Abstract
Matricne metaloproteinaze (MMP) su grupa enzima koja ucestvuje u patoloskoj razgradnji vezivnog tikiva i osnovnih membrana. Ove endopeptidaze, koje sadrze cink, sastoje se od nekoliko podvrsta enzima, u koje dolaze kolagenaze, stromelizini i zelatinaze. Enzimi za konverziju TNF-α (TACE), proinflamatorni citokin, katalizuju stvaranje TNF-α iz prokursorskog proteina TNF-α vezanog u membrani. Ocekuje se da ce mali molekuli, kao inhibitori MMP i TACE, posedovati potencijal za lecenje niza bolesnih stanja. Sadasnji pronalazak daje ne-peptidne inhibitore matricnih metaloproteinaza (MMP) i enzima za konverziju TNF-α (TACE), niske molarne mase, za lecenje artritisa, metastaza tumora, ulceracije tkiva, abnormalnog zarastanja rana, periodontalnih oboljenja, oboljenja kostiju, dijabetesa (rezistentnost prema insulinu) i HIV infekcije. Jedinjenja ovog pronalaska perdstavlja formula (I) gde su R1, R2, R3 i R4 opisani u tekstu.
