Abstract
Esta invencion proporciona un proceso para la preparacion de novedosas sales de hemifumarato polimorfico de la 8-cloro -6, 11-dihidro- 11-(4 -piperidilideno) -5H- benzo [5, 61- ciclohepta [l, 2-b] piridina, denominada posteriormente en la presente como "desloratadina". Estas formas de sales polimorfica muestran una solubilidad mucho mas alta en agua, y tambien en solventes organicos proticos, comparandose con la desloratadina progenitora. Comprendiendo el proceso para preparar las formas polimorficas: a) mezclar la solucion etanolica de desloratadina y acido fumarico a una temperatura de aproximadamente 55°C a 70°C, agitando durante 30 a 45 minutos despues de mezclar, y posteriormente, filtrar el solido preparado en una condicion caliente, para dar la forma polimorfica 2 que tiene una DSC de 232°C ° 2°C; o b) mezclar la solucion etanolica de desloratadina y acido fumarico a una temperatura de aproximadamente 15°C hasta la temperatura ambiente (25°C), agitar a esta temperatura durante 30 a 45 minutos, y luego filtrar a temperatura ambiente, para dar la forma polimorfica 1 que tiene una DSC de 224°C ° 2°C.
