(Bmace2)编码的多肽在农药筛选中的应用。
本发明中的“分离的”、“纯化的”或“基本纯的”DNA是指,该DNA或片段已从天然状态下位于其两侧的序列中分离出来;还指该DNA或片段已经与天然状态下伴随核酸的组份分开,而且已经与在细胞中伴随其的蛋白质分开。
在本发明中,术语“家蚕乙酰胆碱酯酶蛋白(或多肽)编码序列”指编码具有家蚕乙酰胆碱酯酶蛋白(或多肽)的核苷酸序列。
该术语还包括能编码具有与家蚕乙酰胆碱酯酶相同功能的蛋白的序列的变异形式。这些变异形式包括(但并不限于):若干个核苷酸的缺失、插入和或取代,以及在5’和/或3’端添加数个核苷酸。
在本发明中,术语“家蚕乙酰胆碱酯酶蛋白多肽”指具有家蚕乙酰胆碱酯酶蛋白活性的多肽。该术语还包括具有与家蚕乙酰蛋白胆碱酯酶蛋白相同功能的序列的变异形式。这些变异形式包括(但并不限于):若干个氨基酸的缺失、插入和/或取代,以及在C末端和/或N末端添加一个或数个氨基酸。例如,在本领域中,用性能相近或相似的氨基酸进行取代时,通常不会改变蛋白质的功能。又比如,在C末端和/或N末端添加一个或数个氨基酸通常也不会改变蛋白质的功能。该术语还包括家蚕乙酰胆碱酯酶蛋白的活性片段和活性衍生物。
该多肽的变异形式包括:同源序列、等位变异体、天然突变体,诱导突变体,在高或低的严谨度条件下能与家蚕乙酰胆碱酯酶DNA杂交的DNA所编码的蛋白,以及利用抗家蚕乙酰胆碱酯酶多肽的抗血清获得的多肽或蛋白。
本发明中,可选用本领域已知的各种载体,如市售的载体。
在本发明中,术语“宿主细胞”为真核细胞,包括酵母细胞,昆虫细胞和哺乳动物细胞,优选宿主细胞是真核细胞,如Tn细胞、Sf细胞等。
本发明首次克隆得到家蚕的乙酰胆碱酯酶基因,为家蚕乙酰胆碱酯酶的双基因编码提供了直接的证据,且公开家蚕乙酰胆碱酯酶的基因组序列,是鳞翅目昆虫中第一个明确基因结构的昆虫。家蚕是一种驯养了5000多年的模式昆虫,因此家蚕乙酰胆碱酯酶基因具有特别的用途。

家蚕乙酰胆碱酯酶的cDNA的克隆和测定
以家蚕总RNA反转录得cDNA为模版,用两对寡核苷酸为引物,其中正向引物含有Bgl II酶切位点,反向引物含有Xho I酶切位点。

扩增得到2型家蚕乙酰胆碱酯酶基因,编码区长度为1917bp,称为Bmace2基因(SEQ ID No.2),编码一个638个残基的蛋白,预测有一个23个氨基酸残基的信号肽。
通过RT-PCR,分别用两对引物扩增出两条片段。将PCR扩增产物与pGEM-T Easy载体(Promega)连接,转化大肠杆菌TG1用碱法提取质粒,进行测序。
家蚕两种类型乙酰胆碱酯酶基因在真核细胞(Tn细胞株,即粉纹夜蛾细胞株)中的表达
将带有家蚕1型(Bmace1)和2型(Bmace2)乙酰胆碱酯酶基因的质粒pGEM-T-easy载体分别用Bgl II+Xho I进行双酶切,分别回收大小为2052bp和1917bp的片段,并分别与用BamH I+Xho I双酶切的载体pFASTBac1进行连接反应,连接产物转化TG1,经碱法抽取质粒DNA,并通过PCR验证,PCR片段与理论大小相符,说明杆状病毒转移载体构建成功。转移载体分别命名为pFastBac1-Bmace1和pFastBac1-Bmace2。
用重组转移载体pFastBac1-Bmace1和pFastBac1-Bmace2分别转化含有穿梭载体Bacmid的大肠杆菌DH10Bac,挑取发生转座作用生成的白色菌落,提取重组Bacmid DNA,并作PCR分析鉴定,得到了整合位点正确的分别含有Bmace1和Bmace2基因的重组杆状病毒Bacmid载体。
在无血清条件下用Lipofectin介导重组Bacmid DNA转染生长状态良好的粉纹夜蛾细胞,8h后换新鲜培养基(含10%胎牛血清),72h后取10μL含病毒粒子的培养液继续传代感染。连续4~5次传代感染后,收获细胞,3000r/min离心3min,弃上清,PBS缓冲液洗涤1~2次。1×PBS悬浮细胞,加等体积2×SDS上样缓冲液,100℃煮沸5min。同时与被感染Bamid的细胞同时收获进行电泳SDS-PAGE分析。结果表明,感染含有Bmace1的重组病毒的Tn细胞抽提物在71kD左右出现特异性表达条带。进一步对凝胶进行自动扫描分析表明,Bmace1表达量占总蛋白的15.2%(图1);感染含有Bmace2重组病毒的Tn细胞抽提物在66kD处有特异性的表达带。凝胶进行自动扫描分析表明,Bmace2表达量占总蛋白的13%(图2)。
表达产物的生物活性分析和在农药测定中的利用
以0.5mM的碘化硫代乙酰胆碱乙酰胆碱(acetylthiocholine iodide,ATCh)为底物测定了上述表达的Bmace1和Bmace2蛋白的酶的活性。Bmace1酶的活性为28.63nmol/min/mg.pro,Bmace2酶的活性为30.65nmol/min/mg.pro。
同时测定了两种有机磷农药抑制剂即对氧磷和毒扁豆碱对上述表达的Bmace1和Bmace2两种类型的家蚕乙酰胆碱酯酶的抑制作用。与从活体幼虫头部抽提的粗酶液相比,表达产物对两种抑制剂更为敏感。对氧磷农药对粗酶液、Bmace1和Bmace2的抑制中浓度I50分别为9.8×10-7M、2.5×10-7M和0.6×10-7M,Bmace2比Bmace1更为敏感。毒扁豆碱的抑制也表现出同样的趋势,粗酶液、Bmace1和Bmace2的I50分别为4.8×10-7M、0.7×10-7M和0.2×10-7M。同时抑制结果也发现,毒扁豆碱对三种酶的抑制作用高于对氧磷。
结果说明本发明用真核细胞表达的Bmace1和Bmace2基因工程酶对有机磷农药敏感,可以用于筛选有机磷农药。本发明首先表达出具有生物活性的基因工程鳞翅目乙酰胆碱酯酶,为针对农业最重要类群的害虫即鳞翅目害虫的杀虫剂设计和新农药离体筛选提供了便捷途径,在新农药筛选前期可省去了培养鳞翅目昆虫的大量费用和时间。

Claims (7)
1、一种编码家蚕乙酰胆碱酯酶基因的序列,具有序列表中SEQ IDNo.1所述的核苷酸序列。
2、一种编码家蚕乙酰胆碱酯酶基因的序列,具有序列表中SEQ IDNo.2所述的核苷酸序列。
3、如权利要求1所述基因编码的多肽,该多肽具有序列表中SEQ IDNo.3所述的氨基酸序列。
4、如权利要求2所述基因编码的多肽,该多肽具有序列表中SEQ IDNo.4所述的氨基酸序列。
5、如权利要求3所述的多肽的制备方法,其特征在于包括如下步骤:
(a)构建带有目的基因的杆状病毒转移载体;
(b)将杆状病毒转移载体转化含有穿梭载体Bacmid的大肠杆菌DH10Bac,提取重组基因;
(c)将重组基因转染粉纹夜蛾细胞,培养后收获细胞,分离出多肽。
6、如权利要求4所述的多肽的制备方法,其特征在于包括如下步骤:
(a)构建带有目的基因的杆状病毒转移载体;
(b)将杆状病毒转移载体转化含有穿梭载体Bacmid的大肠杆菌DH10Bac,提取重组基因;
(c)将重组基因转染粉纹夜蛾细胞,培养后收获细胞,分离出多肽。
7、如权利要求2或3所述的多肽在农药筛选中的应用。
