Description
The invention relates to the field of pharmacology, more specifically to microbiological methods for the preparation of drugs, namely cytochrome C, used to treat heart failure and other diseases associated with the difficulty of cellular respiration. The known method of obtaining cytochrome C by cultivating the yeast genus Candida under conditions of aeration in a nutrient medium containing sources of carbon, nitrogen and mineral salts does not provide a satisfactory yield of nutritional C and is very economical due to the need to use environment components - vitamin B | glycine and a-ketoglutaric acid. To reduce the cost of the process and increase the yield of the drug, it is proposed to cultivate the yeast species Candida mycoderma in the presence of yeast autolysate and iron salts and under conditions of ionized aeration (sulfite value 0.6-0.8 g OL / h). The concentration of iron salts in the medium during cultivation is maintained at the level of 1-5 mg1l. Example. Culture of Candida mycoderma VKM-U-934 support pas wort agar. The medium for obtaining seed in the main process of growing biomass, the yeast / kei has the following composition: 0.3% ammonium sulfate, 0.07% magnesium sulphate, 0.05% sodium chloride, 0.04% calcium nitrate, 0 , 01% potassium phosphate dibasic, 0.1% potassium phosphate monosubstituted, 2% glucose, 5% brewer's yeast autolysate, and 0.003% iron ammonium alum. Prepare the medium on boiled tap water. The yeast is grown in flasks on iri-28-ZO C shakers under conditions of moderate aeration (0.75 zQiAliac. The sowing material at the age of 1 day is transferred to 2% of the volume of the medium in the flasks. The pH value during the main process of growing the yeast is maintained at a level of 4.0-4.5 by periodically injecting sterile 10% sodium hydroxide solution. It lasts for two days and cultivates two days. The biomass yield is 6.8 g dry cells per liter of medium. The cytochrome C content is 3150 mg per kg dry cells. Subject of the invention 1. Method of preparation Totochrome C by cultivating Candida's yeast under aeration conditions on a nutrient medium containing sources of carbon, nitrogen and minerals, characterized in that, in order to reduce the process and increase the yield of the product, the cultivation of the yeast Candida mycoderma species is conducted in the presence of yeast autolysate and salts of iron and in ulo-5 4, types of reduced aeration (sulfite number 0.6-0.8 g). 2. Particular n. 1, characterized in that the cultivation is carried out and iron ends in medium 1-5 mg / l.
