Abstract

3-(2)-AMINO-ALI TIOL-MODIFIKOVANI, S FLUORESCENTNOM BOJOM VEZANI NUKLEOZIDI, NUKLEOTIDI I OLIGONUKLEOTIDI, POSTUPAK ZA NJIHOVO DOBIJANJE I NJIHOVA UPOTREBA, u 3'- i/ili 2'-položaju nalazeću OH-grupu nukleozida, nukleotida ili oligonukleotida pomoću derivata prevedemo u amino ili tiolnu grupu a potom sjedinimo fluorescentni pigment. Nastale 3'- i/ili 2'-amino i tiolno modificirane nukleozide, nukleotide i oligonukleotide možemo potom da upotrebimo za sintezu prema snopovima u prisustvu šablonsog snopa ili oligonukleotida kao i za detekciju genetskog materijala. Jedino ovi nude prednost, da fluorescentnu oznaku više ne postavimo na 5'-kraju oligonukleotida ili na nukleobazu i time je ne treba uvoditi u hemijsku sintezu kao u dosada poznatim markiranim tehnikama, medjutim pošto poznate i uobičajene metode imaju nedostatak, da možemo da upotrebimo samo neke polimeraze za sintezu, da prihvatljivost trifosfata opadne zbog polimeraza a i osim toga je potreban veći višak supstrata.
